    Hello, thanks for this video ! Just to sum up :
    -You started from a shoot. You took away the roots, cleaned it, put in a media, and get the same shoot. Now you transfer the shoot to another media, but at this point(after 125 days), you still have only one shoot !
    My question is : when is the multiplication part starting ? How will it happen ? looking super forward to knowing !

      Yes, I could have moved the plant after day 50. However, I was busy building the new lab. I moved the plant to multiplication media 17 days ago, and is already doing some interesting things. I'll make an update video soon!

      @@PlantCellTechnology have you made a video of where the actual multiplication of the plant yet? so far it seems to only have been 1 plant in various medias, when/how does the multiplication (division?) happen? thanks

      @@c3235086 No, we don't have a multiplication video currently. Multiplication is the second stage of tissue culture, where small clumps are obtained after a couple of weeks of culturing tissue in the media without hormones.
      The small tissue clump is transferred to a fresh growth hormone-containing media. This leads to the growth of the tissue clump or callus, and after a few days, the formation of small shoots is observed.

    Felicito tu video, yo estoy trabajando con monstera ahora pero en el establecimiento tengo muchos problemas con la oxidacion. Anteriormente no ocurrio este tipo de oxidacion que en 2 semanas esta totalmente negro, he usado todo: Carbon activado, Antioxidantes: Ac ascorb, Ac citrico, y L cysteina, pero es igual. Estamos en pleno verano muy caliente en invernadero , es posible que esto afecte el proceso de establecimiento? que epoca es la mejor?

    I love your videos! I'm just getting started in TC out of sheer curiosity and reading about orchids and carnivorous plants. With a basic understanding of chemistry (lol failed orgo twice in uni) and how to use the tools, I was able to establish a begonia! I recently ordered PPM at the recommendation of a couple forums. It's so cool that I can also receive my it outside the USA, this honestly saved me so much money and headache. If I still lived in DC, I'd love to be your intern lol I only wish I was exposed to this in school.

    Francisco!! Mil gracias por tanta información! Me parece fascinante todo lo que nos cuentas por aquí!
    Estoy muy interesado en esto precisamente para propagar monstera, pero aún no entiendo el proceso completo. Si puedes resolverme algunas dudas te estaría muy agradecido.
    En primer lugar, dices que es necesario establecer primero la planta en el caso de la Monstera Deliciosa, para ello solo sería necesario MS, no? Pero si además queremos que comience a enraizar deberíamos incluir NAA
    Después de que la planta tenga raíces, has quitado estas raíces y las has puesto en un medio de cultivo nuevo, pero este nuevo medio es de multiplicación? Es decir, multiplicas la Mosntera desde las raíces?
    Sería un honor recibir tu respuesta!

    Muy bien video! Cuando pasas el explante a medio de multiplicación, no lo divides por segmentos nodales? Como se multiplica? Gracias

    after making this multiplication media with the PGR, did you put it in the autoclave or did you add the PGR after sterilizing the media

      I added the PGRs while I was making the media. I used heat stable PGR (BA and NAA).

    Can you use purified agar instead of gallan gum for the multiplication media?

      Yes, you can use agar too for the multiplication of plants in tissue culture.

    Awesome videos! So you cannot put the shoot straight into multiplication and skip the first media?

      Yes, for multiplication you can divide the shoots into subsections if you want and transfer them to multiplication media.
      Multiplication media is different from establishing media, as the former one contains plant hormones and the latter one does not.

    My philodendron is in multiplication media now and its been 4 weeks and no roots. Should I be concerned?

      The multiplication media contain a higher concentration of cytokinin that induce shoot development. so, for root development, you need to transfer your developed shoots to the rooting media, which contains a higher concentration of auxins.
      So, I hope you understand why you aren't getting any roots in your plants.

    What is your media for stage 1 ?

      Regular MS media no hormones. You may skip the establishment stage a move directly to multiplication media. However, for some plants including Monstera deliciosa is better to get it established first.

    You are using "media" as it was the singular. It is not! Medium is the singular. Media is plural. So, make one medium or many media.
