    In this video i demonstrate how to make a liquid culture of any mushroom species using Karo light corn syrup. This method is bare bones intro to liquid culture. Showing the most basic entry to grow mushrooms.
    EDIT:
    Use PAPER micropore tape, not plastic micro pore tape. You may use a steam canner (2 hours) to sterilize your culture media instead of a Pressure cooker. using a pressure cooker sometimes will caramelize the sugars (bad) due to the high temp.

    Red rtv high temp silicone- Lowe’s/homedepot/any car place. Use this instead of the tape and it’s a self healing injector port

      @@DAAI741 drill a separate hole for either poly fill (have to be careful not to get that wet) or look up hydrophobic syringe filters and google that with mushroom and you should find the size you need and videos on how to drill and attach to canning jars

      Its best to use liquid culture as spores take up to a week or so to turn into mycelium and spores are also usually contaminated so its best to do a brf tek with the spores, get agar, take a piece of clean mycelium from either the middle of a cake or the middle of a mushroom or something and place it into the agar, once it turns to mycelium you can use that to create your own liquid culture, you have a much greater risk of contam and contam is very hard to see usually in liquid

    Hi my friend I was just watching you on RUclips and I will say I was very impressed by your knowledge and willing to share with us. My son has been growing mushrooms for me to eat because of the research he has been doing about the health benefits of a few different types.... I have a question. We are in the process of making the jar myzilyume. We are in Utah so it's extremely cold. For two to three months his been trying to get some to grow. My question is how long will the myzilyume last in the jar before it's not good to grow anything. We want to be ready for when the weather changes and it's warm enough to grow them. We were trying for a long time and finally I told him to go ahead and keep the room really warm and let's see what happens. Two weeks later we were enjoying some kind mushrooms lol thank you

      Thanks for the kind words! 🙏 mycelium In liquid culture stored at room temp will last around 6 months to a year and if you refrigerate the liquid culture it will last longer than a year.

      Yes! Best thing iv ever bought for this hobby! I think it was like $30 on amazon. As others have commented the stirrer actually brings fresh oxygen into the mixture and speeds healthy growth!

    I’m a newbie at this, so thanks for the instruction…very clear and helpful! My questions: 1) Where/under what temperature and light conditions should I store the jars? 2) For how long can the jars be stored before going bad? 3) Can you use liquid culture (mycelium) from a syringe as well as the spores within a syringe?       Thanks for watching! Store colonizing jars at 70-80 degrees light doesn’t matter but darkness is preferable , once full they will last for 6 months at room temp and can and should be refrigerated to force the mycelium to go dormant for longer than a year.

      @@jeff042854 Liquid culture is just spores that have been germinated by introduction to sterile nutrient media. Spore syringes are made with only water to prevent them from germinating, they can be store for years before use.

      @@jeff042854 Also a spore syringe contains many different strains of the species. A liquid culture syringe is one strain. So the genetic differences will be wide with spore syringe.

      @@SporenSprout I did broke boi Tek about 2 weeks ago. I have vermiculite ready for the monotub with all the alcohol to get it ready.
      I took 11 cc's and divided them between six bags. Some of the bags look like they're about done and some look like they're about halfway done. How do I know when to move the cake to the mono tub? Should I do each one as it's ready, or do I wait till they are all done? I'm going to be watching your videos because this is the first one that I've seen, but if you can answer my question I would be very much appreciative for it. They are already talking to me, I'm excited!

      Same procedure. The spores will germinate inside the culture media and become LC

      If your talking about making your own spore syringe then no it's not the same. You will need a spore print, and you would not add the sugar to the water. There's videos here on RUclips that you can follow.

      You REALLY, FACTually, DO NOT-
      "NEED to buy, all the 'special stuff'- affiliated w/their 'operations'."
      There are MANY, many diff ways to go about "growing your OWN mushrooms"- whether for medicinal/food/fun purposes.
      A good place to start is the Reddit group! "r/mycology"- I believe its called.

    Hey brother thanks again for the content. I have an extra LC jar of Cordyceps but I’m not ready to use it right now. I’ve been told cordy LC doesn’t store well but I was hoping to hear your thoughts on how I can make this LC last a few more weeks.

      The best thing to do is keep it in a cold room. It will be ok there for 1-2 months.

    Hi there, thanks for the video. How many generations of LC can you use before it goes bad? Like you apply LC syringe into LC jars, then you use a bit of the solution from each LC jar to make another LC jar. Usually how many times can you do this before we have to start again?

I couldn't find my question in all the other comments, here it is: when storing the jars in the fridge do you put the jars in something like a sealable plastic bag to protect them from any nasties in the fridge, or do you just put the jars in the fridge as they are?       In a perfect world, you would have a dedicated mini fridge just for culture but to answer your question, I don’t use a plastic bag and I don’t know that anyone else does. If the liquid culture is super thick and full like it’s supposed to be, there is less room for contam also.

    Great video! I don't have a pressure cooker or a magnetic stirrer. I know they're widely used in mycology but I was wondering if there's any way to sterilize and stir without investing in those items? For example a make shift steamer using everyday household pans (instead of the pressure cooker). And simply physically shaking the jars (instead of using the magnetic stirrer). Any broke boi possibilities there?

      You can steam the jars for 2 hours in a pot of water with a lid. You cannot shake the jars if you use a micropore filter because the liquid will touch the filter or even leak from it. You can use a marble and swish it around the bottom to help break up the mycelium instead of shaking or using a magnetic bar.

    NOTE: If there is more than 4% sugar to water ratio the liquid can be come toxic to the mycelium/spores. It's better to use less than more if you decide to guesstimate.

      @@contambrah If the maximum is answered above, isn't the minimum, by default understood to be ZERO - but with no food = no growth. Anything greater than ZERO will allow for reproduction.

    One question: after all that, what's the best way to "duplicate" the LC? Should I take a few CC from the jar and pour them in other LC jars? How many times could I do that?

      Yes you can put a few ml in more LC media or you can put a few drops on a bunch of agar plates to multiply that way. There is no definitive answer to your question, it can be done many times until it stops performing well from old age, some species degrade quicker like cordyceps.

    total newbie here.... am wondering so say not to go spore to liquid culture but to agar first. Any thoughts on this? great tutorial!

      If you send spores to liquid culture, the liquid culture will have many different genetics so your fruit bodies will be not very consistent. When you send spores to agar you have the ability to choose the best growing sector of mycelium and transfer to a new plate. Then you can use a wedge of the new plate to send to liquid culture. Then your liquid cultures genetics will be more consistent. Not a big deal either way though. After replicating liquid culture many times you will have to start back again from spores to keep the culture fresh and strong.

      Only if you are inoculating some wood chips outside in the forest or something. Look up the term “slurry” for growing mushrooms.

    Hi thanks for this video! You said we could use a marble too. Does that mean we need the stirrer machine or would we just shake the jar with the marble in it?

      You wouldn’t need the stirring machine if your using a marble. Just tilting the jar back and forth is good enough for the marble to break up the mycelium that will clump at the bottom.

      Got a stir machine for less than $30. Its worth it. Make sure to put the stir bar in before sterilizing, but sanitize the bars first. I think they were the source of contam the first time.

      I wonder the same thing about how plants can be propagated in pure water and produce nice healthy roots, and then that same plant in soil with too much water gets soggy and the roots rot 🙄

    What is the difference between this method and your another method of a separate injection port plus air filter hole and pressure cooking with 15 psi for 30 minutes ? Also have a question about the regular 2 piece mason jar lid that rusted the filter hole overnight after i left the honey water in the jar for overnight cool off. i am using your liquid culture syringes which i bought from etsy. Do you think i will have to redo everything over again with plastic lids or it will be still okay.

      The difference is this video is I didnt use rtv silicone injection port.
      The filter hole was not rusted, you probably splashed the honey water and it came in contact with the filter. The filter should not get wet. Make sure water level is half way in jar, make sure not to over cook too hot, make sure not to splash. If the filter gets wet, dry it off with laminar flow air.

    Is the magnetic stirrer necessary, or is hard shaking ok? I didn't know if it somehow actually makes that much of a difference.

      Yeah the mycelium can get clogged in the syringe if you don’t break it up good enough

      You can use spores but you will have many different strains growing. Also spores are not entirely clean so you may have to test the final product on agar to see if it’s clean.

      Do not freeze them. You can use the refrigerator. But pink oyster specifically should not be stored in refrigerator.

      You can watch my other videos for information about how to use the liquid cultures. A syringe is used to extract the liquid culture after 2-4 weeks of growing in the liquid. Can be stored for years at room temp or refrigeration

      Keep jars around 75 degrees while colonizing. Once a considerable amount of mycelium has grown, you can store in the refrigerator or room temp and extract many syringes at once or whenever.

      Vu que le mycelium à ce stade est en phase de croissance je dirais qu'il faut env. 21C, et ensuite conserver les pots au frigo une fois que la colonisation est terminé. Je n'ai pas encore utilisé cette méthode donc je ne peux rien garantir!

    Amazing tutorial, man. I just tried this for the first time using my pressure canner and it ended up slightly yellow (caramelized) but not too much. Should i even bother inoculating it or just try again with a shorter cook time?

      I think you should start over. I prefer using a steam canner for 2.5 hours. But you can do 15 psi for 15 minutes in your pressure cooker and it should be ok

I think the two jars i overcooked cost about 14 cents in ingredients so its really not much of a loss. I appreciate the info and thanks again.

    I've got a small cloud forming in jar....now to keep it going, how warm should the room be for it to thrive? It's cold up here in Oregon, should I get a heat pad?

      Nice!! You should stir the cloud every few days so that it breaks apart into tiny pieces. I use a space heater in my room and set it at 70-75 F

      You can keep them in room temp for a few months but they should be put into refrigerator after you are satisfied with the amount that has grown. The mycelium will go dormant in refrigerator until you want to use.

      Yea just throw a marble in the jar before pressure cooking the jars for 30 mins at 15 psi. I used a magnetic stir bar in this video which is preferable

      You would inoculate grain spawn with the syringe and once the grain spawn is colonized, mix with substrate. Yes each bag will be identical. I have other videos that show the whole process.

    Those syringes that you started with are spore syringes right? I’m wanting to make some lc with several spore syringes that I purchased. I’m just getting confused when you call them “liquid culture syringe”.

      No I used a liquid culture syringe. Liquid culture is basically germinated spores. It is mycelium.
      You can use a spore syringe the same way as you would a liquid culture syringe but it is considered multispore inoculation and will result in many genetic differences.
