    I'm 46, and I've had nothing but improvements after months of interchangeably taking Niacin, NMN, & NAD+. Energy levels are more consistent, and my back pain has even improved after 2 years of no improvement.

      well are you an under methylator ..or over methylator ? Maybe you're an over ..which explains why Niacin goes with you.

    Niacin flush 3 gram a day had normalised my cholesterol, been on for 12 months and my readings are well with range( Total, LDL, HDL and Tryclicrides), doctor wanted to put me on statins before hand and decide to take Niacin based on A Hoffer advice, so best decision I made and feel great. Also follow Dr Berg Keto strategy and further improvements in overall health is improving. Not sure I fully understand the point of the video but interesting though...

    I'm 61 and had been taking NR for over 2 years with no negative side effect whatsoever. I started with 100 mg. But, because of these alarming warnings on a Methyl depletion risk, I've added TMZ to my diet a year ago. I felt no difference. But, the experiments are clear, the proper dosage for anti-ageing purposes is not 100mg but 500 mg to 1000 mg per day. Mice get a whole lot more than that relatively to their body mass in the lab to attain the result of a 20% life extension. So, I passed to 2x250mg of NR a day, added 400 mg of NMN, another NAD precursor, and maintain a low dose of TMZ. For the past 3 months now, I finally feel some light effect of energy increase but no negative side effect. I've also been taking all sorts of other supplements, including choline, adrenal support and MCT oil with a low sugar intake (Keto diet) for several years. I will not worry about the negative effects of NR until proven necessary. I haven't been sick, not even a cold since I started taking NR, and this is unique with regards to my past.

    I'm interested in the methyl group conjecture. Anecdotally, my experience over 12 weeks of Niagen use is pretty amazing. I'm 50, and I'm building muscle, my skin looks good, and I get a lot of compliments than I wasn't getting before. However, TMG sounds like a smart addition (really a no-brainer kind of addition) to any Niagen use. Thanks for the episode.

    I have MTHFR and have been taking 500 mg of Tru Niagen 2x per day for over 2 months and the results have been transformative. My energy, mental clarity, and overall health have been greatly enhanced. Everyone is unique and needs to find what works for them.

      I started taking Tru niagen .ended up with macular degeneration and felt really shity on it I wonder what test I should take to find out why

      Having the mrhfr gene can also mean you over metholate .... In which case probably a large part of why you feel better is not just NAD+ production but the fact it brought your metholation and neurotransmitter production back into balance. The fact niacin removes methol can be very advantagous depending on the person. It's a standard treatment protocol for people who over metholate which is indeed the cause of many issues like brain fog, poor memory and concentration, neurotransmitter imbalances, anxiety, alcoholism etc.
      If your an undermetholator however it can cause serious issues.

    I knew exactly where he was going with this! Most people don't know this but he is spot on. Methyl groups, specifically Methylfolate and Methylcobalamin are required for the production of purines and pyrimidines for DNA bases. Also, Methyl groups, such as the production of SAMe is required for a whole host of biological reactions to include the production of creatine and phosphatidylcholine (~80% of methyl groups) as well as degrading neurotransmitters, histamine and the list goes on. Too much Niacin will shut these processes dead in their tracts and I completely agree that the dosing for NR is insanely high. If you have any polymorphisms such as PEMT, GAMT, COMT, SHMT, (all MT's are methyl tranferases requiring CH3) you can pay the price and shut down these biological processes.

      Cholesterol is the building block of your brain and all cells in your body. When a doctor says that Cholesterol can cause a heart attack or stroke he means that your veins or arteries have suffered damage and that your body has dispatched emergency mechanisms using cholesterol to patch up a hole until you can regenerate the wound. Later the body will autophagy the patch and your vein or artery will be almost good as new with some new cells but also some scar tissue and how good you regenerate depends on your age or stem-cells available. Of course autophagy is nearly 100% halted by the consumption of sugars or carbohydrates and so in the standard american diet you may never have this cleansing mechanism started. Lowering cholesterol can only hurt you by slowing your circulatory system's damage control mechanism.
      A good advice to follow its to follow a low carb high fat diet and do fasting to kick start your autophagy mechanism.

      @@mattzobian You are unaware of the legal fight between Dr Abram Hoffer MD and big pharma? He fought the system for years as he was able to put many schizophrenics in remission with B3. Despite his results the journals fought to keep him from publishing his research.

    I had the same thing happen when taking 250mg ... I got extremely depressed and exhausted. I stopped using the nicotine mode riboside and slowly after a week or so my mental health improved and my energy returned. I started taking it at a much lower dose exactly as he states about 30 mg with the TMG and it works wonders for me. I have MTTR, and rapid methylation issues. If it works for you at a higher dose, great! Not everyone is the same. If it wasn’t for this doctor posting about this I never would have known what had caused my issue. Thank you Doctor!

Should this also be a consideration: "When examining the available evidence about B3 vitamins, methyl donors, and liver health, it’s also important to keep in mind that the three B3s are not equivalent. Not only do the B3 vitamins use different biosynthetic pathways to create NAD+, but they’ve also shown unique pharmacokinetic properties in mouse liver tissue.
    Nicotinamide riboside has a much larger molecular weight than the other B3 vitamins, which means that their milligram doses are not directly comparable. Two grams of niacin or nicotinamide is not equivalent to two grams of nicotinamide riboside. These doses deliver very different numbers of molecules, and this matters because it’s the total number of molecules and not their overall weight that determines how many methyl donors get used. To get the same number of molecules found in two grams of niacin-and thus put a similar strain on your methyl donor system-you would need to ingest nearly five grams of nicotinamide riboside. The highest dose clinical trial of NR to date (2 grams per day for 12 weeks) actually showed trends toward improved liver health.9"

      Hi Mary- just wanted to thank you for entering the discussion. The research on these types of supplements and their efficacy is so limited (as far as I can tell) that it seems we're in a new frontier. There's so much "promise", e.g., increased longevity, disease prevention, etc., that many uninformed (such as myself) are jumping in and trying to figure out appropriate supplements and dosages via RUclips, e.g. So, all participation by informed people like yourself is deeply appreciated.

    Been taking NR for four years and love it. Haven't been sick since I've been taking it. Annual blood work always comes back sterling. I won't be giving it up anytime soon.

      NR has been avalible commercially from at least 2014 by elysium health. NR and NMN pill doses of any company are way to low for effect + orally takein isnt as good.

    Last April I began taking about 1 gram of NMN a day and felt tons of energy in the gym every day. In May I went for my pilot medical and cholesterol was way above limits. Never had high cholesterol before. Was sent to endocrinologist. My Thyroid Stimulating Hormone reading was 60. Max is 4.2 The high number actually means it is under-performing. They pulled my medical and started me on a drug to activate the thyroid. A month later I was given the medical back. In November despite taking the medicine every day, TSH had gone up to 12.0. They took my pilot medical away again. I stopped taking the NMN and TSH number dropped like a rock. I am thinking that taking 1.5 grams a day of NMN fools your thyroid into thinking that it doesn't need to produce any.

    Started taking it and within days...days noticed cramping and fatigue at the gym. The only change to my diet was adding this to my supplements. And I do use microbolized creatine. Found this video after searching the web for info on side effects. Deleting Tru from my supplement regimen. Thanks for putting the time into the vid and posting

    Initially I did well on niacin. I was calm, it was great. I worked up to 1500 mg and then I started feeling sick. Low grade fever, lethargic, feeling of generally being unwell. I quit for a few days and then went back to a low dose of 250 mg. Still felt unwell. Since then I have learned I am an undermethylator and taking the niacin used up my methyl groups. So it's not for everyone!

    Thank you Dr. Masterjohn for your video presentation on Nicotinamide Riboside (NR). I found it to be very informative. I would like to share with you additional information and my own experience with NR. NR is in its infancy in the supplement industry. Over time more information- like your informative video presentation- will find its way to the lay public.
    I found your comments about creatine and methyl group inbalance very interesting... I haven't come across anyone who has taken the trouble to address this issue in all the mainstream buzz going on presently about NR.
    I believe that the negative comments to your video from individuals who claim not to have experienced any of the energy loss over time that you describe with NR use may be attributed to their unwitting ingestion of methyl donor foods that supply sufficient dietary amounts of choline, a methyl donor that spares TMG, otherwise known as betaine. Green leafy vegetables, eggs, certain meats, beets, peanuts-if consumed in high enough quantities may supply enough methyl donors, such as choline, to mediate the NR depletion of methyl donors, hence their conclusion that you are wrong because they have been taking NR with no signs of the drop in energy you refer to.
    Certainly the suggestion to ingest TMG is an excellent one, regardless of a person's opinion about their energy levels. TMG lowers L-homocysteine and C-reactive protein, both of which create the environment in the body for high risk of stroke and heart attack, prostate cancer, and other undesirable health conditions which lead to inflammation and disease.
    But why stop with TMG? DMG (dimethyl glycine) is another wonderful supplement that boosts enery levels noticeably through increased oxygen utilization, and also attenuates high levels of L-homocystine and CRP, among other harmful substances. Methyl folate and choline are also incredibly powerful methyl donor supplements, as well as Vit B-12 and B-6 which are also potent methyl donors. If an individual maintains adequate levels in the blood and tissues of all of the methyl donors that I've mentioned- along with TMG- NR supplementation should not be a problem.
    Thank you again for your humanitarian work and good will. And by the way, fellow RUclips viewers, it is PhDs like Masterjohn, that MDs refer to when they read (occasionally whenever they can find the energy and spare time) scholarly material on medical subject matter

      I loved your informative input. My main question after watching Dr. Masterjohn’s video was I wasn’t sure if he meant that losing the methyl donors was a permanent status or would it be temporary and fixable? So are you saying if you boost the amount of methyl donors with eating methyl donor contributing foods to give your blood a “cushion”, while taking NR it will not cause long lasting depletion of the body’s ability to produce methyl donors? I hope I’m understanding and asking in a way that makes sense. I don’t want to take anything that could permanently damage anything. I’m trying to heal not harm. Thanks!

    seems like good advice.. I've noticed an inconsistent effect with 250 Nicotinamide Riboside.. so I take it occasionally with good breaks.. however I love how it (not always) gives me that effect where I wake up in the morning like I did in my 20s.. which is less inflammation and grogginess.
    Or when it lifts my mood an hour or so after taking. Its an interesting supplement for sure.

    Thanks for the info! Life Extension has 100mg of nicotinamide riboside, in capsules. You can buy small gel caps and easily divide them up into halves, thirds or quarters.

      They want to maximise profit even at the coast of lives.

      The problem with natural things is that the risk of failure or worse results are always possible. Also you need to do it at least once for finding out how effective the method is. And you need exactly the right conditions.
      When using artificially made compounds you know exactly how much of the end product you can expect.
      These companies plan ahead. Failing to reach a goal might cause losses of millions of dollars. Plus if it's exactly the same compound it doesn't matter wether it's natural or artificially made.
      When taking stuff like plant based products it's even harder. So if you need a certain dose artificially made products are always better.
      But nevertheless a lot of drugs are naturally made compounds. For example all antibiotics are made naturally.
      I'm not defending these companies, they are doing a lot of crappy stuff, but I am defending artificially made compounds.

    From my personal experience Chris is right. For some people under methylation from too much niacin can be very unpleasant. Both NR and NMM slowed my methylation dramatically.

    For those who simply do not understand.
    First, understand the methylation cycle. Methyl Groups is required for DNA Methylation. And various functions. Since most people are undermethylated, especially if you take unmethylated B-Vitamins, like cyanocobalamin, or folic acid, or pyridoxine hydrochloride.
    Vitamin B3 consumes methyl group.
    You can take high dosage of B3, so long you have enough methyl groups. Here you can see the balance...
    So if you do take high amounts of B3, take methyl groups from TMG or SAMe or Choline Bitartrate. Choline Bitartrate converts into TMG.

      @@notproproductions2025 If you want the benefits of B3, you can take NR or NMN, both are very available. Don't take unmethylated Bs. A very good B-Complex is thr AOR B-Complex Advanced. It contains choline bitartrate in it. You can add a little Trimethylglycine for extra methyl groups. Like 200mg. SAMe is expensive. You can 1 mg of folinic acid not folic acid. Methylfolate is for the methylation cycle. Folinic acid for DNA repair. You want slightly overmethylation, you don't want too much overmethylation as many people have disorder in different areas.

      Depends on the amount other undermethylated Bs. 500mg of TMG is more than enough unless you take huge amount of unmethylated Bs. Tell me your complete Bs list and form of Bs.

      1000mg then trimethylglycine. U may want to consider to switch to a safer bioavailable methylated b complex like AOR B Complex Advanced. At certain days u can take 500mg. Note that pesticides inhibit methylation n also dairy antibodies. High carb as well.

    sounds like this stuff should be used in cycles, like steroids...give your body a bit of a chance to recover and replenish its self on off cycles, then continue.

    Thanks for sharing. I do love the niacin flush. Such a rush of energy. However im concerned it depletes ceruloplasmin. Just like it require methyl supposedly ceruloplasmin spend in the process.

    I believe that what this woman went through that posted to his Facebook was actually likely a mild detox, what a lot of people don’t realise is that niacin and related molecules strongly and quickly detox the body which will manifest in the symptoms she describes, she even mentions she feels toxic!
    In-fact it’s common knowledge that drug users use high dose niacin to pass urinal drug screening.
    Taking it for several days is apparently enough to completely remove metabolites that may take months otherwise.

      I have really bad anxiety I guess it is what it is I really don’t know it happens every month I get a blood rush rush rush through me and it goes to my brain and I get really scared like I feel like I’m going to die and it makes me think of something of head before in the past it will last sometimes two days sometimes one but then I’m weak and tired and I feel depressed for days I am starting niacin I hope and pray this works it seems like it has helped me I’ve only tried it one day and it seems like I’m not having them attacks this is my Third day if you got any suggestions please let me know if it’s helping you thanks for sharing

    I found your talk interesting, however I react really badly to TMG. I am taking a higher dose of niacin due to the fact I have some itchy dry skin. I also have high histamine, gluten sensitivity and CBS snp that causes me to sensitive to high thiol sulfur. The extra niacinamide has really helped, although it hasn't done the full thing. I also seem to be a fast methylator which seems to be connected with excessive reaction/sensitivity to many foods. So perhaps for some of us we actually need more niacin. Of course its a learning process.

    I took NMN/Resveratrol for 9 months, Sinclair sized doses. I am 49. I am vegan. I noticed very little effect, but the one thing it did help is that it dulled the numbness and tingling I have in my feet from nerve damage that started several years ago. It went away real gradually, but about 2 weeks after I quit the Sinclair regimen the tingling and burning/tingling came right back. If I could get NMN cheaper I would do it again.

    It can be very economical to buy way to strong supplement, and then just take a fraction of them. Usually means less crap like fillers and such as well.
    B-vitamins may be a bit bitter but that is probably not an issue if taken in RDI doses. The only capsule I’ve done this with which tasted completely horrendous was some kind of bluish copper/zinc supplement.

    Hi Chris! This is a very interesting and important information, which is very rarely given in such a detail. Thanks a lot for it! I have a question about the connection between TMG (betaine) and TMAO. I know that TMAO is very dangerous and usually is associated with high risks of heart disease. Can TMG become TMAO? If this is so, than what can be done to prevent it?
    My second question is about Methioninum. If I'm not mistaken, it is also a methyl donor. And it is a commonly used liver protector. Can Methioninum be used instead of TMG? Is so, what should be the necessary dosage in comparison to that of TMG, when taken with niacin or nicotinamide?

    I had a similar experience with Niagen. After a short time taking it I noticed my run times were worse and I couldn’t do as many reps in the gym. Just generally felt bad and out of energy. Threw the Niagen in the trash and started feeling better. The TMG is an interesting idea.

      I had a similiar experience with NR. I have found sublingual NMN and NAD+ to not have those effects currently, though I do cycle off at least one day a week and take trimethyglycine, as well as, creatine and phosphatidylcholine to leave more SAM-E for DNA repair and other essential demands.

      I had the same thing happen when taking 250mg ... I got extremely depressed and exhausted. I stopped using the nicotine mode riboside and slowly after a week or so my mental health improved and my energy returned. I started taking it at a much lower dose exactly as he states about 30 mg with the TMG and it works wonders for me. I have MTTR, and rapid methylation issues.

      please help I have MTHFR, liver problems and niacin deficiency. I tried nicotinic acid at low doses but even low doses caused me low methylation issues(sneezing itching and many more).
      How can I take nicotinamide riboside at low levels.

    No problem - just take TMG (which I do for both the methyl groups and the glycine that I combine with NAC). So I get my NAD boosted along with Gly-NAC.

    Thank you for this video. I was taking TruNiagen 150mg for under 3 weeks and for the last week have been feeling really horrible - insomnia, feeling very stressed, no short term memory, anxious mind, tired. I have a partial MTHFR mutation. I just went to test my TSH/T3 because it felt like I was having hyperthyroid, it was that bad.

    Thanks for the great video and explaining it in a way that we can all understand. I personally deal with really bad anxiety and I’ve been told to take VERY HIGH DOSES of niacin and it actually made me feel very dizzy, lightheaded and my anxiety was a lot worse. Currently taking medication for anxiety btw. I’ve learned a lot about methylation and learned that niacin depletes methyl donors in the body. I’ve heard that MOST people that have anxiety and depression are low methylators - do you find that to be the case as well?

    This makes perfect sense to me. The op with the "bs detector" probably runs his petrol car while ignoring water and oil levels because cars run on petrol. True cars do run on petrol but they also use smaller amount of other fluids while consuming the petrol.
    This is a basic analogy but it should ring true to many of us.
    The body works in a very similar yet more complex way.

      The cells in younger people are more efficient at doing the tasks required of them to produce NAD when they are youthful. I heard the communications breakdown between the cell and something else as we age. NMN can get them talking again and working better together again.

    Hi Chris! I know this is a LITE episode, so you don't go into all the details, but I'm curious what you think about the risks associated with Niacin. It seems that it can cause Liver toxicity, so many doctors will check liver function every 6 to 8 weeks for the 1st year after starting their patients on it. Is this true of all forms or is it specific to Niacin? Also, I've read that Niacin can increase Blood sugar. Again, how significant is this and does it occur with all forms or just Niacin itself? Thanks for your episodes, these are awesome!

      I take niacin along with chromium, to balance out the blood sugar. (Chromium increases insulin sensitivity, and tends to lower blood glucose.) Re: liver problems, they happen with the SLOW RELEASE (non-flush) version, not with regular niacin.

    From what I have read, it seems that “no flush niacin” is replacing true B3 in most available products. And the no flush form does not have the lipid reduction effect of unadulterated niacin. Thisis important.

    What about absorbing Niacinamide through skin? It dissolves in glycerol and can be applied to skin. To scalp of your head, as an example. It may be beneficial for hair growth too.
    I got problems with my liver when I tried above 3000 mg a day of Niacinamide a day. With lower dosage like 500 and 1000 mg it was no problem. Also, I am keeping OMAD and have a late dinner while I was taking Niacinamide at morning to increase my energy for morning exercises - 45 min cycling.

    I've been looking into NR. I am an elder, and have been looking into nutrition for decades... it is an endless task and I expect to study for the rest of my life.
    I appreciate your input and will look into your suggestions... including getting a mg scale.
    BTW... off subject... your shirt... although an exciting color, is way too small... the buttons are screaming.

      I think the shirt has suffered from too much wear and tear. And maybe my chest and shoulders are getting bigger. In any case, I ordered a new red shirt today, in the fit of the other shirts I wear. The new fit will be designed for athletic instead of slim body so it fits better. Should be in the videos that come out in February. Haha.

I had my 23andMe test results analysed by NutraHacker which advised that I "avoid methyl donors" (due to COMT & CBS SNPs) so I guess that means I can't take TMG & therefore also can't take niacin? I previously took 500 mg daily of niacinamide which caused aching around my eyes & increased tiredness so, instead, I've been taking 250 mg with no noticeable ill effects but now fear that's problematic! SNPs seem to make supplementation extremely difficult. Any input greatly appreciated!

    Based on my own experience and some anecdotal reports from Amazon product reviews and web forum posts, I suspect that the problem with boosting NAD+ (via NR, anyway) is that it futzes with purinergic signalling and so messes with autonomic function. Sorry for the vague and not so scientific language, but it is the best I can do today.

    I have often went months taking between 500 and 3000mg of niacin daily and I've always felt great after it. It has been known for years to be a great treatment for schizophrenia cholesterol levels and arthritis to name of few ailments. 35 mg a day is absolutely useless. Look at abhram hoffers years of work on niacin for starters. This guy here speaks a lot and you would think he knows what hes on about if you didnt know better from experience, and listened to doctors who have treated thousands of people with high doses of niacin. He hasnt a clue of how to take or use niacin therapeutically. Phd doesnt mean he knows everything.

    What about taking NADH instead. It's supposed to be the active form of NAD and usually comes in no more than 20mg. I just started taking 15mg MethylFolate with a B complex for anxiety along with Sam-e and creatine because I heard most of the Sam-e you take ends up being used to help your body create creatine... so if you takr creatine powder it will prevent your body from using up the Sam-e supplement.
    But from what I just heard you say about taking too much niacin (NAD) and your body excreting methyl groups along with it has me worried that taking a B-complex along with NADH even though its only 20 mg... might be compromising the 15mg MethylFolate im taking. I just looked at my B-complex bottle and it says it has 100 mg of niacin... is that too much along with the 20 mg of NADH im taking? I dont want to be compromising the MethylFolate im taking.
    Maybe I should find a B-complex with less niacin or take each B vitamin separately to avoid all that niacin because my multi vitamin has 50mg of niacinamide too!

    Thanks for the info, Chris! If I get any side effects from the NR, I'll try adding TMG. I know you are HIGHLY respected in the field of nutrition/biochemistry and know FAR MORE than your average MD (who just aren't educated in nutrition). Sorry that there are so many degrading comments, you're just trying to educate us on what to do if anyone experiences negative side effects on NR.

    Isnt this where Glycine can help? Found in collagen it offsets methylation

    Appreciate your recommendation to cut doses via powders or splitting capsules. I do this personally for creatine, pre-workout/caffeine to personalize doses.

      Fantastic! Thank you! I’ve been splitting my 100mg nico in 3parts and doing great. We can also skip, and take it once every tow days. We don’t NEEEEED all supp and vitamins daily. Great quality foods make up for a lot. Supplements are for support. You won’t see me ingest 24 caps a day of anything. I have them all on an intelligent rotation and that’s been working for me. Let’s see how I do if Covid hits me smack in the face 😳🤷‍♀️🤪

    Fill the scoop with water. Pour the water into a measuring cup, note level. Scoop desired divided portion of niacinamide with a correlating measuring spoon.

    I need 1000 mg on straight niacin (flash one) because I'm getting off anti psychotic medicine
    its really spending lot of methyl ?
    because I have under methylation
