    Regarding the step 3 of transgenesis, the procedure through which the transgene is introduced is called TRANSFECTION. Transformation is done autonomously by micro-organisms ( as demonstrated by the Griffith experiment).

    Could i use this technique to over-express a gene already in a mouse? If not, what other methods can i use for this?

      Transgenic insertion of a plasmid containing the mouse gene of interest may be performed to generate an overexpression model, but this has an unknown number of gene insertions. For a single-copy insertion overexpression model, we can perform PiggyBac-on-BAC transgenesis.

    plasmids occur in prokaryotes. can recombinant plasmids be inserted into mice?

    in the 3rd step of transgenesis you insert gene of intrest with vector (plasmid) , why cant we just directly inject naked gene into the male pronucleus
