I've been experimenting with mushroom growth, and this method is like, the only way I've gotten any mycelium growth that actually looks good. This feels like it should be the first step to getting a lot of mycelium every time.
Nice video! Making a liquid culture from spores is not ideal. Keep in mind a spore print is not just sterile spores, they are all contaminated to some degree. Thats why most people will put a few drops of a spore syring on agar x 3 or more plates, then find isolated mycelium and move it to another plate, grow that up and check for contamination, isolate and repeat until you get a clean plate. Then use some of that mycelium to add to a sterile honey water jar, which is then used to inoculate as many jars as you need. Then at that point you can make a liquid culture syringe that contains uncontaminated mycelium and store it in the fridge for years.
@@SporenSprout Thank you! As it happens I do have a question for you. I prepared two identical kilner jars with modified lids and one tablespoon of the same honey in each jar with 600ml of distilled water in each jar. I have just got them both out of the canner and one jar is clear and golden with a little sediment in it which is fine but the other jar is slightly opaque and ever so slightly cloudy but I see no sediment?! How can this be with two identical jars set up the same way? Should I bin the cloudy looking one or just go ahead and keep both? Thanks for getting back to me so soon, appreciate it.
@@cyberbid That is a common problem that was very hard to diagnose but I figured it out after a lot of trouble. Make sure you are using a 23 quart presto cooker with 3 quarts of distilled water in the bottom. Raise the jars off the bottom with a short rack and make sure when all jars are set inside that the water level is coming up to touch the bottom 2” of the jars. So the problem comes from either not using enough water in the pressure cooker to where it’s bathing the jars or the pressure cooker itself is not good for this application. For example I had to stop using my 41 quart all American electric sterilizer because the same problem occurred every time. I only use a presto 23 quart now and the problem never happens. It’s important that the jars come out dark golden. You will be surprised by the difference when you apply what I suggest.
@@ShouldaWaved For 600ml I use half a gram of light malt extract, 1 gram of peptone and roughly 10ml of karo. Any corn syrup can be used, karo is just what I happen to have here. Honey is fine as well. A tsp is enough 1.5 tsp is absolutely plenty, as high as 2 tsp is as much as I have used and not made it clouded. The light malt extract is the main thing that makes it clouded if you use a whole gram. That is just my finding. It stays clear and grows incredibly fast.
Appreciate the video! Had sediment and didn't know what caused it. I PC'd @ 15psi for 30mins,but left it in there to cool down and it must've kept cooking it. Thanks again!
I have also noticed if the water is too hot when mixing the honey into the jar it can cause the sediment as well. I make sure the water is warm not boiling hot when stirring in the honey. Good luck!
I’m just getting started in liquid culture. My question is after you inculcate the jars how do you store the jars and at what temperature? And how often should I stir ?
You can just store them on a shelf in a clean room that is around 75-78 degrees F. Stir every 2-3 days. Once full of mycelium you should move to a room that is 40-60 degrees F to slow down the mycelium metabolism.

You can use a canning pot to steam bath the jars for 2 hours the same as you would can a jar of food. For grain spawn, corn can be steam bathed the same way but rye grain would need a pressure cooker.

Yes you are right, refrigeration and stirring once per week is good. Yes they should be good over 12 months, and then new master liquid culture can be created from master slants at that point.

Always make sure you have enough water in the bottom of the pressure cooker and you will not have problems with the bottom warping. That seems to be the most common problem. I love my all American sterilizer!
What are your thoughts when the culture mix looks cloudy vs semi transparent? Some little bits of white culture floating and at the bottom there is darker masses?
I don’t use cloudy jars. It is normal for mycelium to grow and float on the surface. Try to stir frequently to avoid it. The mycelium mass at the bottom is not supposed to be dark colored it’s supposed to be a light colored mass.
I've had this idea for a while. Instead of adding water to the grain when sterilizing why not only add a little then pump the bag with a ton of liquid culture after sterilization? While that'll run more risk of contamination of the lc I'd bet it's actually safer for the grow bag to have such a headstart. People treat lc like it's hard to multiply but once you have one you have as many jars as you want.
I do something similar for growing Cordyceps. I use a whole syringe to inoculate one small jar and that’s normal. It’s to allow faster colonization and works just like you are describing. Sometimes my rye grain jars start to dry out and I always think about squirting in more sterilized water haha keep up the good thoughts and I would try it!

They are good at 70-75 degrees F for a couple months and then they should be moved to a cold room (60 degrees F) or refrigerator. At room temperature they can last 6 months sometimes longer, and in the refrigerator they last over a year. 2-4 ml per inoculation, spores are the reproductive solution for mushrooms. The spores come from the cap, they germinate into cottony mycelium and replicate continuously when in contact with nutrients.
It would be unwise to inoculate it with dried mushroom. The only possible way is to put a few drops of clean water on the dried mushroom cap and it will rehydrate just enough to possibly drop spores on tinfoil as a spore print. Scrape the spores up and mix with sterilized water and inoculate agar plates and transfer until a clean plate is achieved. You can then add a wedge of the colonized agar to the liquid culture media.
I have a couple is your LC syringes , and some from another company . Can’t wait to make LC with them . This video was really great Once I add the LC to the jars , do I stir once mycelium grows? Or stir before pulling into a syringe ?

It is ready to use whenever there’s enough mycelium for whatever your needs are. It can be stored for 12 months or longer at room temp or preferably in a cold room. Stir every other day at the beginning and then you can stir once a week.
I have 6 jars of liquid culture, various strains of mushrooms, that are all around a year and a half old. All of them are still healthy, no apparent problems. I will be taking a sample and transferring them to new LQ soon to be safe though.
@@SporenSprout how long does it last this way? i suppose when im starting running low, i just make another batch from the old one right? or is it like grain to grain trasfer and the culture will start to loose vigor if i do this few times
I cut a wedge and open the lid to drop it in but you can try disturbing the mycelium on the agar with a scalpel and then squirt some sterile water on the plates, tilt it around and then suck the liquid back up and use that hoping for success but may take longer the grow at first.

rather late reply.... but you can also take a syringe, draw a little liquid from the culture jar, cut a small wedge from the plate and put into into something like a shot glass. Now mix the wedge and liquid in shotglass. Very shortly you should be able to draw the mixture back into a syringe and then back into intended jar.

2-4 ml is what I use for most species in any size. I like to use more to speed up the process, especially for slower species like lions mane or shiitake.
Do you keep the magnetic stirrer in the jar until it’s full of culture? & when it’s full of culture do you keep stirring or it can be stored in refrigerator for up to year/room temp for 6 months and that storing time, no stirring? & magnetic stirrer, when is it removed?

can you tell me what the tin foil cover is for? also i have a jar that seems to have a mycelium cloud only at the bottom and it doesn't seem to expand any higher, is that normal or has it stopped growing?
The foil prevents moisture from wetting the filter on the lid during sterilization. It is fairly normal for the mycelium to grow only on the bottom of the jar and not further depending on the quality of the LC Media and genetics. If you don’t have a magnetic stir bar to break up the cloud then it will grow very slow.

Yes or even a wedge from an agar plate. Each method has its particular usefulness. Agar is good for longer term storage and isolating better genetics. Liquid culture and grain to grain transfer is the best way to scale the mycelium in bulk.
What do we do if we don’t have a pressure cooker. Also can you make a tutorial on how to grow lentinus crinitus with everyday materials because I unfortunately don’t have the money to buy a kit.
You can use a giant canner pot with a rack in the bottom with water and steam the jars for 2.5 hours. Yes I have grown tiger sawgill mushroom. I got sediment after cooking it for 30 minutes at 15 psi. Do I take pressure weight off immediately to cool? Is it still ok to use liquid culture if there is sediment from over cookong?
@@Bonecarpenter. It’s because you didn’t use enough water in the pressure cooker. Make sure the water level is touching bottom 2” of the jars while cooking.
So I made two batches of LC, maybe 3 days apart. The first one, I definitely didn’t use enough light corn syrup(distracted), so the 2nd jar is developing faster than the first one. The first one seems to have stopped growing mycelium. Can I sterilize a corn syrup mixture and add it to the first one or just leave it alone? Tia
I like your idea and think it is worth trying as an experiment if you can successfully sterilize a concentrated syrup/water mix without caramelizing/overcooking the sugars and inject it into the jar. I’d make sure the jar doesn’t have sediment or cloudiness before attempting. I usually dump out jars that stall because most of the time it is not worth waiting for, when a fresh batch that is made perfectly will yield way faster and better results. Good luck!
@@keemzero4151 Big pot steamed for 2 hours without pressure. Pressure cooked for 20-30 minutes using pressure and higher temperature. The pressure makes the honey sugars caramelize too much and it becomes cloudy. I prefer big pot.
I use filtered well water. You can check the ph of your well water and shock-chlorinate your well to bring it back down to 6.5 PH if it’s very high. You can also use laundry bleach to clean your well 1-2 per year if it’s smells like rotten eggs from sulfur bacteria.
Some people do but it’s not very consistent. You will be missing the nutrients and moisture provided by the grains. The grain also acts as a carrier agent.
@@SporenSprout will try that. Also I can add grains to my substrate. I see this method to be the best for anti sterile home environment. Of course I can be wrong, as I am brand new to growing and just learning every theory I can find.
Where I’m at I can only get mason jars 470ml, could i just cut the recipe in half and still get the same results? So 300ml water and 1 teaspoon instead of 1 tablespoon?
Hello, i wanted to try this method with a melmac Monster lc but it didnt work. The cloning worked fine ithink it was growing pretty quickly but when i inoculated it it Just wouldnt grow... After a few Weeks of Notting Happening i opened one Container to See If the Environment was good for mold and Stuff and surely enough after a week it was full of mold so i guess the Environment was good so im thinking maybe i stored the lc wrong?? Or didnt use enough? Would really apprecciate some help ✌🏻 Oh and im using coco coir and vermiculite with some gipsum as my substrate Just pasteurized it

Yes spore syringe works but you will have many different genetics. I use 2-5 ml of liquid culture(1 strain) not spores. it would be better for you to use 1-2 ml of spore syringe to cut down on the amount of strain variance.

Hi, just followed your video and looks like a bit of sediment at the bottom of each jar….600cc of water, (1) tablespoon of honey dissolved into hot water. Tops made the same as yours, covered /foil, 30 minutes at 15lbs. Should I start over?
I would start over if the honey water is not golden. There can still be a tiny bit of floaters as long as it’s still golden. I’ve found that overcooking causes the golden color to turn into a lighter color.
Thanks, I decided to start fresh with a new jar of honey!! Last one was a few years old from Block Island, RI.! I don’t have a magnetic stirring plate so I’m using (5) stainless steel ball bearings… maybe I picked up some oils from them. Forgot to give them a scalding bath. I’ll try again in a few and if same result then I’ll track down some glass marbles! Tough to find in downtown Boston!!
I just took out a bunch of mushroom stems from button white mushrooms because I'm not going to eat those, can I turn that into a jar of liquid? You probably have a video on it, I'll look but a link would be very handy
The amount of water you use isn't really necessary for liquid culture as far as the mycelium goes, it will colonize pretty much however much water you use. Just make sure you're using around a tablespoon of honey per jar of water. The ratios don't necessarily need to be perfect.
It is likely to become contaminated that way. I recommend the presto 23 quart pressure cooker from your local hardware store or grocery store. They are around $100.

Sediment is usually from over cooking the liquid media. You will have to keep an eye on it to see if the mycelium eats the sediment or not. I will never use any liquid culture that does not look like the ones in this video. I can tell a liquid culture is contaminated with yeast by observing the clarity of the liquid media, if it becomes murky and non-see through even when undisturbed then I know it is contaminated. Also by letting it sit undisturbed eventually any contamination such as trich should start growing on the surface as a clear indication. The best way to test it is to put a drop of it on agar plates and let it grow out on a 2D surface.

@@RugMann Actually rice should be good just steam sterilized as well as popcorn, but I would do at least 2 hours. Grains like rye berries, millet, and oats need pressure cooker because they are more nutritious.
@@BriggsWare Just use whatever will maintain 15 PSI for 30 minutes. Make sure the water level in the pressure cooker is touching the bottom 2 inches of the jars while pressure cooking.
@@BriggsWare just PC it a e longer. But for LC not much longer. 25 minutes tops! I use LME and it comes out clear and grows culture fast, in a days it starts a nice chunk... I use a protein shake mixing ball in it...
In my experience the mycelium will refuse to grow aggressively or at all in the LC. I always start over if it happens. Make sure to only use warm water when mixing the honey in and not very hot water.

Yes you can propagate the mycelium of morels this way but you have to make sure the genetics you are using are capable of fruiting without the mycorrhizal connection to a tree host. Also I understand that the method of cultivation requires implementation of forced nutrient deficiencies and addition of exogenous nutrient bags.

You will end up with many different strains growing together and will get inconsistent results. Put the drop of spore syringe on agar plates to narrow it down.
You can steam LC media and in a pot of water for 2 hours instead of pressure cooking for 30 mins but you cannot get around pressure cooking grain spawn.
I think i am misunderstanding the inoculation step. What are you incolatating the liquid with? Water mixed with spores off of a spore print or a small piece of the mushroom you want to grow mixed with water or something else completely? Also could i use tablespoon of cane sugar instead of honey?

I use 1g of LME to 600 ML of water for my LC's. Simple but yet effective. Get insane growth within 2 days from a LC syringe
If you add peptone it really goes fast.
Honey or corn syrup added helps also.

I do something similar for growing Cordyceps. I use a whole syringe to inoculate one small jar and that’s normal. It’s to allow faster colonization and works just like you are describing. Sometimes my rye grain jars start to dry out and I always think about squirting in more sterilized water haha keep up the good thoughts and I would try it!

They are good at 70-75 degrees F for a couple months and then they should be moved to a cold room (60 degrees F) or refrigerator. At room temperature they can last 6 months sometimes longer, and in the refrigerator they last over a year. Master Culture slants can be used to extend that window even longer to preserve your best genetics. Here’s the video for culture slants:

Thank you for taking the time to educate. How would you inoculate the liquid culture with a dried mushroom sample?
It would be unwise to inoculate it with dried mushroom. The only possible way is to put a few drops of clean water on the dried mushroom cap and it will rehydrate just enough to possibly drop spores on tinfoil as a spore print. Scrape the spores up and mix with sterilized water and inoculate agar plates and transfer until a clean plate is achieved. You can then add a wedge of the colonized agar to the liquid culture media.

It is ready to use whenever there’s enough mycelium for whatever your needs are. It can be stored for 12 months or longer at room temp or preferably in a cold room. Stir every other day at the beginning and then you can stir once a week.
I have 6 jars of liquid culture, various strains of mushrooms, that are all around a year and a half old. All of them are still healthy, no apparent problems. I will be taking a sample and transferring them to new LQ soon to be safe though.

I cut a wedge and open the lid to drop it in but you can try disturbing the mycelium on the agar with a scalpel and then squirt some sterile water on the plates, tilt it around and then suck the liquid back up and use that hoping for success but may take longer the grow at first.

rather late reply.... but you can also take a syringe, draw a little liquid from the culture jar, cut a small wedge from the plate and put into into something like a shot glass. Now mix the wedge and liquid in shotglass. Very shortly you should be able to draw the mixture back into a syringe and then back into intended jar.

2-4 ml is what I use for most species in any size. I like to use more to speed up the process, especially for slower species like lions mane or shiitake.

The magnetic stir bar is left in the entire time. After it’s full of culture you can store in the cold and stir every other week or so.

The foil prevents moisture from wetting the filter on the lid during sterilization.
It is fairly normal for the mycelium to grow only on the bottom of the jar and not further depending on the quality of the LC Media and genetics. If you don’t have a magnetic stir bar to break up the cloud then it will grow very slow.

Yes or even a wedge from an agar plate. Each method has its particular usefulness. Agar is good for longer term storage and isolating better genetics. Liquid culture and grain to grain transfer is the best way to scale the mycelium in bulk.

You can use a giant canner pot with a rack in the bottom with water and steam the jars for 2.5 hours. Yes I have grown tiger sawgill mushroom. It prefers a wood substrate, I recommend using the dub tub method but substitute the coco coir for sawdust. Here’s the video:

I got sediment after cooking it for 30 minutes at 15 psi. Do I take pressure weight off immediately to cool? Is it still ok to use liquid culture if there is sediment from over cookong?
@@Bonecarpenter. It’s because you didn’t use enough water in the pressure cooker. Make sure the water level is touching bottom 2” of the jars while cooking.
So I made two batches of LC, maybe 3 days apart. The first one, I definitely didn’t use enough light corn syrup(distracted), so the 2nd jar is developing faster than the first one. The first one seems to have stopped growing mycelium. Can I sterilize a corn syrup mixture and add it to the first one or just leave it alone? Tia
I like your idea and think it is worth trying as an experiment if you can successfully sterilize a concentrated syrup/water mix without caramelizing/overcooking the sugars and inject it into the jar. I’d make sure the jar doesn’t have sediment or cloudiness before attempting. I usually dump out jars that stall because most of the time it is not worth waiting for, when a fresh batch that is made perfectly will yield way faster and better results. Good luck!

I use filtered well water. You can check the ph of your well water and shock-chlorinate your well to bring it back down to 6.5 PH if it’s very high. You can also use laundry bleach to clean your well 1-2 per year if it’s smells like rotten eggs from sulfur bacteria.

Some people do but it’s not very consistent. You will be missing the nutrients and moisture provided by the grains. The grain also acts as a carrier agent.

Hello, i wanted to try this method with a melmac Monster lc but it didnt work.
The cloning worked fine ithink it was growing pretty quickly but when i inoculated it it Just wouldnt grow...

Yes spore syringe works but you will have many different genetics. I use 2-5 ml of liquid culture(1 strain) not spores. it would be better for you to use 1-2 ml of spore syringe to cut down on the amount of strain variance.

Hi, just followed your video and looks like a bit of sediment at the bottom of each jar….600cc of water, (1) tablespoon of honey dissolved into hot water. Tops made the same as yours, covered /foil, 30 minutes at 15lbs. Should I start over?
I would start over if the honey water is not golden. There can still be a tiny bit of floaters as long as it’s still golden. I’ve found that overcooking causes the golden color to turn into a lighter color.
Thanks, I decided to start fresh with a new jar of honey!! Last one was a few years old from Block Island, RI.! I don’t have a magnetic stirring plate so I’m using (5) stainless steel ball bearings… maybe I picked up some oils from them. Forgot to give them a scalding bath. I’ll try again in a few and if same result then I’ll track down some glass marbles! Tough to find in downtown Boston!!

The amount of water you use isn't really necessary for liquid culture as far as the mycelium goes, it will colonize pretty much however much water you use. Just make sure you're using around a tablespoon of honey per jar of water. The ratios don't necessarily need to be perfect.

I used to do the 2 hour steaming pot but after more testing I found that it is NOT reliable. Always use a pressure cooker.

Sediment is usually from over cooking the liquid media. You will have to keep an eye on it to see if the mycelium eats the sediment or not. I will never use any liquid culture that does not look like the ones in this video. I can tell a liquid culture is contaminated with yeast by observing the clarity of the liquid media, if it becomes murky and non-see through even when undisturbed then I know it is contaminated. Also by letting it sit undisturbed eventually any contamination such as trich should start growing on the surface as a clear indication. The best way to test it is to put a drop of it on agar plates and let it grow out on a 2D surface.

In my experience the mycelium will refuse to grow aggressively or at all in the LC. I always start over if it happens. Make sure to only use warm water when mixing the honey in and not very hot water.
