Enzyme linked antibody can not be called secondary antibody. Rather it's a second antibody (it is also a primary Ab) that can recognise an epitope on the antigen that is different from the epitope recognised by the primary ( 1st Ab).
Why the number of cells is equal to the number of spots? If many cells produce lots of antigen, it will form complexes very near one of another, therefore forming one large spot, so one spot isn't equal to one cell!
