Vaders face before he kills her is both sad and funny at the same time. Sad because it shows how depressed of a character he is to the point where he doesn't care if anyone has anything positive to say about him. Its funny because he doesn't kill her immediately and waits until see lets her obsession with him out of her mouth before stabbing her. This might imply that it isn’t the first time this happened to him and if so, good boy Annie for staying loyal to Padme.

This story is textbook example of Limerence. What it is for those that do not know, it is a romantic obsession/hyper infatuation with a love object (it can be a person or thing). Instead of being true love, limerence is an imitation. It treats the love object as the savior or solution to a perceived problem. If I just had that girl or guy everything would be perfect, then I would be happy, or if I had that body or lost this much weight then my life will be perfect etc. It's all fantasy, it can also be considered mall adaptive daydreaming.
One thing that I think is that Vader probably knew about these feelings from Nurse since he is able to sense the surrounding area for feelings even he doesn't see the person. This would make sense because Vader usually kills anyone on sight the minute they step into his quarters but he didn't do it to instantly to her and let her speak which makes me believe that he knew.

While she did eventually fall in love with him, she came to realize that "Mr. J." viewed her as more of a useful minion than a romantic partner. As for this Imperial nurse, yes, she always seemed to harbor fantasies of being Vader's lover despite all evidence to the contrary, and it took nothing less than his crimson lightsaber through her gut for her to understand that it was only a fantasy.
