in sickle cell test Dithionite can also be used but sickle cell test can't differentiate between homozygous & heterozygous state so Hb electrophoresis is used..... but for any Hemoglobinopathy High Performance Liquid Chromatography (HPLC) is best.
for osteomielitis, S.Aureus is the most common organsim, but in sickle cell anemia, Salmonella becomes number one due to the inability of the spleen to do its job due to the functional asplenia. So your more prone to encapsulated organisms like salmonella.. But im not sure, i could be wrong.
Thank you but why you divided case of Vein and artery ?. this confused me . you mentioned that sickling occurs only in ( acidosis /hypoxia etc) but the vein and artry case this mean patient is suffering of sickling all the time due to deoxy Hb in vein . im confused can you explain?

Both. But it depends on the issue, and depends on whether or not splenectomy is going to be performed, and many other things. Only a doctor can help you with that, because I am not a doctor.
When I was born in the mid 80s no one tested for it. I wasn't diagnosed until i caught mono when i was about 5 or 6. In my state they test everyone at birth.
