  • Question: What cofactors are needed to synthesize and recycle BH4?
    Short Answer: Zinc, magnesium, potassium, and niacin are the cofactors needed for the synthesis and recycling of BH4. Folate and methylation are not involved, though high-dose folate or folic acid could hypothetically hurt BH4 recycling since both are recycled by dihydrofolate reductase (DHFR).

    So essentially if i want to increase BH4 and lower excitotoxicity of my Arginine pathway, I should increase dietary Zinc, Magnesium, Potassium, and Niacin?

    So going into this a bit more ...if someone has ENOS3 mutations...not me..but someone.... what is the best way to go about supporting the ENOS pathway?

    I have A1298c homozygous gene which causes low BH4, and I use Liposomal Vitamin C to increase it.

      Can you share the vitamin c supplement you use?

      @@jfrm334 Nutrivein is the cheapest Liposomal Vitamin C on amazon and I find that it works fine. I used to buy the very expensive stuff but then I realized how incredibly strong the cheap ones are.

      @@jfrm334 I recommend if you take a lot of Liposomal Vitamin C to also use copper glycinate, it will help because most people have low copper anyway. I use "Solgar 2.5mg copper" supplement. Technically it's possible to make copper supplements for free by using copper sulfate.

    Is it accurate to say that Folic Acid causes competitive inhibition of DHFR?

      The key folic acid question 😅 everyone mentions this yet no one provides any proof. Hoping for a definitive answer from Chris.

      @@tonipieleanu I don't think we can get any definitive answer from Chris. He is saying absolutely there is no correlation between directly folate and bh4, while in cerebral folate deficiency bh4 is depleted and folinic acid increase the amount of bh4.

      It is plausible that you would have extra competition and it probably depends on the tissue. Notably you have competition from dihydrofolate anyway.
