The explant contains multiple types of differentiated and undifferentiated cells, does this matter? Since most plant cells retain totipotency, couldn't you use any part of the plant? Is it faster to use differentiated vs. undifferentiated cells?
Do you use 1% Clorox for sterilization? For example, in 100mL of distilled water add 1mL of Clorox? What is the concentration of sodium hypochlorite in Clorox? 5.25%?
To culture the same plant you can use the same process. But for different plants, you might need to find a procedure suitable for that, as different plants have different requirements. About the starter kit, you can use it to culture any plant.

I have a mature Birkin that I kinda abandoned when it completely stopped growing. It's still green but looks pretty sad. Would cutting it all back down to the piece you did in the video be a viable menas of rescuing and propagating this plant?
Hey David, You can cut all the leaves that had already yellowed and are dying (these might tend toward the bottom of the plant as it grows the new leaves from the top start white part and they get green over time and then yellow when it's dying). After cutting, transfer the plant to leca balls so they are consistently in water. You might observe the growth of new leaves after this!
In our lab we use the following media formulation: For multiplication: Full MS media 25 g of sucrose 3-5 ml/l of BAP 0.3-0.5 ml/l of NAA 1 ml/l of PPM 6 g of Agar or 3 g of Gellan gum For rooting: Full MS media 25 g of sucrose 3-5 ml/l of NAA 0.3-0.5 ml/l of BAP 1 ml/l of PPM 6 g of Agar or 3 g of Gellan gum
will this work with a Philodendron Caramel Marble? Second question: did you make out of one plant, 3 plants (because the initial plant splits in 3 leaves)?
You can try the recipe. However, media components and concentration differs based on the plant species. And, the division is based on the shoots. so, if you have more shoots in numbers or longer shoots, you can divide them accordingly.

i have an expensive tissue culture coming in the mail(hopefully), curious your thoughts on how much light to give them. i have a sealed terrarium cabinet with grow lights but have read the small tissue cultures don’t respond well to those lights immediately. Hoping do not botch a pretty expensive experiment, so your thoughts are appreciated
Hi Brennan, The duration and intensity of light given to the plants depend on the plant species. So, based on which plant are you trying to introduce in tissue culture, follow a particular procedure of tissue culture of the plant to get better results with lesser efforts.

Sir, i have watch and understand your sterilization protocol. My question is .. Is it ok for after treatment of hydrogen peroxide + vinegar we could wash explant several time with sterile distilled water to remove traces of sterilizing agent and dried for few seconds before inoculation.?

@@amtd3808 You should follow only a specific protocol serial-wise. Don't try n mix two protocols. If you are following the complete procedure from explant collection to acclimation from the research paper, follow it critically. Don't mix the steps of the two or more protocols. They are specifically designed with some specific combination of chemicals.

Washing several times is not required. However, 2-3 gentle washes with distilled water will work to remove the traces of chemicals.
