  • In this video, Biology Professor (Twitter: @DrWhitneyHolden) will describe the Indole test procedure and discuss the background information to understand how this test works. Specifically, this test identifies bacteria that can convert tryptophan to indole with tryptophanase enzymes, which is made recognizable by use of Kovac's reagent to give a cherry red dye in indole-positive bacteria.

    i am studing from spain. I understand english a little a beleive me microbiological test are very difficult for me, I never study this before. Now I am preparing an exam and I need to say, you are my heroe teacher, (((((thank you))))

    Thank you so much for all the information, it is very clear and simple. Just a suggestion, you may need a new red marker since the notes in red are very blurry and have many other color markers to address different points while teaching. Again, thanks so much for your knowledgeable lessons are very helpful.

    Does this test only work Indoles or can you also do it Outdoles.

    if an organism possess tryptophanase complex that could turn tryptophan in to pyruvate and indole, that might be an indication that the organism is aerobic or facultative anaerobe but definitely not obligate anaerobe, right?

      Not necessarily. There are anaerobic fermentation pathways that use pyruvate. It isn’t only for aerobic respiration.

    Miss I have a question... I want to know that can we take any sample for this test I mean can we take water as sample to perform indole test or do I have to take other some specific test sample...Thank you

      You need a pure bacterial culture that has been growing in the lab for at least 24 hours. You can't just use a water sample, as it will have multiple species of organisms in it and also have not nearly a large enough amount of bacteria to make the test work.

    Hi there! Thanks for the great video.
    Is indole test a reliable test in identifying bacteria or does it need other biochemical tests to support it like the IMVIC tests?

      The indole test will tell you if a bacterium is indole-positive or indole-negative. You would need additional biochemical tests or PCR/sequencing to be certain of a species identification.
