  • In this video, Biology Professor (Twitter: @DrWhitneyHolden) will cover the Disk Diffusion test, also known as the Kirby-Bauer test, and discusses the background information necessary to understand how this test works. Specifically, this diagnostic test uses a highly standardized procedure to assess the level of effectiveness of different antibiotics against a particular bacterial isolate, by measuring the size of zones of inhibition.

    Hello! Is it possible to adapt this method to see the effect of visible light (blue, red, ..) on bacterial growth/inhibition? If not, which other method can be utilised? Thankyou!

      I would put the bacteria on different plates and expose each plate to one wavelength of light. You could then count the number of colonies growing after exposure to different light. Or you could grow bacteria in tubes of broth exposed to different wavelengths of light and then measure growth in the tubes using a spectrophotometer.

    Regarding the procedure for the antibiotic susceptibility test, would it still work if a spread plate technique was done opposed to using a swap to cover the HE plate?

    Can I ask a question :
    As we know the Anthrax toxins consist from 3 main proteins, One of them is Edema Factor ( EF) which is adenylate cyclase class2 , the genes for this class are known as cyaA, one of which is anthrax toxin.
    This cyaA is Bifunctional hemolysin/ adenylate cyclase, after cleaved in two proteins a calmodulin-sensitive adenylate cyclase (cyaA-ACD) and hemolysin , and this hemolysin is responsible for hemolysis of RBCs ,
    So finally How B.anthrax does not produce hemolysis on blood agar in laboratory diagnosis?

    Can nutrient agar be used in this test?

      I’ve used LB and nutrient agar with my students. We call it a “modified disc diffusion assay” since it is not the standard protocol.

    Hi, is there a std method of testing effectiveness of a biostatic agent applied to a surface? I’ve synthesized several colloidal copper hydrosols (suspended copper nanoparticles) to see if spraying on PPE will increase its antimicrobial properties. I was thinking of spraying discs and placing them in a lawn culture, but the nanoparticles are not intended to diffuse through the agar but rather stay attached to the material (discs). Is there a simple “zone of inhibition” type test, or can I just spray the hydrosol directly on part of the agar and look for growth? Heeeeeeelp!

      You could use paper discs dipped in your hydrosols to “touch” inoculated agar, then remove, leaving a layer of hydrosol behind. Do it at various concentrations and see if bacteria are inhibited. Can you just culture bacteria in 96-well plates with varying concentrations for hydrosols?

I’d like to verify whether copper treated fabrics inhibit microbial growth and for how long. Unfortunately I only understand the chemistry part. Any direction would be greatly appreciated.

    I am coming across the reporting of 0 mm diameter for the first time. I know of reporting
    6 mm, which is the diameter of the disc.

      I know it’s counterintuitive, but recording 0 mM for no zone of inhibition is pretty standard. It’s the only thing I’ve ever seen.
