    600 ml H2O
    18 g corn syrup
    1.2 g light malt extract
    1 g peptone
    Stir to dissolve.
    Add stir bar (or stone)
    Cap jar and wrap lid in foil.
    Pressure cook @ 15 psi for 30 minutes.
    When completely cooled, inoculate.
    (Took notes for myself, but also wanted to say thank you for the video!)

      How do you inoculate this mixture with mycelium? Can you use part of a fresh mushroom?

    • @TheQueenOfDreams
      @TheQueenOfDreams 5 месяцев назад +9

      @@cyberbid not my video, but I use it to culture a LSS into mycelium, and then I put a sample on an agar plate to test for contam. If it grows clean, I go back to the liquid culture and set the mycelium to grain spawn.
      You could use a piece of a fresh cap, but might have a higher risk of contamination. Good luck!!

    I seem to have done almost the same research you did and went about things very similarly. However I used dry corn sugar, the four pound bag, and made a pre-mix with the dry malt extract. I also cooked for 40 minutes because I figured a darker solution would be good for the mycelium, but it certainly is harder to see what is going on.

      I've recently seen a few videos where they REDUCE the nutrition in the agar mix, and claim it increases chance of rhyzomatic mycellium. The idea is reduced nutrition causes the mycellium to have to stretch out to find nutrients. Too nutritional of an agar mix might cause nothing but the fluffy pomentous (sp?) Mycellium to grow in your plate...

    • @tdtrecordsmusic
      @tdtrecordsmusic 10 месяцев назад +1

      they all have different outcomes for what u need to do. if ur just starting, u really can use anything that has sugar or starch. ur main concern is cooking to sterile. getting ur stuff clean & sterile is the main concern. after that just experiment and take note of what happens. yeah, LME is a luxury/not needed.@@SachaLotus

    • @tdtrecordsmusic
      @tdtrecordsmusic 10 месяцев назад +1

      potato is hard to get clear if I remember correctly, so if u need to see whats going on i would suggest something more pure like karo. i do remember potato being fast tho. the myc said yum yum, i like potato. the karo, is a little slower(but crystal clear) and can give more time to work like do an isolation or separation.
      i use a clear LC to find the winners and make sure everything is clean. once u get a colony that is hardy hungry and ready to rock, use a nute mix that has fast growth for expansion. the LC mixture that is used for expansion might not be clear or have a good color, but it'll get consumed fast so u can achieve a good volume. ofcourse, its vital to get it clean first so u gotta b able to SEE it in the beginning. i suppose u could do all the work on agar, but doing that work in the LC is my goto technique. plus agar is a lot of steps.. i do all that in the LC and have gr8 success. I like the small surface area of jars instead of the wide agar plates
      consider > those little guys are hungry and stuck in their home. they will eat whatever caz they gotta survive. I bet even white sugar will work.
      p.s remember to leave one jar NOT innoc'd as a control so u know ur technique was good. do all same steps, just no innoc.

So wish me luck and hopefully they all do well. I’m still gonna do some agar plates to test for contaminations in the near future. Great tip about leaving one LC jar not inoculated to test for contams. Mush love from Peru 🇵🇪🙌🏼🙏

    What kind of mix is that?
    Here in Germany it is relatively difficult to get peptone. It is simply reserved for laboratories.
    But I got some and did this LQ. Within 12 hours a solid mycelium carpet has formed. How fast is that !?!?

    If I use this recipe and add agar powder to set it. Will this work as petri agar and slants?

    Can you use glucose syrup instead? It’s difficult to find corn syrup in Denmark.

      Same thing, I believe. From everything I know, you want a glucose source. Hence, why the dextrose in potato dextrose agar.

    How do you visually identify contams? Mold looks so similar to mushroom mycelium, and I have some growth that looks like mycelium but is... threaded differently idk how to explain it all clumps with the normal looking mycelium too
