    About a year ago I took nicotinamide riboside for a short period of time and absolutely hated it. It was tanking my workouts. Whether I was running or doing resistance workouts, muscle fatigue was coming on quicker, performance was going down and I felt like I was dying during the workout. Thank you Chris for explaining what was going on. Metabolizing the nicotinamide was causing me a methylation deficit and a creatine deficiency.

    Thank you for this video and all your videos. I had an adverse effect with Tru Niagen. Bad muscle cramps, insomnia, anxiety, and I was going to the bathroom a lot and was very fatigued. I’m MTHFR positive (from one parent). Once I stopped taking it, all my symptoms went away!!!

      @Manny Santiago According to that paper, the bovine milk has an average concentration of 1.7 uM of nicotinamide riboside (NR). The molecular weight of NR is 255.2 g/mole. That means, you get only 0.4 mg of NR when you drink 1 L of bovine milk. That is such an insignificant amount compared to what you get from supplements (between 150 and 600 mg). Remember, it's all about concentration that does the work in every aspect of life. Do math before you speak nonsense.

    As people pointed out already, you get methyl groups from virtually everything you eat. The fact that there is methyl group deposition on nicotinamide is not equivalent to a safety concern. There have been no adverse events reported in any clinical testing of NR. We have tested 8 weeks of NR at 3 doses and looked for depressed 1-carbon metabolism using exactly the assay that was used for nicotinic acid and nicotinamide. Whereas single doses of nicotinic acid and nicotinamide increase plasma homocysteine at 300 mg, we saw NO CHANGE after 8 weeks at 3 different doses of NR. Moreover, the idea that you would get "sticky brain" from any form of niacin is inconsistent with DECADES of use of gram levels of nicotinic acid. Abram Hoffer is literally in the Orthomolecular Medicine Hall of Fame for advocating high dose niacin for psychiatric conditions. It's not clear that it works for all-comers but it does not produce sticky brain and it's not unsafe. There's a danger to people saying there's a danger when you don't have the data....

      I got very sick from taking just one pill (it came to me at 2 pm and I can't wait so I take one intending to take another on evening). I had muscle pain, nerve pain, dizziness and generally feeling very debilitated mind and body. It was pure poison for me. The maker of this drug should be a little bit more responsible by perhaps including an antidote just in case some people like me will have this problem with the drug.then I would not have to suffer for two days. But I guess I have to thank God for not being on a Boeing 737 max that dived to the ground.

      Dr Brenner I read a recent study that smokers that take b6 or b12 it doubles there chances of lung cancer. I'm an off and on smoker and take NR now I know NR is B3, should there be concern b3 may do the same thing?

      Dr Brenner - I'm about to start taking Tru Niagen and am wondering about whether I should supplement it with TMG.
      This video says that I should, but my inyuition says that taking TMG would positively BLOCK the beneficial effect of NR.
      The reason behind taking NR is to demethylate DNA. But if NR can easily bind to methyl groups from the TMG instead of forcing them out of DNA, would it even be effective? What's your recommendation?

      * Fenbendazole 200-500mg daily
      * 10-30 Apricot seeds and/or Vitamin B17, 500mg twice a day. Please remember according to lab tests, approximately 20mg of B17 is found in each apricot seed and should be taken with 30mg of Zinc citrate for improved b17 absorption. Word of Caution: Try just 2 or 3 seeds and see how you feel. Depending on the potency of the seeds Hunza Canadian brand I sometimes can't handle more than 7 daily or I start to develop hypoxia symptoms. Otherwise with some other brands I can eat 15 or more seeds daily,easily.
      * Vitamin C as much as you can handle before diarrhea sets in. - Perhaps liposomal Vitamin C you can make yourself has better absorption than tablet form.
      Might also help:
      * Metformin 250mg up to 2.5g daily or (over the counter) Berberine 500mg to? from what read, Metformin works better.
      * b15 for improved blood oxygen
      * ginseng (to help increase blood pressure in case yours drops to low from apricot/b17
      * 60mg melatonin 3-4x daily
      * 500-800mg Vitamin E daily(gamma)
      * 300-600mg Curcurin daily
      * 50/50 CBD THC oil few drops daily under tongue.
      Reverstrol mg?, Reishi Mushrooms, Essiac Tea

    Is TMG preferable to straight glycine? I realize there are methyl groups attached to the TMG, and if you're losing methyl groups by taking NR you want to replace them, but isn't the purpose of taking it to get the glycine rather than the methyl groups? You can get more methyl groups by eating more protein. It's the glycine that's important here, right?

    Thanks Doc. Because of your last vid on the dangers of niacin and my own experience I was led to discover TMG which I respond really well too. I think I had what you described - an initial lowering of histamine but then an increase in homocysteine. Now I try and get by through a balance of the two. Maybe I'm MTHFR? I plan on taking a test soon...

    My opinion on supplements is that we should only take what our bodies specifically need. I had an adenine deficiency and did research and found NAD+ helps. They also found that most addicts are deficient in adenine and NAD+ helps that too. I really feel it helped with my addiction and energy and my adenine levels are back in range so I’m going to continue taking it.

    I take niacin and NR as well as TMG. Will taking TMG at the same time with niacin or NR change their effectiveness or bio-availability?

    So after researching these types of videos I went to Amazon, and I looked up some NR suppliments, and came across Thorne ReservaCel. It seems to have 150 mg of nicotinamide riboside chloride 75mg of resveratrol and 25 mg of trimethyglycine per pill. The instructions recommend to take 2 pills one or more times per day, but based on these vids that seems like overkill. I prefer to take one pill 2 times a day. Are these good ratios? Is it too much, or too little? I'm feeling good by the way. No side effects yet one month in.

      Jesus Alvarez I’m using the Resveracel myself, and had much more energy and mental clarity the first 10 days. I don’t have that anymore and I take 1 capsule in the morning. I’d say I’m almost a month in.

    Where can you find TMG in appropriate dosage to your recommendation - capsules of 750 mg were the smallest dose I found and that wouldn't be easily dosed to meet your recommended 100 mg per 200 mg NR assuming daily divided dose of no more than 450 mg. Thanks

    I have been taking niagen for four years. Currently taking 2 tablets of Elysium which is 250mg of NR-E (Crystaline Nicotinamide Riboside) and 50mg of Pterostbene(similar to Reversatol). I do it on an empty stomach. Is that bad? It seems to work. Also How much TMG and Creatine should I be taking?

    Hi Chris. I am wondering does the mitochondria need FADH2 (flavin adenine dinucleotide) together with NADH (Nicotinamide adenine dinucleotide) do make ATP (in the electron transport chain)? Or does the NADH donate electrons separetely to make ATP from the FADH2?

    What do you recommend for people only consuming one meal a day? Right now, I'm taking 150 mg in the morning on an empty stomach and 150 mg with my meal.

      NR seems to last in my system the best part of 24 hours, certainly 12 hours, therefore you might be okay just taking it with your main meal. If you want you could take a booster dose at the other end of the day.

    Oh my. I’ve been taking NR Ttru Niagen and have been lower in energy. On the other hand, when I take Resveratrol I definitely have increased endurance.

    Hi Chris, thanks very much for this very informative video. I personally take NR at a dose of 300mg daily; been doing that for 2.5 years now. I just read Wheat bran, Spinach and beets are very rich in TMG. Eating them every day is probably going to add an extra gram of TMG to the body. Do you believer adding these three foods to the daily diet is enough to compensate methyl consumption through NR excretion?

    I take my NR immediately after breakfast every other day with a dose of only 150 mg. I assume that the low dose of NR and taking it after breakfast will minimize those metabolic pathways which might divert the NR from converting into metabolic products other than NAD. I hold a B.S. in Biochemistry.

      q10 is great for mitochrodria. Ubiquinol has better absorbtion rates. NOW has 60x 100Mg ubiquinol for $20, or you can find 30grams of q10 on nootropicsdepot for $25

      Interesting. I'm only quoting some guy in a Reddit but 'Nad+ in the bloodstream must be converted to NR before it can be reconverted to NAD+ inside the cell, cell nucleus, and mitochondria. This is why it would be more efficacious to do the IV with NR instead of NAD+.'
      It makes wonder about sublingual NR or NAM, whether that might be better than swallowing it straight down?

      @@redberries8039 I think this guy on Reddit is behind on the studies. NAD+ in the blood is def taken up by cells. See Dr Masterjohn's last video.       Rone thanks for posting that... I just spent a grip ton of money on NAD IV’s and have been wondering if I woulda been better off taking nicotinamide riboside supplements, preferably sublingual... but, the NAD IV’s really were reviving... but, at age 50, i was also fully expecting to look 25 again by now 🤪... that did not exactly happen... maybe I need five more IV’s ... unfortunately, the FDA has banned the sales of NAD IV’s (and a slew of other safe and effective IV’s that mostly ND’s use) ... it looks like pharma wants certain IV’s out of their way as there is evidence that they are developing products using some of what is in these IV’s ... otherwise there is absolutely no reason to remove the IV’s... none were dangerous... they will be off the market in January 2020... my ND said he’s been stocking up on NAD bags... if you got em, you can administer them, but, once your doc runs out, that’s it... this also means it won’t be avail for drug addiction recovery either

    Chris, So it seems that you disagree with David Sinclair who has done all the research about this. Fair enough. However to help us could you please indicate the links to the research papers that support your claims eg taking NR at mealtimes etc,

      Sinclair wasn’t first to the research and you might want to consider the fact that he is working with pharmas to develop some kind of NAD products... not calling him a liar or anything... but, anyone working with pharma products is highly suspicious and likely going to omit or exaggerate info or the final product won’t be any better than the actual herb or vitamin they are basing their product off of. If you think pharmas don’t constantly fraudulently represent their products, and avoid ensuring people know of risks associated with their products, maybe you should look into that. Vioxx, might be a good scandal to look into. And apparently, there is currently a court cas involving Merck and their fraudulent misrepresentation of the efficacy of their MMR vaccine. It will be interesting how that case turns out.

      Dr Edward Omron has been using nicotinamide (aka niacinamide) successfully.

      Niacin has side effects that NR doesn't have, also in my experience I don't get any noticeable benefit from niacin, but the benefit is quite clear from NR.

    Very helpful. In terms of the danger mitigation techniques you describe, would you recommend the same things for NMN taken sublingually? Including taking with meals? And do you have a recommended dose of TR. Tru Niagen's version comes in 150 millgrams capsules. So if I have three Paleo-ish meals in a day, I should take one TR pill for each meal? Plus 75 mg of TMG with each meal,I guess.

    Well too methyl can also negatively impact a mental health and cause depression, anxiety, panic attacks. There are people with slow/super slow COMT gene which can affect with building up a methyl group which worsen a mental health. So taking NR in a higher dose can actually help those people by excreting methyl and improve mental health.

      I got my niagen supplement in the mail, but I’m a little scared to start taking it right now because I have a ballroom competition coming up and I don’t want to risk tanking my creatinine supply (just in case I screw up the TMG thing). I also supplement with creatinine, so I should be fine right?

      I started when I was 35, it gave (and gives) quite a noticable benefit. Some people I know tried it in their 20s and early 30s and they didn't notice anything. Perhaps it restores you in someway to a baseline of someone in their 20s, so maybe you don't need it till mid 30s?

    Is this one of the times that having mthfr gene variance is beneficial? I don’t know much about all this still learning and overwhelmed but I’ve been told my body doesn’t methlyate so as I listen to this I think hey maybe that’s a good thing in this case...:

      It may in fact make things worse, MTHFR as I understand it means you have less ability to methylate not an inability to methylate. To which there are varying degrees of inefficiency depending on whether you carry one or two copies of the gene, people carrying 2 being the worse affected. So this means that NR will take up more methyl groups leaving less for B vitamin methylation.

      Erik Fernández Resveratrol is something David Sinclair is pushing. It is highly insoluable and very little of it is bio available. Stay away from it and stick with NR

      Marko Jakovljevic Hey man thanks for the comment, I think I will still try resveratrol as I am getting 99% trans-resveratrol obtained from the Knotweed plant. It’s kinda pricey but the purest I could find. I’m probably going to stick to 200mg a day as I am younger than Sinclair and I probably shouldn’t take his dose .

      WeDon’tExist my understanding is that one would need to take impossibly high doses of tesveratrol for it to have a desired effect... but, otherwise I don’t know all that much about resveratrol... just what I heard

      I know how you feel! I started loosing my hair when I was living in S. Korea. It was terrible! I take a billion supplements and do oil treatments regularly now because I’m so paranoid about it, but I honestly don’t think that any of that is why it grew back. When I was in Korea, I couldn’t get enough calories to maintain my weight, so I think that’s the main reason it was falling out. Now that I’m back in America, it hasn’t been a problem. I’m guessing you’ll need to start by identifying the underlying cause of it. I’m curious to know what Dr. CMJ has to say about it.

      It works, that for sure. And I really felt it when I stacked with TMG. Im 3 months in, and Im gonna keep doing it. I do 500mg nicotinamide a day

      The process of metabolizing niacin uses up methyl groups. Fasting does not. So if you fast, you don't need to supplement with TMG.

    Can you recommend a TMG supplier? All of the manufacturers on Amazon make 500mg-1g capsules, which is massive overkill if I'm taking 300mg of NR and following your protocol (which would be 150mg of TMG).

    Very helpful. I've been taking what now seem to be large doses of TMG ~3000mg [level teaspoon] with my NAM at 300mg, two or three times a day. I don't think there's a danger with that much TMG?? I also take creatine ~2g twice a day. Also a high strength B-complex as I remember reading somthing about NA/NAM messing with ...something ?? maybe homocystene?? ...that is put right with one of the B vits in my complex [can't remember which] ??
    Twice a week I've just started taking NA in the mornings on an empty stomach, repeat dosing after an hour and again after an hour to supress FFA in the blood and stimulate GH ready for resistance training in a fasted state [there's research to back that up]. I've included TMG with those doses.
    Any thoughts welcome.

      I take quite large doses of TMG too. Around 3g twice a day. Maybe I should take a B-Complex alongside?

      @@rossuk2779 I remember the B-complex was niacin/niacinamide related rather than TMG related [I'm pretty sure the TMG has helped with increased reps in the gym]

    You recommended taking 100 mg of TMG for every 200 mg of NR. If you split the 2 pills of Tru Niagen over 2 meals each table has 150 mgs.
    So that would be 75 mg's of TMG, I couldn't find any products with anything close to that, all much higher? Any advice?

    i've been taking NR on empty stomach--300 mg (2 pills) and then an hour later still on empty stomach 300 mg more. believe this has increased yoverall energy level quite a bit. I work out 1.5 hrs. day and take a slew of other supplements at age 72. After this Masterjohn vid i will make another effort to see if there's more info re when and how much to take, but i do notice that David Sinclair takes nmn on empty stomach and the Resveratrol mixed with a little Yogurt.

      NMN is not NR, they have the same ultimate target, but they are different. They also have noticeable different effects in my experience, although there's a lot of overlap.

    Judging by what you said in this video it seems that you are unaware that some people do not get any athletic benefits from creatine. Also having a muscular body is not conducive to longevity. I've never seen a supercentenarian who's worried about how much muscle they have.

    Sounds like nonsense psuedoscience, you need to do tests to reach a conclusion that you need extra TMG.
