      Feel free to add more ingredients, you can use 1 gram of LME and .5 grams peptone for stronger solution. Just make sure there is enough water in the pressure cooker so that the water level is touching the bottom 2 inches of the jars while pressure cooking. I used low amount of ingredients because I kept getting sediment but found that I wasn’t using enough water in the pressure cooker which caused the sediment.

    I've been using this recipe for a out 6 months now, i use a little more peptone and malt extract but it looked like we get the same kind of results, as soon as you showed the ingredients I got excited, this is by far the best LC recipe I've used

      I once stored it in cold cooler and it ruined my LC. It wouldnt grow anything anymore.
      Can I just keep it in a kitchen cupboard for a few months? Did that only take you 2 weeks to have that much mycelium from just 1 cc of LC injected in that jar?

    Thank you for your videos! They are very educational.
    I am more familiar with your old videos. Trying to catch up now. I feel my knowledge is a bit rusty after over a year break from studying mycology. I had experimented little and due to health conditions I was forced to have a break and my LC spoiled meanwhile.
    I’m ready to restart but now all I have is 1 spore print and 5ml spores suspended on water. Unfortunately I am not able to get more.
    I have materials for grains on jars, bags, agar, LC, bulk substrate… What would you recommend I start with so I make the best of the little I have? For grains I have access to rye and popcorn by the way. I am interested in starting a good self sustaining cycle loop so I don’t waste the spores I have.
    Your help would be greatly appreciated. Thank you in advance!

      @@emekaalex288 Tht syrup provides food for the mycelium to consume and grow in within the liquid.
      That's what a liquid culture is. Then you just draw some up in a syringe and use the LC to inoculate grain.

    Good afternoon . What do you think about replacing petri dishes and test tubes with glass flat-walled bottles? It is essentially a hybrid of a test tube and a petri dish. The only thing that worries me is the convenience of isolating the mycelium strain in a bottle.
    I would like to hear your opinion.

      The Myc actually runs for nutes, instead of being surrounded by tons of sugar. Think it helps expand better @@PPP-on3vl

      Because most sugars like corn or honey you would use at a 4% solution. LME is a complex carbohydrate and can be used at far lower concentrations such as in this video

    After trying the original recipe with a 64th of peptone and LME and karo I just put together another 7 quarts of the suggested recipe for more nutrition. The first way shown in this video worked very well. I simply want growth for another round as fast as i can get it and plenty of nutrition to fill the jar with mycelium. The suggested recipe was 1G peptone, 1G LME and half tbsp of honey or corn syrup. I mixed the first quart that way and while stirring it i noticed it was definitely not completely clear. i lowered the LME to between.5 and .75 if a gram, not measuring it exactly just leaving the 1\4 teaspoon less than full and that solved the problem if the slight haze.
    Absolutely clear LC with that recipe. 1G peptone which is 1\4 teaspoon, .5 to .75G LME, light malt extract and half tbsp 7ml of Karo. I used karo only because that is what was there and honey seems to caramelize when i use it at 4% I have not tried it at lower concentrations, it might be perfectly fine.
    I wanted to list my findings for others who are trying to get completely clear LC but still wanting high nutrition.
    Thanks a million for all your help, especially the trick with adding 2 inches of water covering the jars in the bottom of the all American pressure cooker so I don't cause a haze after sterilization. It worked perfectly. I ran it between 12 and 13 psi after getting it to that pressure for 35 to 40 minutes and immediately vented. Perfect results.

      If you use filtered air through a Luer loc filter disc you will see the mycelium grow in huge leaps and bounds. One in and one out and a 45 dollar aquarium air compressor. I say the 45 dollar one because you will want numerous jars going at one time.

      I have all of those parts right now.
      I use a blunt tip needle that goes all the way to the bottom of the jar so putting air in that should cause a lot of oxygen going through the liquid and to the bottom of the jar.

      @@ZCRAIZED1 Excellent my friend! In Early 2000's I was part of a company in Kennet Square Pa. that had a culture collection that rivaled the National Type Culture Collections. I started cultivating medicinal and gourmet mushroom spawn that we supplied to large mushroom grow houses. With fresh air I had to double the amount of peptone or malt extract because the mycelium grew so fast. We ended up selling the company to Campbell's soup and I suppose it still exists. It was called Mushroom Science Laboratories. keep up the great work.

    So the idea is to sterilize the f2ck out of the jar and fill it with some liquid carbs? Would you consider trying a really crude version of this? Like wiping down the jar with some off the shelf isopropyl alcohol and then dumping some sugar water in with some innoculant? Idk I think it'd be cool to see if it works. Anyway, really cool footage.

    I didn't see this and as usual I mixed everything in hot water, the peptone took a small amount of agitation to get it mixed but all three ingredients were very easy to mix in hot water. LME and I'm using light Karo just because that's what is here. I have honey but at 4% it badly clouded and a lot of sediment. I'm guessing I caramelized it with too much heat and too long of a temperature.

    Hi, I appreciate your concise and simple instructions to create liquid culture. What is the suggested time to let the culture grow mycelium or is it by sight? (i.e. 50% of the liquid has mycelium) I'm trying to build up my culture backlog and extend my spore purchases. Thank you!

      Yes I’d shoot for 50% and it usually takes 1-2 months

    i love the clearness and very fast growth of this recipe. thank you! however, most of my strains now start to make significant rafts. i first thought it was contam, but surely they are not all suddenly contaminated. it gathers up in big floaters on top that is very hard to break. have you experienced the same?

      @@SporenSprout i was thinking if they get a bit too much nutrition still? have a feeling it could be related. not feeling super comfortable with these rafts, and the additional step of blending i would also like to avoid. what do you reckon? i'm coming from honey LC and that's been working fine, but i feel the clarity of this method makes it easier to spot bacterial contam.

    This might be a dumb question but I'm a newbie so please don't make fun of me lol. Is it possible to take liquid culture that has already been formed, and inoculate another jar with it to allow it to grow? Essentially can you use one jar to make more jars?

      Yes you only need 1-2 ml of liquid culture to make a whole new jar and you can keep doing that for a while.

    Thank you for this recipe. What do you think about using powdered corn syrup? Does mycelium have a preference of bone peptone over soy, or vice versa?
    Do you feel that this broth will work for pestilotiopsis microspora, initially anyway?
    We are excited to find out how this works with all of our different species. We have a number of projects going on both professionally and personally that this may be a nice addtion too.

      I’d go with soy over bone. The bone peptone makes the mycelium grow on the surface too much and also smells bad. Yes I think it will work great for any microorganisms really. I have never heard of corn syrup powder but for me corn syrup is never strong enough. I like honey.

      It works well but if you don’t stir it everyday the mycelium tends to clump and tangle which can be annoying. I prefer honey LC for that reason.

    My presto states to fill 3 quarts of water when using it.. does the LC recipe glass mason jar have to sit completely above the water during the 20 minutes or is it ok if the bottom portion of the mason jar is submerged in the water?

      An inch or 2 covering the bottom of the jars is normal. You can use jar lid rings to help raise the bottom rack further if needed.

      Yeah. Next time germinate the spores on agar first.

    Great video and thanks. Thats 1/64 tablespoon. A teaspoon is about 1/8 of a tablespoon. So 4x that small one you got there or 1/4 of the big one (the size of a nail) would be a teaspoon.

      Why does the spoon say “1/64th TSP” right on it then? It would say TBS if it was 1/64th tablespoon. Either way it doesn’t matter, you can use more nutrients than that. The only reason I used such small amounts was because I had sediment but I found out it was from not using enough water in the pressure cooker. Make sure the water level is touching the bottom 2” of the jars while pressure cooking.

    Thanks to these LC videos promoting this bacteriological peptone, Amazon is sold out. I decided to order the soy peptone instead.
    Given these results from something sooo diluted, we all need to promote this recipe so there's more peptone to go around as this mycological revolution ramps up 😉.
    Good to know at least that LC solutions can be successful over such a broad range of concentrations. I don't think I'll even bother measuring after watching this; just a couple of pinches of each to be sure I have at least this amount, but obviously waaay less than other recipes I've seen.

      Doesn’t matter really, just use whatever you have to stir it at the beginning.

    Hi what an informative video. thanks, I am making mine to your recipe and need to know what temperature to store the jar at?

      You can store them at 70-75 F. When mixing the ingredients you should start with room temperature water. The ingredients do not dissolve well in hot water. You can use 1/4 teaspoon (1 gram) of each powder as long as the water is not hot. It should dissolve perfectly in cold/room temp water. It also helps to reduce PSI from 15 to 13-14 to prevent opaque cloudy LC media. Good luck!

      I wish I would have seen this video last night. I only saw the short one and didn't catch the statement about watch the full video. I made 7 quarts of the lighter nutrient content. I'll be making this heavier recipe tonight if I can figure out what to do with my all American canner to get it to not make the mixture clouded. I'll definitely take your advice about running it at less than 15 PSI.

    ive had cultures last 3 years in a dark cool closet..........dont disturb the water wall inside and the mycelium will give off a digestion enzyme that sterilizes the water and essentially puts it in stasis.

    Hello! First of all - amazing video and results! I would use some advice if you don't mind: I'm using 8 grams of fructose in 200 ml of distilled water. Sterilized the solution and inoculated it, kept the jar wrapped in the aluminium foil at 24-26 Celsius. It's been 4 days now and the only thing that appeared in the solution is a very thin and short transparent (or white, it's hard too tell they are so tiny) strings of supposedly mycelium. Also the spores (they appear as very dark brown dust) are doubled in size.
    Is it going well and I should proceed with waiting or the result I have now (4 days) is not enough and I should just scrap it and start all over again?

      It might be ok but spores are not very clean to begin with so it’s not adviced to inoculate with spores. You should germinate the spores on agar first and then transfer to LC media. You will need to check the liquid you made on agar to know for sure if it’s good

      can boil in a pot (with lid, creating steam) for 90mins. make sure the water is a couple inches above the jar and u put some sort of rack on the bottom of the pot.

    Is this sterilisation is 30 minutes at 15psi? .should we Weight until the pressure cooker turns to 15 psi and the 30 minutes or all together 30 minutes from the minutes I started pressure cooking?

    Question: I followed this recipe but I forgot to tighten the lids after I took them out of the pressure cooker. I left the foil on top though. I took the foil off 3.5 hours after removing them from the pressure cooker and noticed the lids were loose so I tightened them at that point. Do you think the jars could be contaminated? Let me know if you think I should start over.

    I tried this exact recipe which looked amazing and the only thing I didn't do was use your brand of malt. The corn syrup and the pep tone was the same brand from Amazon. I used Anthony's malt and it will not dissolve. Right now I'm not sure if it's the malt or the peptone but something isn't dissolving no matter what I do with the magnetic stir. They're the same color so it's hard to tell. And guess my only other avenue is eliminating one and then the other to see if they work at dissolving entirely in the liquid

      Increase the amount of water in the pressure cooker. The water should be touching the bottom 2 inches of the jars while pressure cooking.

      I did a jar without the Anthony's malt and it dissolve fine with the magnetic stir before pressure cooking. I'll try your suggestion on that with the one that still has Anthony's malt in it

      It's very easy if you haven't already figured it out. You just cut the area of agar that you want and drop it in the jar.

    Question so Ive seen online that people have said you only need a few drops of a spore syringe to 1ml. So what is the truth can you get away with less ? Or do you need more spores

      A spore syringe is not perfectly clean so you should only use 1 ml. If you germinate and clean the spores on agar and make liquid culture from it than you can use 2-5 ml

      No, after inoculating the jar it is allowed to grow at 75 F and once it’s full of mycelium you can put in the refrigerator but I prefer to just move them to an air conditioned room instead

    if I'm using half gallon mason jar should I still PC at 15 psi for 30 mins or go longer than 30 mins? also if using spores syringe, how many cc should I use for the half gallon jar if I need fast colonization?

      30 mins is still fine. 1-2 ccs although spores are not guaranteed clean. Best to germinate on agar first and then transfer to liquid. You can do it though, just make sure to test a drop of the final product on agar.

      ​​@@SporenSprout doing agar currently is not possible, I got those spore syringes from a very reputable supplier that I had successful spores to grain 10 years ago from their syringes, I'm also following a very strict hygiene including sterilizing neddles in PC and inoculating in well sanitized closed area and using self healing port like the one in video and for the GE I'm using 4× 1/4 inch holes and on each hole I'm using tyvek and on top of it GE filter disk..

    Is time for growth the same regardless of what mushroom you’re growing? For example I thought Cordyceps were ready after like 4 days tops?

      With this recipe the mycelium initially grows faster but will not completely fill the jar like other recipes with more nutrients like honey. Cordyceps can grow for 1-2 weeks

    Is boiling tap water sufficient for this method or have you got any advice on how to distil water at home. Having a terrible time getting LC to work with honey currently so definitely going to give this a go!

      If you are having trouble making the honey LC media. I recommend making sure the water in the pressure cooker is touching the bottom 2 inches of the LC jars. If you had the water level barely touching the jars or not at all then that was your problem.

    Just curious, could you explain why this Recipe works with just a fraction of the nutrients vs all of the other ones that use grams of this stuff?

      The low nutrients force the mycelium to quickly expand in search of more food but won’t grow as full. You can use more nutrients and that’s fine. I just had problems with cloudy media when using lots of nutrients but then I found out that my all American sterilizer is what was causing the cloudiness so now I only use my presto 23 quart pressure cooker for LC media.

    My first LCs (3 jars of golden oyster, and 3 jars of lions mane) are 2 weeks old now and seem to have stalled. The golden oyster blob rises about 1/3 of the way to the surface, and lions mane is only about 1/5 or 1/4. This video inspired me to try a milder solution than most LCs recipes you find, although by my math, my recipe is still 5.5x richer than this (1/16th tsp each of DME and soy peptone per 350 mL jar). I'm also running 2 jars with a sloppy half teaspoon of honey added to make note of any differences, and so far there aren't any.
    Another difference is the fact I'm using a 0.22 micron syringe filter installed in the lid to provide the FAE. Do you suppose the syringe filter provides inadequate FAE for a 350 mL liquid culture in a 500 mL jar? I've given them a vigorous stir twice now. Do you think perhaps I need to stir more frequently, or for a longer duration? Or perhaps I'm just being impatient. What level of growth should I be seeing at 2 weeks?

      The lid has 2 holes. 1 hole has RTV injection port and the other hole is covered with 2 layers of micropore tape.

    I've been using a recipe that uses about 1/2 tsp peptone and 1/4 tsp LME, but the same amount of corn syrup and water. You've found these much smaller amounts of peptone and LME to be superior to larger amounts like that?

      Yes in my experience the smaller amounts work better. If you can fit more in without turning the solution cloudy than keep doing that. If there is any cloudiness the mycelium will be initially inhibited in my experience. I’m thinking a hot plate will allow you to dissolve more without it compromising clarity.

      @@jobbingactor you can use more ingredients as long as you make sure the water level in the pressure cooker is touching the bottom 2” of the jars while sterilizing. The amount of nutrients does not have a big effect on shelf life.

      @@SporenSprout Credit where its due your one of a tiny few you-tubers that takes time out there day to post replies and on top of that, very quickly. It doesn't go un- noticed. Message received. Ive prepped some jars very similar to yours, I had Malt yeast extract but no peptone so went for a gram of yeast nutrient. Yesterday's jars were clocking on for 20g per litre so I watered those down with distilled water to about 5% and will PC them again.

      Just use a gram of LME and .5 gram peptone and make sure to use presto 23 quart with 3 liters of water in it. I used low ingredients in this video because my all American pressure cooker always ruined my media by making it cloudy and I didn’t realize that for a long time.

    Questions about stirring: You say to stir once at the end and then let sit for a week. Then stir everyday after that so it doesn't form at the surface? How many days do you stir until? When do you put it in the cold room? Do you stir once it's in the cold room?
