      How are the B+ doing for you? I just ordered some prints and have heard great things. Only have experienced the Tidal Wave and Golden Teachers to date. Both great!

      @@masonreed5684 it doesn’t matter which variety you grow of Cubensis. The process is exactly the same…exactly! A cube is a cube. The only difference are PE’s, and that’s when you spawn them to bulk because you need to case them or you get blobs and mutations. But the LC and agar process is the same. PE (penis envy) take longer to colonize as well.

    I make my own maple syrup, I use that in my agar and liquid culture. They seem to love it and stays very clear.

    Hi Teach, I just wanted to say thank you for your videos. I successfully harvested 870 wet grams of mushrooms on my first grow from your videos. :) I now have decided to take my grows out of the closet and into the entire spare bedroom. I ordered most of a mushroom lab and I am looking forward to grow #2 and then further agar plate/slant work and liquid culture production :)

      @@commiecrusher I have tried them, probably before you was born. Not that it matters. My experience wasn't amazing was more on the nauseating side. Now I just take chaga, lions mane and reishi. I'm currently trying to grow cordiceps.

      I wish people would just say the amount in dry weight, I know wet is a bigger number but it's less accurate since water content varies.

    The Presto PC is nice man. I want to thank you for all the tuts and all the knowledge. I’ve been doing this for about a year now and I’ve made quite a few adjustments based on your teachings. I strictly only grow APE, PE uncut and golden teacher now. Not a day goes by where I’m not working on Mycology. 🙏

      I have the 28 qt Presto and It works very well. I just started doing LC's and for 1 or 2 quart jars, I use the instapot. Wife is not happy about that though. I'm going to start using 1/2 pint wide mouth jars for LC's rather than Qts.

    Thank you sir, and everyone else out there for sharing tips, tricks and knowledge. In a lot of ways it's similar to the 3d printer circles. Great thing about having so many teks and personal tweaks (for all skill levels) is that you can "Chinese Menu" stuff. A 5 gallon bucket for cultivation, someone else's personal substrate mix, and little add on/tweaks/personal touches from 3 different people.

    Nice one PGT! I prefer to use Li (liquid innoculant) for anyone thats interested it is sterilised water and mycelium (no nutrients) straight into grain.
    I do the step you do at 8:55 with the agar plate and go straight to grain, i find tjis method excellent.
    I never thought about sucking up the mycelium directly from the agar dish until i watched this so thank you for that.

      Just put some lc on agar and isolate the clean mycelium takes extra time but beats contam right?

      @@sheeeitmayn4384 well surprisingly. I made two different LC''s B+ and Pink Buffalo. The PB spores LC I pulled them into 10cc syringes and I had about the same amount of contaminated jars as I had just using spores. That's using an SAB.
      BUT.... I just did up some of the B+ using same exact method and so far I'm almost to 100% colonized in my grain spawn and so far I have zero contamination. Only thing I did different was instead of inoculating jars in my kitchen I did it in the bedroom. And TBH I thought it would be a disaster because I keep reptiles, and I have had issues with cobweb mold before I got into growing mushies. But everything is going good so far. I did do a serious deep clean though because the web mold is super annoying just in snake cages. Especially in mine because they basically are perfect environments for growing mold. I don't use glass tanks. They're a humidity nightmare if you want it be consistent.
      Also I'm still working on probably going to end up building my own flow hood. Still doing the proper research on how a proper one is built. I almost went full on with that myco willy dudes build and learned that he doesn't mention a pre filter and I'm told this could mean the difference in getting months vs years from a HEPA filter. And right now we all need to save as much money as we possibly can.

      @@ChondroMan so you had some luck using a spore syringe straight to liquid culture? Im at the moment going to agar and brown rice flower jars with my spore syringes but was wondering if i could shoot some into LC jar and it work out fine??

    Great video! You having the agar open while you scrape is keeping it open and exposed while cutting a chunk of agar and dropping it in keeps exposure to a minimum.

    Started 2 GT and 2 B+ dec 9 and 4 more of all PE the 22nd. One gt and one b+ didn't even spawn. Other two gonna go to bulk in a day or two. This my first ever grow. Also got agar plates and a microscope with plenty of slides. Following a lot of RUclips videos and I'm having an absolute blast. Thanks PGT.

      @@chrishudson4940 always works quite well for me and the spores seem to enjoy themselves. Plus I get a visual indicator when all the suagrs are consumed as the fluid goes from a golden hue to clear

      @@chrishudson4940 I im always a big advocate for baby steps. Especially since once you get your first flush you can make a spore print and make an LC from there. Take your time, do your reaserch and everything will be fine!! Best of luck! Plus golden teacher gives great advice!! Can't go wrong listening to this man

Right now my outdoor cyanescens patches are keeping me pretty excited. One of the patches has hundreds and I think will put up a thousand mushrooms this year. It’s so damn cool

    Easy way to measure your 4-5% of sugar nums is to just use a syringe. 300 ml mark on a regular pint size canning jar and so a 12ml syringe of sugar nums will do.

    The reason I asked because I have an agar slant of wine cap dried up in the fridge for 9-10 years. I tried several attempts recusing the culture and finally have it on a single clean agar ketchup cup. The rests are not growing, contaminated, partially grow/partially infected. I used it to multiply on mor agar, all fail but one second ketchup cup. I do not want to repeat the process again but go straight to LC. I am afraid to fail and lost them all. 😢Thank you.

    I just ordered some premade agar plates as my plan was to go from spore syringe to LC. I just started learning with oyster mushrooms and didn't have to worry about that step. I'll head over to your vids on that now lol thanks for the info

    Thank you for helping me along this learning journey! I did go from Spore syringe to Liquid Culture, and so far, no signs of contamination. I don't have anything inside the jar to help with the stirring so not sure if that will hamper growth or trying to extract any LC since it's not being broken up for easier getting.

      Drag the needle through the glob you want to pull and then suck up what you broke off. And this is what took to the millet the fastest.

    After you have removed mycelium from the agar dish, do you use the same agar dish to populate more liquid culture jars until no visible mycelium are present, or is it best to discard that agar dish because it's been exposed to air? Thank you for your amazing contributions to our knowledge.

    Sensei! I love you videos! I recently started using your recipe with peptone and using Boomer shroomers method of inoculation (taking part of the fruit through a syringe and injecting it in) and every jar I’ve had is cloudy. While I’m sure most of the problem is my sterile technique, I was wondering if you or anyone else has anyone tried this method of inoculation with this recipe? Is there a reason you inoculate from agar only? Thank you so much!

    Let’s say I got LC syringe from a PGT recommended supplier, can this used to inoculate or does it need to go to agar to be screened for contam? Seems like they would have followed a similar process and it already be clean?

      You can expand LC to LC. It is always best to test things or start from a clean sample off agar. But to answer your question, yes, you can use it to inoculate 🙏

      yes it does, but it isn't like you never use alcohol directly to it

      If you spray alcohol directly on. It will harm and bruise mycelium.

    So.. dont do spores>agar>liquid culture/slurry>grow?
    What would the better option be?
    Inoculating several agar plates from a single syringe>isolating clean growth> isolating aggressive mycelium>liquid cluture/slurry>grow?

    You mention loosening the lids before you pressurecook them, do you tighten them after taking the lid off the pressure cooker (Once you've let them cool down?) I'm Using your video as a guide to grow Yellow Oyster mushrooms.

The question is: After storing liquid culture in the fridge, does it need to be back at room temperature before injecting it into grain jars for colonization?

    Pssssssst - FYI PGT Yikes!! @ 9:19 you stuck your 18g hypo needle into your HEPA filter creating a hole of appx. one thousand six hundred and fifty microns... that filter would fail certification.
    You might want to squirt some silicone caulk in and around that hole.

      @@harleyjackson4208 It did penetrate but the reality is you can't even touch that extremely fragile filter media with something sharp, specially a hypodermic needle without compromising the filter integrity as HEPA filters are tested to

    Hi Sensei, I have watched all of your video's and was wondering can you just make a clone through LC instead of going through agar first. Wouldn't it be the same process? Of course it would have to be done in sterile conditions. Thanks for your time, take care and be safe.

      @@ozoneswiftak yes, took 6 different samples placed 3 into LC. Did all work in my SAB and apparently 2 got contam somehow. 1 jar is chugging along. Put the other 3 samples to agar, working on 3 plate to get the best sample to send to LC. Hope this helped.

    I got a 23 quart pressure cooker but it’s really old and I can’t find a new gasket for the model number I got. The ones I bought were just a little too big. Might have to just buy a new pressure cooker Got this one from my parents and it has got to be over 40 years old

    Thank you so much for a simple but effective recipe for expanding LC. The recipe I've been using with Soy Peptone just doesn't work for the Black Pearl King Oysters. :)

    Can I do the same with pepton , light malt extract, corn syrup to scrape off the mycelium off the agar? Also can I instead puncture the ketchup cup instead of removing the lid completely & squirting in the mixture in and scrape off and suck back out into the broth ?

    Hey, i'm currios, can you inoculate your jar with spores? just like the mycelium just scraping the spores from the sporeprint.

      It is possible but not ideal. Spores are inherently dirty with bacteria and potentially mold spores from exposure to open air. Ideally you want to clean up a culture on agar to use

    I'm debating what's a better method? The 250 degree oven or the SAB. I feel like the oven is better. I'm not going to be dropping the coin for a flow hood any time soon! One day maby. Btw love that you reply to comments man, I'm shire I'm not the only one who greatly appreciates it!!!!

      The oven can be easier to work with because it is open to moving freely. But the SAB is better practice and suggested! 🙏

    PGT, I had some bad luck with grains that were not sterilised long enough, they all colonised with LC but with contam. This time I did sterilise everything right, used LC from the fridge (had been standing there for a 6 to 7 weeks or so)... nothing is happening unfortunately.. 2 weeks in.

      Maybe the mycelium was damaged too much as I shook the jars streight after inoculation. Some jars I did not shake (like the unmodified lids as there is a higher chance for contamn) and those do grow a few tiny bits of mycelium.. so it should still be alive somewhat

      Make sure you have proper gas exchange, otherwise it will stall things out.

    Konnichiwa Sensei, excellent! Now maybe I can get a successful LC. Q: Does powder dextrose work? how much for 500ML? You mentioned LME. How much? How does the LME aid in LC?

      Lme aids in providing nutrient. About 1.2-1.5g in 500ml solution.

    If you are using agar you want to grow the agar out for 2 weeks to make sure that there is no contamination

    Can we just put distilled water in the syringe and put it inthe agar and scrape those off and re-suck those into the syringe for a one time use instead? If I don't wanna a lot of it in a jar.

    Have you used the latest corn syrup craze? It's got vanilla salt And ethyl alcohol.
    Will that crap also work? Can't find old fashion corn syrup anywhere besides online

    You can also just keep the agar in a sterile jar with an injection port. Take from the un-inoculated culture and mix it with the agar, then just transfer it back to the liquid culture jar though a injection port.

    What are your thoughts on squirting water in a jar of rice culture and sucking it back out if you don’t have agar?

      It's risky. But not impossible. I personally wouldn't do it. You might as well just do grain 2 grain transfers at that point.

      Because spore solution contains only spores + distilled water, nothing nutritional.

    Sorry if this is a dumb question, I'm just learning. After you have the LC colonized do you add that to your sub for fruiting? I've never tried this. I always use rye berries to colonize. Thanks! Love your videos... so much great info! ✌🏼

      i think a lq is still for inoculation of grains. This is just the absolute fastest way i think. (im also new so maybe someone more experienced can better answer)
