  • This T cell development lecture explains about the T cell maturation and differentiation pathway. It explains the mechanism of how T cell is trained in thymus after the activation and the chemical activation and differentiation of T cells in Thymus. This video also explains the thymic selection and clonal propagation of T cells.

    In addition to cell cycle proteins, cytokines and their receptors are produced following antigen activation of T cells. These are involved in the expansion and further differentiation of the T cells into memory and effector cells. IL-2 is an autocrine factor which leads to T cell proliferation. Other surface molecules induced following activation include CD40L (CD154) which interacts with CD40 on dendritic cells inducing them to produce cytokines (e.g. IL-12) required for T cell proliferation and differentiation into Th1 cells. This leads to the specific T cell priming and clonal expansion necessary for their effector function and the development of memory.

    Just to confirm, when the thymocyte comes in through the HEV it will be CD4-CD8-, the t-cell receptor will begin to form via VDJ recombination, and then they will interact with the thymic epithelial cells to become CD4 or CD8 in the inner cortex and this is where positive selection happens (with MHC I or MHC II) becoming CD8 and CD4 respectively --> then in the medulla they'll undergo negative selection with dendritic cells or medullary thymic cells (these cells have peripheral antigens which induce the negative expression) --> then they go on to the lymph nodes or in the blood as naive T-cells awaiting to differentiate in T-helper or cytotoxic T-cells

      Bro, can u explain the concept of MHC restriction? Like why some cells cannot bind to the mhc, even though they have the same tcr having alpha and beta chains !!

    i have a doubt.... in the video he says that the thymocytes undergo apoptosis if they dont recognize the protein present on the the self cell, so how odes the thymus recognize if the thymocyte has previously bound to it or not and subject it towards apoptosis
