Interpretation chart u will get it from the antibiotic company site / u can enquire with the suppliers.

Sir I prepare sample 10mg/ml concentration. For standard I use ofloxacin so I make the standard in same conc as test samples are?
No need to prepare standard in same concentration.
In stokes method same antibiotics disc we've to use and same bacteria for test and control?
Same bacteria for test & the control.. e.g: for the test u can use identified Hospital sample and for the control u need to use standard strain (ATCC strains).
Is incubation not required if bacterial suspension is prepared using MH broth, as with glucose broth or peptone water where the suspension is incubated before making a lawn
Incubation is required after inoculation

Why the bacterial suspension is compared to 0.5 Mc farland standard
It helps to ensure that the number of bacteria will be within a given range to standardize microbial testing.

Yes. U can take the average and then compare with interpretation chart. But the difference of the trip. results should be near by.
if ur doing sensitivity test with ur own prepared antibiotic then u need to do triplicate otherwise no need.
