Can you please brief few things: 1) Why do we incubate every time at 24 degree centigrade and for 30 minutes? 2) Once you said that GP120 is masked and in rare cases it happens that it gets exposed while sending its RNA, and rarely produces antibody, then how come ELISA considered as confirmatory test for AIDS? 3) Which complex produces colour to be detected through spectrophotometer and at which wavelength?
This is simple/direct ELISA. 1. It is not necessary to incubate 24 degree C for 30 min. One has to standardise the protocol. 2. Anti-HIV antibodies are produced but not sufficient to control the infection. 3. One need to add substrate after adding enzyme. The substrate is colourless which converted into colour product by the enzyme. The intensity of the colour will indicate the titer of Ab.

Hi Dr neela, I have a qn. Say for example a person is hiv positive, can we test the patient's antigen with a ready made antibody or is the reverse better as u have mentioned in your video? Because if the virus has an ability to mask itself, will antibodies even be produced. This I'm not very clear

if antibodies are taken out of serum why can't these antibodies can be sold for others as ready made antibodies. so that the person can get cure from HIV people generally say there is no medicine for HIV .
Actually these retroviruses are very nasty things.... They have spikes on there outer layer, this makes sure that they are masked by the host organisms' membrane... Hence the host isn't able to recognize them...

@@shanthipriya3117 see it's not always masked or capped as also mentioned by mam. The viruses are masked most of the time as a result body fails or is delayed producing the antigens. Got it? And I'm not mam I'm a student like u

many explanations are wrong. alkaline phosphatase can not bind directly to Ag-Ab complex. enzyme linked Ab are synthesized artificially in the laboratory.

Santosh Kumar may times it gives false results..
as far as I know, each tip of the “Y” (antibody) in the variable region is highly specific and
binds to only one particular antigen, suppose GP120.

Medam how antibodies are produced in serum for hiv antigens, when Gp120 is masked?
That's why body doesn't recognise HIV and it gets worst. It doesn't produce any antibodies.
