- MHC class 1 and 2 - This immunology lecture explains about the structure and functions of MHC class 1 and 2. It explains how MHC class 1 and 2 are processed by the antigen presenting cell and the mechanism by which these major histocompatibility complex receptors engage in interaction with the helper T cells that stimulates the immune response in body. This lecture also explains the difference between MHC 1 and 2 with respect to their functions.

I agree with many of the other comments -- the content here is made exceptionally clear. Example, no one ever explains the damn name: Major Histocompatibility Complex -- in terms of graft/ host. Your body is using MHGs to check whether the tissue (histo) is compatible with you/SELF. This is but ONE function.
He explains this, and also how only the macrophages, dendritic and B cells possess the MHC2 receptor -- its all made so clear in a way that keeps looping back to the overall picture of the immune system. Congratulate Shomu, and yourself for finding him! He seems to really love the subject and this is contagious.

Loading of antigen onto MHC class 2 does not occur in the ER and does not require the proteasome, however loading onto MHC class 1 does require a proteasome and does occur in the ER but it is not for pathogens that have been engulfed from the extra cellular environment, that mechanism is for pathogens that replicate within a cell and thus there is no need for an uptake stage during antigen processing. Please correct that mistake.
Clip replacement occurs in the cytosol and it occurs with the help of HLA-DM. It does not occur freely and randomly as explained in the video.

One thing i was curious to ask while watching the video was, that means these MHC complexes are not present on the cell surface all the time , and only if the pathogen is recognised , the ER will release the MHC complex bounded with pathogen. Is it so ?

Sir, please help me on this question! If I mix 2 mismatched MHC donors’ T cells together, are T cells going to kill each other? Or it has to have dendritic cells to involve? I mean if DC is necessary for T cell to recognize that it is not my cell? Thank you very much!

What happens to the antigen fragments that are transported into to endoplasmic reticulum??...
Do ER has abundant MHC complexes that carries the antigen fragment and present it on surface??
On surface of cell .. only one MHC complex is represented or it can be more?

Does MHC Restriction explain Organ/Graft rejection by T cells? ( How can T cells attack organ cells as that organ has antigens on different set of MHC molecules which acc to MHC Restriction T Cells cannot kill )

Great video. But I have a confusion. , My notes suggest that the Alpha chains of the MHC 2 are encoded by the HLA DP DQ DR. If that is true who is responsible for encoding the Beta 1 & 2 chains?
