Notch Signaling Pathway | Purpose and Mechanism

  • Lesson on the Notch Signaling Pathway. The Notch signaling pathway is a highly conserved and critically important pathway in homeostasis and the development of sensory hair cells and branched arterial networks. The Notch signaling pathway derives its name from the NOTCH receptors, which are activated through ligands located on the cell membrane of other cells (sending cells). The NOTCH receptor is composed of three domains, the extracellular domain, a single transmembrane domain, and a notch intracellular domain (NICD). Upon ligand binding to the Notch receptor, the Notch receptor undergoes cleavage reactions, releasing the NICD into the cytosol. The NICD binds to the complex of proteins, which translocate into the nucleus and activate Notch target genes involved in cellular proliferation, differentiation and apoptosis.

    Brilliant concise video! I think the cleavage of intracellular domain of Notch is usually called gamma-secretase.

    From what I read, I thought the NICD translocates into the nucleus first and then binds CSL? Am I wrong?

      you're right. NICD translocates to the nucleus where it interacts with CSL

      It is not an acronym. It comes from Drosophila research: the phenotype which showed a little 'notch' in the wing. As this is a highly conserved pathway, similar genes are found in humans.
