    Beer's law -- applied to proteins; that's neat. But it looks like the assessment to determine the quantity of protein in a sample is strongly dependent on historical data; where you can compare similar samples to each other. ... ... or just read the data sheet provided by the supplier of the ELISA. After doing some reading on this... it looks like most ELISA samples are actually created by specialized laboratories that hire a lot of Peptide Chemists. Surprisingly... Peptide Chemists are paid around $45,000-75,000 per year (well; for ONE datapoint, but that ONE datapoint was requiring that the applicant had a PhD). I am a bit surprised at how saturated the Chemistry job market really is at the moment; that seems like low pay for pretty highly skilled work.

    So whats the diffrence between western blot and elisa indirect variation?

      I assume you have noticed that both techniques use an antibody against the antigen and another secondary antibody which is detectable!
      The difference is that western blot (WB) is able to reveal the protein size, whereas ELISA is not. Also, ELISA allows for a more accurate quantification that WB.
