  • Biomedical lesson on AKT (Protein Kinase B) signaling, regulation and downstream effects. The AKT signalling pathway is a highly conserved pathway that induces several important functions including glucose uptake and utilization, protein synthesis, fatty acid synthesis, glycogenesis and cell survival and proliferation.
    Hey everyone. This lesson introduces the AKT (PKB) signalling pathway, how the pathway is regulated and controlled, and what are the major cellular outcomes of AKT signaling. Additionally, we also discuss how diseases such as cancer can be associated with abnormal AKT signaling.

    I'm glad you saved the simple explanation for the end because it helped me digest such a detailed explanation of all of the macronutrients it regulates by blocking them... I think 😅

    1. AKT is an inhibitor of TSC1/2
    2. TSC1/2 is an inhibitor if Rheb.
    3. Rheb is an activator of mTORC1
    AKT inhibits the inhibitor of Rheb (TSC1/2). Thereby allowing more Rheb to activate mTORC1 allowing for more activation of TSC1/2 as compared to if AKT was NOT present

    Is GSK3 the same as GSK3-ß from Wnt-signaling (Activation of Akt leads to inactivation of beta-Catenin Destruction Complex, leading to a proliferative signal through a accumulation of beta-Catenin in the nucleus)? This would be an additional way how Akt stimulates cell proliferation?

    Great Job! Really helpful! But in the part relation to Glycogen synthesis is little bit confusing to me. If possible please clear me! Does by GSK3 phosphorylation, GSK3 become inactive? and GS become phosphorylated or dephosphorylated?
