  • This video will give a comprehensive view about lac operon mutations. Thw following questions would be answered.
How do mutations affect the lac operon?
What happens if lac Z is mutated?
What are the 4 components of the lac operon?
What does the lac operon detect?

    Question:
    1) In LacOc and LacI- conditions, what if glucose is still present? It is stated earlier that lac operon is only expressed if glucose is absent and lactose present because CAP increases RNA Pol efficiency. LacOc and LacI- only mutates the operator or repressor, so whether lactose is present or absent does not affect transcription, but what about glucose levels? Wouldn't this still control transcription?
    2) Cis vs Trans/Complement
So if a mutant is not rescued, a mutation acts in cis and if a mutant is rescued, the mutation acts in trans?

I thought lactose doesn't necessarily bind to the repressor, it's the allolactose effector that binds to the repressor, allowing activation. ?
