  • Thia video describes how to fit a receptor mechanism to single ion channel reordings.
Ciick 'SHOW MORE' to see refernces and more details.
The fits are done by the methid of maximum likelihood, i'e.the transition rates (rate constants) in a specified mechanism are adjusted to maximise their likelihood (the probability of seeing the observations given the mechanism)/In order to calculate the likelihood we need the distributions of the apparent open and shut times, allowing for the fact that both open times will appear to be longer than their true value when we miss short shut times (and similarly for shut times). These are known as HJC distributions (for Hawkes, Jalali & Colquhoun, who solved the missedbrief event problem).
