    Hi there, love your videos and I have gone through all the Q&A and have not found a clear answer to the sugar question if you can please clarify. 1% VOLUME of sugar I believe is confusing for everyone. If we make a h2o2 solution at 1:19, let’s say 2cups/500ml, sugar isn’t really something anyone measures in “volume” so when you state 1% would it be safe to say add 1 tsp sugar (5ml)?? Please let us know, this process is something I will test on my old seeds! thrilled to have found your channel!

    Thanks for the video! I have germinated seeds on peroxide before but I forgot the ratio, so started to search and found your video. I didn't know about adding sugar, that's an extra I will need because my seeds are old! Also you may check coppersulfate as an addition to your "seed reviving mix" as I remember having seen someone also using that in combination with peroxide to germinate very old seeds. Not sure if that is useful, but you may wanna check that out!

    Two questions about some things I didn't understand well:
    First: how much would be 1% sugar, considering the solution is measured by volume and the sugar by weight? Let's say I have 500 ml of water, would that be like about a teaspoon? Or how can I calculate it?
    Second: won't soaking the seeds in the solution for several days waiting for them to sprout damage or kill the seeds? I've read in some other places that you're supposed to soak them for only a couple of hours, especially with smaller seeds.

    • @PlantCellTechnology
      @PlantCellTechnology  5 месяцев назад +2

      1% solution means you have 1 gram of sugar in 100 milliliters (mL) of solution.
      As soon as you see the seeds sprouting in a couple days, you need to transfer them to fresh tissue culture media.

    • @PlantCellTechnology
      @PlantCellTechnology  10 месяцев назад +2

      That's good. But at least for sterilizing seeds to place in tissue culture, I found that 19:1 works for almost all the seeds. I have tested.

    • @Санчес-я1т
      @Санчес-я1т 9 месяцев назад

      Perhaps they recommend this concentration simply for sterilization, soaking for two minutes. Because if you leave the seeds for several days in a solution with such a high concentration of hydrogen peroxide, it can have a negative effect. Subsequently they will grow very slowly.

  • @alyssa0411
    @alyssa0411 9 месяцев назад

    One question because I’m having trouble finding an answer online. Is the H2O2 method used ONLY for old seeds? Or can I use it on new seeds as an added way to get better germination results? I have used the baggy method with fairly good results, but I was wondering if I did a seed soak in a test tube and added the correct H2O2 to water ratio, would that help with a better germination rate. My issue is not knowing if H2O2 might in some way hurt or damage new seeds.

    • @PlantCellTechnology
      @PlantCellTechnology  9 месяцев назад

      Yes, absolutely, you can use the technique for new seeds as well. Hydrogen peroxide is also an anti-fungal, so it will prevent the growth of mold in the seeds
