Do you let them cool down to room temp with the filter on overnight?
So, because I have a flow hood I actually don’t use the sleeve and keep my plates in front of the hood until they are cool and all the condensation has gone away. I don’t usually use the boiling water trick anymore because I have a hood. When I was working in an SAB once I was done pouring my plates I would put the sleeves back on them and then remove the SAB and put the mason jars of hot water on them and leave them until they cool completely
Thank you for the video! You mention "leaving the plates until the next day". Is this in front of a running flowhood overnight or is it turned off after pouring? I wonder if contaminated air could get sucked in while the temperature drops. Do your petri dishes have ventilation nocks?
I leave my hood running, if not I would just cover them in the sleeve and keep them in there. They'll be fine.
Thanks for the vid!! Question, my LCs I’ve made which have Nutritional Yeast are instantly cloudy before even inoculating and after sterilizing. How would you tell contamination then? Recent LCs I’ve taken the NY out and no problems now however, I see random “floaties” on top, once I stir, it disappears
that's normal, once the mycelium starts to feed on the nutrients in the jar and grow you should notice that cloudiness subside, the jars here with both inoculated the same day.
@ awesome, I’ve noticed some LCs if they’re not stirred often enough they start to colonize and layer of mycelium on the surface of the LC, is this ok or is this actual contamination?

@RealSimpleMushrooms I've been using grafting tape for a while now, since it has pretty similar properties to parafilm, at least the ones we need. (I come from a fruit growing background). Your videos are very informative, devoid of speculation and completely in line with the theory, maximally simplified for a wider audience, and at the same time entertaining, which makes the information easy to understand and adopt. All in all, just as it should be. I wish you, and us, good health and new videos in the New Year 👍🏻

Thank you. You said you'd do this video, and ya got right on it. I poured 20 on New Years Day. 4 plates are stinking like yeast. (Maybe i didn't mix it well enough or long enough) Poured too hot and fogged up the SAB. Getting ready to order the products you put links to on the last video.
It happens. I'd say for every 50 plates I pour I get 5 contaminated ones

All part of a plan, I didn’t post for a very long time and I’ve been working on the SEO on the channel so I’m posting 10 videos to give the algorithm a bunch of audience data so it can recommend my videos more and to the right people. It’s been working so far. Every video does better the the prior one. Once I hit that 10 video mark I’m going to get on a schedule of either weekly or bi-weekly with maybe some live stuff sprinkled in

As someone who is new to growing and learning about mushrooms, I’ve only tipped my toes into growing from kits and just started outdoors in beds. Really excited to learn more and have greater knowledge. What are some of the best beginner resources, thank you
North Spore, Renegade Mushrooms, and FreshCap Mushrooms are good channels to learn beginner techniques. Lab work is a little advanced.
@@TheUnhousedWanderer I’ve been using the box kits from north spore and stated trying fruiting bags. It’s good to know I’m in the right direction. Yes the lab stuff is somewhat over my head at times, but the more I learn the more my deeper understanding of how it all works increases. Thanks for your advice.
There are a lot of good ones out there but I gotta say Philly Golden Teacher is awesome and all of his teachings are solid. Once you get more advanced (and he has some great beginner content too) is Edward Grand. He’s got a TON of content to sift through

Glad I found your channel :) Thanks for the tipps! Two questions regarding the pressure cooking: I assume you have unscrewed all the lids slightly, correct? Also, how much water did you put in the PC? Are the jars/bottle submerged slighty? Mush love man
If you have a gas exchange hole or are using a lab media bottle you can screw them in tight, if you just use the metal lid upside down you need to crack that one loose a bit. And yes I have them slightly submerged…maybe an inch or so. Glad you’re enjoying the content. My follow up to this video is scheduled to go live at 3pm est today

What makes a "true casing layer" if what you did on top isnt??
A “true” casing layer is a layer of 50/50 peat moss/vermiculite. Peat can hold 20x its weight in water and the mixture is non nutritious. It helps with humidity and moisture retention

Your skepticism of potatoes aggravated me throughout your video. This has been the gold standard for mycology since it's inception. Anyone that thinks this is weird has never read a book about it.

Thanks! So transfer to a water agar.... no nutrients for bacteria to feed on. But, the mycelium can still grow there and get stronger. Between your last video and this one, this is what I am hearing. I had not heard of water agar til you showed us how to make it. I think you are my fav creator right now. Helping me to understand agar work. Btway..... On water agar, it will often stretch out quickly in search of nutrients. Pipetting by mouth is strictly prohibited, but subculturing via butthole is just asking for it.
The water agar is a good idea. Btw, if you have an Asian market near you, agar is pretty cheap. You can also get packets for single use... if you're doing a good amount of agar work lol. They're like 25g or so

Is the trench just a cut in the agar or does there need to be a small gap? I find it hard to do anything other than a clean cut.
You need to have the blade hot and turn it sideways to carve a gap with the broad side of the blade. It needs to be a clear gap all the way to the bare Petri dish.

You can really use either. The meat will speed up the growth needed to achieve the hopefully clean transfer. But personally I prefer to make that transfer to water agar..then transfer one more time after that back to MEA
@@RealSimpleMushrooms thanks for the response! i put it back to water agar this morning , but i find the growth incredibly slow and very fine strands ... I will try both these methods on water next .... i was on T6 and it was making me crazy with the aseptic technique .....i was considering antibiotic agar , but i would rather avoid if possible
You only need one of those fine hairs ;)
When I put something to water agar usually it will go rhizo (but be very faint) and as soon as it shoots off from the transfer I’ll grab a piece and toss it back onto MEA

@@RealSimpleMushrooms I tried a no pour version on glass plates. Your recipe for lme. The sterilization bags sorta broke down. So I iso'ed 70 the area as I removed the plates and wrapped them to seal them and ziplocked them. Maybe tent the bags in foil next time. In a week I'll see if they are still clean and test some lc samples I have saved. The stir plate and Corning Pyrex are useful for coffee too lol. Already had a nice pressure canner that made it easy. Lc to agar testing and spore to agar are my next in line.

I've been growing mushrooms and doing plate work for the last three years. I make up my agar for plates in cold water with a good swirl and then straight into the pressure cooker. Everything dissolves fine by the time it comes out. (I had to close my eyes when you poured the boiling water into the mason jars. If there is any flaw in the glass jar, it can shatter from the thermal shock.) Good video, looking forward to the next one. Keep them coming.

Hi, nice video. I have a question about cloning and obtaining spores. Are spores obtained to renew the culture? Cloning a mushroom preserves the age of the mycelium? What are you looking for with cloning and spores? Stabilize genetics or continue looking for phenotypes
Short answer, cloning you're looking for specific traits (large fruits, large clusters) to hopefully continue to grow that same phenotype (its takes a few selections and then going back to spore to stabilize it, that's a much deeper conversation) Spores are how you pheno hunt. I will swab a plate with spores and as long as they are clean I will just take that whole plate right to grain and grow it out and start hunting. I will also try to isolate a monokayron culture for potenial breeding down the road (again another deep topic)

@@RealSimpleMushrooms I have another question if you don't mind, I was growing certain genetics with peat, worm castings and vermicurite. (with great resoults, yields and flushes) Some easy genetics.(like creepers or golden t) Now I wanted to try some exotic ones like yeti and I have noticed that the fruits come out too small. Could it be that some species like different types of substrate? I have had bad experiences with coconut and vermicurite so I decided to make my own substrate.
