    I'm glad someone finally covered this. I'm in the process of doing this in a couple days. I was wondering if all the parts were autoclavable. Thanks for finding out for me. I plan on using a one-way check valve instead of the stop-cock. Until now I used 8 gauge metal dispensing needles, but epoxied them to the lid so they couldn't spin or move. I also use UV glue wherever I can. Keep making great videos, we need them.

      Yeah, well it was kind of crazy because while it was chugging along at 15 psi, one of those red caps popped off of 1/4 LC jars I made and it was shootin steam out the top of the jar hose so much so that it made the pressure force It's way out the weight valve I almost jumped out of my seat because I'm a quadriplegic 😂but that one jar it happened to was like 150 mL low so it was like a mini little pressure cooker, but inside of a big pressure cooker 😱😱

      Why not just use regular lids during the PC and then swap them out for these in front of the laminar flow hood after ?

      @@goodtoknow1331 any time you open the jar after it has been sterilized you are massivly risking contamination. Even in front of a flow hood. Also if you use regular lids (i assume you mean regular canning lids) they will be vacuum sealed to the jars. That requires a bit of prying to get off. The whole point is to never have to open them after thye have been sterilized. I thnk that is why he also has the inection port so that he can use it to inoculate. If you used the tube to inoculate you would probably have to flush the tube out (down into the jar) with sterile water in order to get the inoculant MYC down into the nutrient liquid. Otherwise it would only be in the tube.

    Great Video Very Good Idea I am growiing in the tropics where contamination is a huge problem I will try that solution. One question though. Is it possible to also inoculate the LC through that silicon hose? Or will too much stuff get stuck in the hose?

      So for me, when my jar is ready to extract I prefer to pull it all in one sitting. Once the mycelium has colonized the jar to my liking I will empty it into as many syringes as it will fill. If you're only filling a couple you can try to sterilize with ISO, or you just put a fresh sterile stopcock on each time (which will cost more down the road).

    super helpful Content. i have 4"x14g Stainless needles im currently using for my LC, and this is a great way to not need sharps while also being able to reach all of the LC in the jar with less flame sterilization. can just buy blunt fill needles in bulk for inoculating Grain

    I was having trouble sourcing female luer to 1/4 barb adapters available on Amzn that were polypropylene. I opted with male adapters which will still connect with a 3 way stopcock and leave 1 connection available for a syringe at the stopcock. Throwing this solution out there to anyone having supply issues with the female adapters rn.

    Great video but quick question, how do you think inoculating through the stopcock instead of through the injection port would go?

      If you're inoculating from another LC it would work just fine. You could also sterilize some water, draw some into a sterile syinge and then use the needle to basically biopsy an agar plate then inject as well. Just need enough of a sterile carrier fluid so that some mycelium makes it all the way down the pickup tube.

    Use a high temp flow control clamp, that way you can pc the whole setup.

    When you finish your jar, what have you found to be the most efficient way to re-sterilize the stopcock for next use?

      These are one time use, if you want to spend the $ on stainless ones you can just chuck them in the PC when you sterilize your grains

    Why do we need an injection port? Can’t we use the stopcock to Inject instead of pulling?

      I use the injection port if I have a known clean LC I want to expand.....or if I just want to draw one syringe to agar test. Once you use the stopcock it's no longer sterile. Me, I will empty the LC jar into syringes in one sitting.

      Best practice is these are one time use…for me anyway. That’s why I just empty the jar in one sitting and store the syringes in the fridge until I use them

      @@RealSimpleMushrooms Awesome! Will that filter patch suffice for air exchange? A friend of mine told me to never fully seal jars because its probable it can burst in the PC. I'm wondering if the filter patch mitigates that risk

    Is the stopcock necessary? What if you just took the red luer lock off and hooked the syringe straight to it without the stopcock? Also, how is the stopcock cleaned and used again for next jar of LC? Is this a one time use? Since you cant autoclave it, how do they get sterilized?

      It’s not necessary, I like to use them in case I don’t empty the entire jar into syringes in 1 sitting. As far as re-use they are a 1 and done deal.

    You did not list the injection ports in the materials and you did list syringe filters but did not go over anything about them in the video but had the jar with them on one. So exactly why did I purchase them if I'm not using them and you didn't show what they were for?

      So I use the injection ports for 2 reasons.
      1. in case I want to inoculate the LC jar with a current syringe I know it viable and clean
      2. Sometimes I will pull a quick sample through it to agar test it for contam.

    Hi I’ve been using these lids a couple times now and they’re the dream. Just wondering if it would still be sterile to refeed through liquid culture through the luer lock with PC liquid culture solution syringes? Or have you already tried this?

    Very interesting, does that stopcock leak over time? That'd be my only concern because if it doesn't stay air tight at all times gravity will pull the liquid down and draw in dirty air

      So for me I will just empty the entire jar into syringes in one sitting, but I also sell cultures. I'll store them in the fridge to slow the growth but they last upwards of a year in the syringe as well

    Instead of home made jars, you can use dextrose water 5% serum, medical grade sterile, with self healing injection port, and use serum set for injecting and aspirating, you could buy from a pharmacy

      You're talking about iv saline bags. Yes?
      I can see that being effective and easy. I would prefer to leave that much medical waste for medical purposes, though, if possible.
      There's also the cost and the volume. I'm not going to need THAT much culture for any one strain, not for a LOOOONG time, anyway. I'd also have to figure out storage. Combine that with the cost per bag (that I've found so far, anyway) and unless I have a family member with "sticky fingers" working in a hospital setting (and I was willing to do that), that could get pretty costly kinda quick.
      I'll try these reusable forms. I'll keep in mind the bags as a backup, though. I can actually see them being very organized and efficient in a larger, professional setting even. For a home or small-scale mycology enthusiast, though, I think these will tend to be the better option overall. I also will be trying to minimize how much landfill I produce per "grow" where and when I can.
      All that said, if you have sources for smaller bags or for what you see as more cost effective, please let me know? I'd appreciate it!

      No, I would just take up some sterile water in a syringe, then use the needle to biopsy the mycelium, (push the needle into the agar which should pick up enough mycelium) then just use the sterile water to push the myc into the jar.....does that make sense?@@KOKO-uu7yd

      @@KOKO-uu7yd fyi, 5%dextrose water bags, not salin ones, they come in a verity volumes, 300ml up to 1000ml, these sugar bags are relatively cheep, sterile, equipped with self healing injection port, easy to buy from a drugstore , mostly come with sterile serum set which could coupled with syringes and cross connectors for easy usage, could be stored in cabinets for long periods of time, could save time and energy compared to home made sugar jars, with much much less risk of failure/contams.
      On the other hand, you may get satisfied if you make your own liquid jar while doing mycology as a hobby at home.

      So yeah, I think you're talking about using the cockstop to inoculate? If i understand correctly, something already liquid should be fine. If I'm working with something like agar or spore prints, I'll still need to have a "carrier" of sterile solution. Other than that, I should keep itnsimple and not over think. I got thst all about right?😂

      Me personally when my LC is ready I will empty the entire jar into syringes in one session. I really only like to access the jar once.

    I bought everything in the description and did everything according to this video… Everything went great, except for the little red cap with the Luer lock melted and had aluminum foil covering as well, so what did I do wrong? 😂

      No, you didn’t do anything wrong. Those Lids seem to be a little hit or miss. I’ve had some survive and I’ve had some melt, I’m looking for a better solution now.

      Second, I just did the same and had the same issue with the red cap.
      I went back to Amazon and read the reviews, and older reviews didn't mention an issue with the caps but more recent ones did.
      I had just ordered new caps from a lab supply company and will reprocessed the jars when they are delivered.
      Not the end of the world , just annoying.

    very neat, But aren't you risking contamination more by having to take off that red cap and putting the valve on? And then having to take off the valve cap and putting the syringe on and off ? I love the tubes though. If there was a way to combine an injection port with the tubes (not seperate on the same lid, but together as a single line) I think that would be close to ideal. Have you ever tried that? I aslo dont think you really need high temp RTV as that is usually for the exhaust ares of engines. Regular silicone should be able to handle 250°F easy. Have you tried it and had failures? LOve you videos clear consice simple, very nice. Thanks.
    Also any reason to use a 3-way valve when 2-way are cheaper and just as easy to get. And why even use the valve if you cannot sterilize it? Use it once then trash it? I am just trying to understand as I am currently building up my LC cultures and plan to have about 20 jars, and will use at least a full jar a week. for inoculating grain jars

      You're correct, any changes in open air you risk contam. Just be sure to do that in an SAB or in front of a flow hood. There are stainless stopcocks out there but they are quite pricey at over 100 buck or more each, but those would be ideal as you could then just PC the entire setup......AND as it turns out those red PP5 caps melt anyway.     I made a very similar one, but I didn't use a 3-way tap. Is it really necessary? Any reason not to connect the syringe directly to the female?

      I only used the 3 way because they were readily available on Amazon. The only reason I wouldn’t connect it right to the female on the bulkhead is storage if you don’t pull all your LC into syringes in one sitting. The stopcock gives you the ability to close it up and store it without worrying about contamination

    Question for you (I'm rewatching now) - you have an injection port, but could one inoculate with lc or spore syringe via the stopcock? Perhaps a flush of sterile H²O would be needed to get everything down the tube and into the medium... 🤔🤷‍♀️
    I'm not sure of this would ve an advantage or not, or even if it would work? Your thoughts?

    Those pp5 lids are not pressure cook safe, I just built this exact setup 25 minutes in the pressure cooker deformed and melted the lid. Gotta buy another pack now😢 other than that I’m excited to try out once I get some more

    Do you use magnetic stir bars and does it get in the way?

      Yes I use stir bars, the hose usually has a small curve to it and you can move the jar slightly when stirring to avoid hitting the pickup tube. They work just fine

      So after I build them I’ll run them through my dishwasher on the sanitize setting then I will put them in a mushroom grain bag and PC them for 60 min at 15psi. I will leave them in the bag until I’m ready to use them.

      Soooo Sorry, I've had a stroke, and I'm still stroke stoopid. :-(
      Thank you. I see it, should have looked there and I thank you for shared knowledge. Stroke cause me to experience the near death experience, but also to hallucinate for about 4.5 years. I discovered mrooms in my quest to answer, 'what the hell did I see?' Fortunately, mrooms did not reproduce the quality of graphics that I saw. DMT did not produce the quality of graphics I saw. Nor did acid. Salvia Divinorum came closest, and scared the daylights out of me multiple times. However, SD nearly duplicated the quality of the graphics of my near death experience, but I saw unrealistic devices and mechanisms. There was none of that in what I identify as penducular hallusinosis. I did lay prostate while a small man put his hand inside of my head. He also pulled the nerves in my head such that my eye was outturned after the hallucination 😲.
      if you want more of these kinds of narratives but with far more detail including being kidnapped and charged with very strange crimes, you might enjoy, I mean appreciate 'The Mysterious Red Light Camera Murder' I'm looking for a publisher, and prissy literary agents are objecting to the use of curse words and low-life expressions such as 'Cops were trying to inventory my dingle berries.'
      By far, mrooms were the most prosocial of all the things I mentioned.

    now that weve covered the basic stuff, how about we get more in depth and talk about recipes for cultures, good substrates for mushrooms and how different species have different needs.
    I've heard you can just use honey and water as a liquid culture recipe.

      Honey, dextrose, Karo Corn Syrup....lots of stuff. I'll be doing a video on the recipe that I use very soon!

    Your information is amazing! Thanks so much for your information especially where to get the items on Amazon.
    My question is this: if you don't want to use up all of the liquid culture, can you put a cap on top of the stopcock, after using 70% isopropyl alcohol to sterilize it? It seemss that even if you use a new, sterile stopcock, wouldn't it still have possible contamination when you remove the previous stopcock? Thanks.
    PS) I think you should patent it! I've seen the Myco lid using a metal transferring needle instead of using your silicone tubing, and your lids are superior!

      As long as your sterile tek is good you should have no problem using the stockcock more than once. What i generally like to do is have a small jar with 70% iso in it and I'll just drop the caps in there while I'm filling, then wipe them down and put them back in when I'm done. It's been working great for me.
