This is a hard one, since there's no the best version here. Each has it's own strengths and weaknesses.
    - Sega Megadrive/Genesis uses high-resolution 320x240 and is very smooth and responsive. However it lacks in audio, FM Synth isn't great for Lion King Soundtrack. Also Genesis limited color palette and only 64-color on screen make this game a dithered mess.
    - Amiga port requires Amiga 1200 or similar, it won't run on Amiga 500 or any ECS/OCS Amigas. However it's rather bad. It lacks few level, starting with second one "Can't Wait to be king". Colors are better than on Sega, since Amiga 1200 had full 24-bit palette (more than SNES or PC VGA), but parallax scrolling mode halves bitplanes so game runs in 16 + 16 color mode. Better colors than on Genesis, but less colors on screen. It has the highest quality music though with 44 kHz playback, but limited to 4 PCM channels only.
    - SNES port uses most colorful tiles and has 6+ channel music. Yet it uses the lowest resolution which makes this game much less enjoyable. Music also has low quality feel to it.
    - MS-DOS port combines high resolution from Amiga and Genesis, with SNES graphics and music from Amiga. It would be the best port, especially that it uses 320x200 16:10 resolution that gives this game wide-screen aspect for modern displays (sprites aren't scaled for non-square DOS pixels). However there are game breaking bugs. Gameplay is very sensitive to CPU speed. You need Pentium+ machine to get smooth 60 fps animation, but on such machines game break. Roar status bar get stuck and you can't finish level 2 and geyser interval in Level 3 gets too short which makes that one climb impossible on fast PCs. Bonus stages with Timon are also unfinished in MS-DOS.
Overall Sega Megadrive/Genesis one has best gameplay and MS-DOS port is best overall, although with issues I've mentioned above.
Anyway all 16/32-bit versions are playable. Only SNES port is a bit annoying with reduced view size due to lower resolution.

I like how both Genesis and Amiga have a slightly faster and more upbeat stage music here.

I like the graphics on the Amiga, but based on this video, the Hyena battle seems way too easy. I'm impressed by the Gameboy version even if the Hyena was the same size as Simba. My favourite and yes I am biased here is the DOS version because I actually owned it and played it. Also not shown here is I just can't wait to be king, which I believe and correct me if I'm wrong the Amiga version doesn't have.
