  • From the Creation of the First Knockout Mouse Scientists have come very far; with CRISPR technologies being the main method used by genetic knockout scientists today, the traditional esc based technology explained in this video is something of the past, mainly due to the length of time these projects take to complete (8-12 months).
    Please note that any percentages mentioned in this video should be taken with a grain of salt as they are not exact values. For example, the targeting efficiency of vectors vary greatly not only from loci to loci, but also from vectors to vectors. 10% is just a rough average targeting efficiency in our hands.

    Hi Cyagen I have a quick question: how are we putting an antibiotic resistance gene into a eukaryotic cell? The stem cells from the blastocyst are from a eukaryotic organism.. so I'd assume that antibiotics wouldn't be able to kill these cells anyways? Please let me know if I'm misunderstanding something. Thanks!!

      I was actually wondering the same thing, why do we add neomycin, if eukaryotic cells don’t get affected by antibiotics.

    If we do PCR of all ESC, how are we left with cells to be implanted in the new mice blastocyst?

      Typically, one or more subcultures of cells can be made for analysis and selection of the correctly modified cells.
