    What about sublingual NMN? I have absorption issues all around so I use sublinguals and patches a lot. I had a weird thing happen when I started using injectible insulin where it all became active at once when I took a sublingual NMN. Turns out you need B3 to activate that form of insulin and I was deficit.

    Hey Chris, I was wondering if you also can take Msm (methylsulfonylmethane) instead tmg for providing methyl groups? I only hear about tmg every time...

    There is some discussion online among people who take NMN or NR regarding whether or not to refrigerate it. And some have asked if it is already spoiled by the time they get it at the store. Any thoughts?

      Rule-of-thumb for the temperature dependence of a reaction rate is that a ten degree rise in the temperature approximately doubles the rate. The converse then, is that a ten degree drop halves the reaction rate. Why important? To minimize the degradation reactions with impurities in supplements and drugs. Bottom line: yes, refrigerate (and keep them dry).

    Hi Chris,. Instead of TMG any thoughts on going with food and just eating a beet every day? For reference I'm taking 300 mg NR. Experimented earlier with 500 mg of NMN. Prefer NR for one of the reasons you brought up, more human studies.

    Thanks for vid. For past week I have taken NR sublingual. Mornings, I dissolve 300 mg in shot glass with a bit of water and pour under tongue, holding 20 minutes. I like the effect so far. Biggest NR benefits over last 4 months for me have been better flexibility, skin, mood. Hope I'm not demethylating!

    Can I take both Nicotinic acid and nr at the same time?

      I have me/cfs, and am currently taking Nicotinic acid. It helps, but still a way to go. Want to try adding on nr to see if it helps. As you mentioned it goes through different pathways. Another factor could be the gpr109a receptor protein, and/or the tryptophan recovery pathway. Any insight you might have would be helpful

    David Sinclair said that he did the experiment (NMN) on mice with small doses and there is no effect.

      David sinclair said he takes NMN and it boosts the effects of resveratrol so it does have an effect.

    If I intend to take 100 mg NAD + per day, how much TMG powder should I add to my diet ? or should I consume the raw version of NAD instead ?

    There have been some more research this past year on NAD+ boosters, specifically human studies on NMN. Also, liposomal NMN has come on the market, although one could always make it at home. It seems to me, since the liposomal form doesn't degrade and crosses the cell membrane intact, there would be no need for supplementing methyl groups.

      I could add a comment about NR. Many argue it's an inferior NAD+ booster. The evidence can be interpreted that NR degrades so fully in the gut and bloodstream that little of it, if any of it, remains intact so as to be transported into cells. It effectively might be nothing more than an expensive niacin supplement.
      One might be able to avoid this issue with liposomal NR. But, as far as I know, there is no liposomal NR on the market. Of course, if one could get NR in powder form, it could be made into liposomal form with some phospholipids and a high speed blender. For whatever reason, I've never hard of an NR advocate speak of liposomes.
      But, considering NMN is further on the salvage pathway than NR, why not use liposomal NMN? Or why not just skip the salvage pathway by directly supplementing with liposomal NAD+? Both of those are available as high quality products that will guarantee high bioavailability. I'm simply not seeing the benefits of non-liposomal NR or, for that matter, NR in general.

    Can you take NR at night with TMG? Or when is the best time to take it? I watched the video about diabetes and taking NR where you talk about carbs and fats being used differently

    N=1 but I've noticed a steep difference in mental agility and sleep pattern after being off 300mg of NR (ran out of my Thorne Resveracel) for 2 - 3 weeks, once.
    By the way, I've been running 600mg of TMG for 300mg of NR, is there any problem with that? Just curious :)..
    I'll try splitting the intake between meals and see how to goes.

    I believe Sinclair says NR needs to be converted into NMN in order to enter the cell via NMN receptor. (Found my fitness podcast).

    Do you think that Choline(Alpha GPC) could work as effectively for
    methylation as TMG does? It's a lot easier to get quality Choline form
    than TMG.

    I'm having trouble finding info on how or if to take NMN as one who under-methylates. Anyone have some experience with this?

      All forms of B3, including NMN, NR, NA and NAM, use up methyls as they are metabolized. To compensate for this, many people are taking either methyl folate or TMG.

      @@Nilsosmar I would be really careful if I were you, mscoldham, about taking it if you are already an undermethylator. Have you had a whole blood histamine test done to confirm you're an undermethylator? Dr. Walsh, who is a scientist and expert on methylation and mental health and other disorders (you may know of him :), strongly advises against folates & niacin for undermethylators. But, if you really want to take B3 or NR - if it were me anyway - I would really monitor things closely and get bloodwork done as well as keeping an eye on how you are feeling. If I were an undermethylator, I would do more research on what supplements can be taken to balance things out with methylation. I recommend Dr. Walsh's book, Nutrient Power, or his website also has a lot of information.

      @@LifeDIY Yes, blood was tested last year after I watched and studied quite a bit of Dr Walsh's info. SAMe made me feel great. I did stop taking it because I am now eating lots of meat. I am just confused bc of what Dr Walsh says about not taking Folate if under-methylating. I will have to do more research. Thanks for the replies!

    Hello, I have one problem taking NMN. Hope you can help with your knowledge 😃🙏 I take 0,5 - 1 gramm Resveratrol in Oliveoil, 0,5 gramm of NMN and 1 gramm of TMG. I started taking TMG as I noticed, that after about 3 days I feel a certain pressure on my heart or let's say on the left side of the breast and I feel like having higher blood pressure. Then I stop the life extension treatment and everything is fine again. When I take only Resveratrol or only TMG this phenomenon doesn't appear. I thought with taking TMG this would stop, but that's not the case. After starting again the same reaction appears. I bought NMN even from Prohealth. The same thing. The dose is really not that high, so I am wondering as the studies don't have such results. I am 45, 1,75 m and about 83 kg. So what's your opinion on that? Thanks in advance, Thomas

    can anti-oxident supplements like vitamin C - interfere with a body naturally producing nitric oxide?

      Dont take antioxidants before or after training. It will most likely blunt the positive effects of the training

      Dr Sinclair said NR converts to NMN convert to NAD. And NR is less stable, and when NR expires, it does harm.

    I don't think nmn going to do anything..our human organ more complex than mice...and we need surtuin Gene to activation ..and what sert 1 to 7 do to our body. And base on it .we have to find molecules...nad will nothing will do with anti aging... promise on it
