    Nice video. Um my question is how do we get to immobilize the antigens since they are our analyte provided that we are not sure if they are present or not in the patient's sample?

      Yeah! How do we immobilised the Ag?
      I hope we are not using Ab to immobilise it or else what will differentiate the Direct And Sandwich ELISA.

    By Elisa there are many positive igg as ebv hhv6 cmv But with negative igm. And negative pcr? What do this mean with clinical symptoms?

      On the Elisa test for ebv, cmv, hv6 the igg are positive and the igm are normal. Also the pcr for these viruses are normal. And the person is have symptoms. What to make out of this situation.

      ELISA is used both as a quantitative and qualitative assay. But Direct ELISA is not the preferred method. Indirect and sandwich ELISA are preferred.

    A) know Antibody
    B) know Antigen
    C) Patient Antibody
    d) substrate
    Please answer with reference
    One of Rajasthan health university of science is claiming it's answer B while as per your video it's answer is A

      I find this question incomplete. This is because ELISA test can be used to detect both antigens and antibodies.
      In case, we want to detect the presence of specific antigens in the patient's serum, we should have known antibodies for this antigen (DIRECT and SANDWICH ELISA).
      And in case, we want to detect the presence of specific antibodies in the patient's serum, we should have known antigen (INDIRECT ELISA).
      Now, because most common application of ELISA test which we study is the HIV screening test. So, according to that, answer should be B. Because in HIV screening test, we detect antibodies (INDIRECT ELISA). Known antigens are immobilized in the wells.

    What is substrate in this?

      It is the molecule or compound specific to the enzyme used to conjugate the antibody. It (complex of enz and substrate) is chosen in such a way that it gives colour after substrate is added to the mixture/ well containing the enzyme conjugated-Ab. Some examples are HRP enzyme with TMB substrate to give green colour.
