  • Penicillin pass through porins of gram negative bacterial cell wall. The penicillin then binds to penicillin binding protein linked the cell membrane to be activated. Active penicillin binds to and inactivate transpeptidase enzyme. As a result peptidoglycans NAM and NAG sugars are not cross-linked and the cell wall collapse

    Tetrapeptide is attached to NAM
    These Tetrapeptides are linked together by pentaglycine cross-link with the aid of transpeptiase ( penicillin binding protein ) and Penicillin inhibits this enzyme .

      Yes, this is correct though in one book I'm reading, it says pentapeptide instead of tetrapeptide

    Correction - PBP's are transpeptidases that link different segments of the peptidoglycans together.

    5 amino acids chain come from nam( N acetyl muramic acid). Not from nag( N acetyl glucosamine.

      These protein are not meant for pencillins but it is property of pencillins to bind them and that's why bacteria alter them and acquire resistance

      One inaccuracy in the video is the representation of PBPs. Penicillin-binding proteins *are* the transpeptidases. They aren't separate molecules.

    Why does penicillin not work on candida albicans? Is it due to it being made up of Chitin, not peptidoglycan? or is there more to it?

      Candida is fungus and cell wall is different, some what more complex ergosterol, glucan and chitin are targets
