  • In this video, Biology Professor (Twitter: @DrWhitneyHolden) will cover the Coagulase test procedure and discuss the background information to understand how this test works. Specifically, this diagnostic test indicates the presence of the enzyme coagulase in a bacterial culture. This is an enzyme that acts as a virulence factor, helping to disguise the bacterium as a blood clot so it might hide from phagocytes. This test is used to differentiate coagulase-positive bacteria, like S. aureus, from coagulase-negative staphylococci. The two versions of this test, the slide test and tube test, are described.

    Why do we have two separate tests for the two forms of the enzyme?Why can’t we detect bound coagulase in the tube test OR free form in the slide test?

    Why coagulase converts fibrnogin to fibrin

      It results in blood clotting. The resulting clot protects the bacterium from phagocytosis (we think).

      The proteins that clot in the presence of coagulase are more concentrated in the plasma.

I did not see a video of yours for this test (which I prefer), but if you have a source you recommend for other microbio tests, I would love to hear it!

    Thank you for this wonderful knowledge, Professor. May I have a question, please? Why do we check for the coagulase tube test after 90 minutes, instead of leaving it overnight? Is there any change if that was a positive test and we leave it too long?

      You might get a false positive if you leave it too long.

      No, there is no melting. I the clump could be disrupted by vigorous shaking, but I’ve never tried.

      From Wikipedia: List of coagulase-positive staphylococci: Staphylococcus aureus subsp. anaerobius, S. aureus subsp. aureus, S. delphini, S. hyicus, S. intermedius, S. lutrae, and Staphylococcus schleiferi subsp. coagulans.
