Could you clarify the significant cholesterol increase with resveratrol? It was one of the main points Dr. Brad mentioned in his "Why I stopped taking them" video (7min.40secs.) and I did not find you mentioned that *No beneficial effects of resveratrol on metabolic syndrome* study in your video. I take resveratrol every day and exercise every day, including intense exercise roughly every other day, and I haven't noticed any issues with being able to exercise or recover. If anything, I am in excellent shape and have only gotten better lately. I think I'll just keep taking it daily.
Same here, I run nearly every morning fasted after having my coffee and taking 250mg nmn and feel great. And most NMN supplements already include resveratrol so separating it is impossible.

I say this because throughout the pages of history, too many people, smart people, fell for the fountain of youth scam. This youth potion is nothing new and very powerful in its influence. I’ve been taking daily but I also run long distances 4-5 days a week. And I’ve been noticing my VO2 max has been declining and I’m not recovering well. Definitely going to take a break for a week now then taking every other day
I’d like to know the truth about Resveratrol: is it good for you or is it potentially dangerous? Why did I have a very obvious, repeatable result from taking resveratrol?… I never expected any visible results from taking it but I began to see that it was indeed responsible for increased endurance in a strenuous “ yoga with weights” class during which we increased heart rate and worked our muscles to their limit. I tested it over and over again. I stopped taking resveratrol because of the negativity around it, but I also had to stop taking the class!

How come Dr. Sinclair gets nervous when confronted about this. He looks down and away and say’s “It’s confusing” …. What do you mean by saying “Dr. Brad isn’t a researcher” when all he’s doing is clearly explaining what went wrong with David Sinclair’s research, the mistakes he made and the inaccurate conclusions he drew, and presents the evidence contradicting Sinclair’s claims! Hi A G, I don't mention brands because 1) I don't cooperate with them so you could trust me 2) I once mentioned a brand and they changed the formula leaving me with an egg on my face. However... I'm working on huge resveratrol project, where I show labels of products I use. I don't say the brands. And you could search and find that for yourself based on that label. The resveratrol dose matters greatly, not just the brand.

Good video. Indeed there are some questions still pending in my opinion. The impact on resistance exercise for instance: the inflammation and post recovery and overcompensation is based on stress (caused by mechanical and metabolic work): protein synthesis occurs in the hours post exercise so I do assume resveratrol should not be taken anyway post exercise in these cases. Following your good work and perhaps we'll have a podcast together in the future;-)

How are you? I think cycling is not needed with NMN. It works in a "build up" mode. It more like taking magnesium supplement. NAD+ mechanism isn't susceptible no benefited by cycling as far as current data. For example, Sinclair found it takes 9 days for NMN to reach it peak active mode, and you don't want to go down from there.

You're very kind Otto. First, fisetin is very interesting but it has 1% of data compared to resveratrol. Having said, that In 2 weeks' time, you'll get a mind-blowing video on that exact subject. It's a project I've been working on for the last 2 months, took me covering nearly 200 studies, and will include polyphenols as a group. I saw your comment below about whether NMN should also be cycled and was wondering what you would think about taking astaxanthin every day. I take it for my skin but maybe shouldnt everyday? I take it to help protect against UV rays.
Hi Claire, can't give personalized advice. I can tell you those fat-soluble antioxidants - unless taken for a specific time for a targeted goal (e.g. 1-3 weeks) can turn off the natural defense system and reduce longevity - WHEN we're young. I"m very careful with them. Astaxanthin should work better after age 70 when we begin to oxidize our brains, eyes, and ears...all fatty tissues. And from what I've seen, it can cross the blood-brain barrier. Today, applied topically against sunshine should be rather safe - I think. Also maybe taking orally before a very sunny vacation and that's it.
Brad did not make money unlike Sinclair, nor did he discredit resveratrol. He simply cited the numerous studies that discredited resveratrol to include CRISPR. Are you saying CRISPR is so incompetent that they misunderstood the studies? Lol maybe. But it's more than likely it is Sinclair that botched the study and refuses to engage in peer reviewed debate.
So what I understand from the video is the benefits of resveratrol are the most when taking it every other day, and dr. Sinclaire also confirms it. So I wonder why dr. sinclaire says in hes book Lifespan that he takes it ever day.

Stanfield for all his emphasis on methodology and rigorous/accurate interpretation of data, all too often selectively cherrypicks research just for the sake of being contrarian. His assertion that it is not even clear that activating sirtuins is an anti-aging mechanism based on 1 sirt-2 study is downright ridiculous. To make such a statement while omitting literal piles of research estabilishing the crucial role of sirtuins (and specifically sirt-1) in aging of various tissues from bones to ovaries, is either ignorant (again, Stanfield is MD, not a real researcher) or malicious. He has proven himself to be a somewhat sleazy opportunist and considering his considerable lack of expertise or even academic merit, his public persona is all the more inapproperiate.

Hi, I hope this comments finds you well. I’m a scuba diver- few months ago I started to take resveratrol , nmn, metformin and some other molecules mentioned by Mr Sinclair. For the first time during my dives (I’m a nitrox diver) I started to experience scary heart palpitations when I was below 20m - it usually would happen at 30-25m depth. At first I thought it was narcosis but then I decided to stop taking resveratrol (intuition) and my dives were going perfectly again. The same problem occurred when I used a hyperbaric chamber (I have it at my gym) - I experienced the same reaction as during diving and they had to take me out of the chamber. When I used it without resveratrol I was again- absolutely fine. Does it make sense to you? I hope you can shine some light on this as it’s been really on my mind. This days I never take resveratrol when I’m on a diving trip or when I know I may use hypoberic. Tks for your time .

It sounds like something that causes your body to become more irritable under physical stress (Water, oxygen levels). I don't have an explainable mechanism. Possible explanations: Allergy to resveraratrol Allergy to compound in the capsule itself, including remains from the plant from which resveratrol was extracted. Over activation of the stress system by resveratrol via an unknown mechanism. Too much high dose. Have you asked your doctor maybe? In the next video on this channel, I show how resveratrol dose changes the reaction of our bodies.
Hi Rimon, I’m very well thank you. Yes , it wasn’t the best experience to get a major heart palpitations at 27m and trying to ascend as slowly as possible. I’m glad I worked out what it was tbh as diving is my passion. I do think it might have been a combination of high doze (1g) and high oxygen… I do breathe 30-33% O2 being on nitrox. I was even more certain it was resveratrol after it happened in hyperbaric. Anyway I do agree with your comment of taking it every other day- I’ve also decreased a dose to 0.5g. I noticed that 1g makes me anxious somehow but I feel fine with 0.5g. Regarding my doctor- I’ve only recently had my first visit with functional medicine doctor but we didn’t have a chance to talk Abt this particular problem. Just a wild guess Edyta, resveratrol increased total cholesterol significantly, from the *No beneficial effects of resveratrol on methabolic syndrom* study Rimon forgot to mention at 1:25 (from Dr. Brad's video around the 8th min. mark). In higher pressurized environments, serum-cholesterol may clot arteries more easily which could cause palpitations... I bet you'd not be the only one showing those symptoms under those conditions.
There is possibly a link between cycling creatine and Resveratrol. One creates cellular on a small scale promoting recovery demands and the other providing the energy needed to meet those demands. Research would be needed but this is possibly a compass.
Do you have a reference for the paper David Sinclair said showed regular diet fed mice had benefit when resveratrol was given on alternating days? I've looked at some of his papers but could not find that data in the supplemental material.
Hi David! I remember seeing that in 2019 when I delved into this for my own supplementation. Now, I went intuitively to the original study, and you're right - No mention there in the supplemental data. But it says in the paper: "Analyses of the other groups will be presented at a later date." The standard diet + resveratrol on and off had to be "other groups". I think that I've seen it in a subsequent paper, that I've seen that. Supplementary data: "Beginning at one year of age, SD and EOD (ME -"every other day") groups were fed a standard AIN-93G diet or AIN-93G plus 0.01% or 0.04% resveratrol... The EODLR had the longest survival times..." 2. Sinclair mentions that clearly in this paper from 2014 I think that's what I read in 2019: www.ncbi.nlm.nih.gov/pmc/articles/PMC3970218/ I'm quoting: "However, resveratrol does extend mouse lifespan when provided in a high calorie diet or in standard chow fed to mice every other day"

Interesting that Sinclair appears to take it every day (skipping days when he forgets for whatever reason). Would be good if somebody could ask him about this in the context of these studies.
I've noticed that supplements seem to reduce my kidney function. So I've cut back to just good diet, daily exercise (alternating RT, Zone 2, HIIT and 10K steps) and occasional fasting. Maybe that's enough. I used to take resveratrol, but can't imagine taking it now.
there is no controversy, just a GP (Stanfield) that knows about as much molecular cell biology as my english school teacher. There is *some* discussion about the exact involvement of sirtuins, but the overwhelming evidence (and not just from the Sinclair lab) shows that they are absolutely involved in CR mediated lifespan increase. Obviously they are not the only pathway, but there is never just one pathway regulating "big" functions like cell senescene, so its not an either/or with respect to sirtuins and rapamycin. If anything, supplementing nutritionally, with resveratrol and NMN is far preferable than taking a drug like rapamycin. Also, if you read the paper showing the low level replicative stress carefully, they do discuss their results in the context of the plethora of evidence on resveratrol and lifespan extension, and they are not actually mutually exclusive because low level relicative stress is actually one of the putative mechanisms unerpining lifespan extension even accroding to Sinclair's layman explanation. OVerall the literature has an astounding amount of data linking caloric restriction, sirtuins and sirtuin activator compounds and the rest is various randos on the internet trying to catch a bit of the spotlight and make a name for themselves (like that stanfield guy) on youtube by throwing around provovative statements like "convtoversy about resveratrol", which is laughable. Thankfully real science doesn't care about them

I am finding it so hard to parse information on ageing when the top ageing experts like Professor David Sinclair, Charles Brenner, Matt Kaeberlein and Brian Kennedy all disagree with each other and argue so vehemently about it on Twitter. It's honestly so bad for science communication and for lay people like me wanting to know if they should take a supplement and which molecule is better. I wish the ageing experts would come together for some sort of friendly joint debate or discussion. My impression is that while there are some studies studies, there maybe aren't large-scale, strong human trials on supplements like NR, NMN, metformin, rapamycin, so forth? Or they are still underway? It's so confusing, honestly lol.
@Jessica _S I agree with you, it's very confusing. Remember that each lab researcher has seen different results. I think it's -- part of their identity. Also, the answer many times is intuition. Because each study gives a tiny, tiny cube of evidence (that may be incorrect), as well. Thus, lab researchers, many have different intuitions about where we should go. And it's very difficult to put intuition into a debate. You can only prove the cubes. Only your mind can see the big picture - hard to argue around that. The results I have put were never done in a single study but followed by the intuition of how the human body works. Metformin - many, many human studies. Been in use in humans for 60-70 years now. NR& NMN - Most of the human studies are from NR because it has a patent. So there is money in it. It doesn't take anything from NMN There is a lot of benefit in mice studies, our evolutionary cousins.
Did you not do a video where you suggested too much resveratrol was more beneficial to those who were sick (I.e. cancer patients) and less is more beneficial for healthy people? Why are you taking 1g, isn’t that higher dose? I thought you suggested between 250-500mg for healthy people?

Good question Christian. Do you mean, you stay in fat-burning mode the whole exercise session? Can't give personalized advice, if it were me, any cardiovascular activity (and even resistance) I'd delay resveratrol afterward. It may even help with the recovery this way.

That study about exercise was short term, people forget your body adapts to things over time. Why don't they run blood test on people who have taken resverstrol for years. Cause a lot of those people seem in great shape. It appears resverstrol didn't harm their exercise.

External antioxidants = supplements. The same results have not been showed in consuming antioxidant rich whole plant foods. In fact, the opposite results have been showed
@ How aer you? If I got your point right (not sure), also plants have many other molecules and their levels of antioxidants are not as high as in supplements. They have additional benefits beyond the scope of youtube comments. So you have a point.
@@WellnessMessiah I’m good man thanks :) My point being is that these studies which indicate that antioxidants are bad have studied supplements. However, when you study plant foods rich in antioxidants (Blueberries, Cacao, etc) you do not find the same results. So, you cannot say antioxidants are bad, only that supplemental antioxidants are bad. Otherwise, this would a very reductionistic statement. Plants contain thousands of phytochemicals, when ingested as a whole, they provide antioxidant effects but also other effects that have not been shown to be bad on exercise or on youth. I’m not saying supplements are bad, I take more than 20 supplements. But I have noticed people claiming antioxidants are bad and they’re basing these claims on these studies which only studied supplements. That’s like studying an anthocyanin supplement, getting a result, and claiming that antioxidants are bad. But antioxidants from blueberries aren’t bad. In fact, they’re good. Plants contain hundreds of thousands of phytochemicals in a food matrix and this significantly changes how it acts on the body. You see it time and time again. Isolating one compound and studying it and seeing it doesn’t work. However, when you take plant itself, it works better. Resveratrol might be an exception because of its low bioavailability and that it exists only in very small milligrams in plants.
Your initial premise is WRONG!! There is no such thing as reversing aging, unless you go back in time. You can slow down the aging process, but nit reverse it.

However... I'm working on huge resveratrol project, where I show labels of products I use. I don't say the brands. And you could search and find that for yourself based on that label.
The resveratrol dose matters greatly, not just the brand.

Good video. Indeed there are some questions still pending in my opinion. The impact on resistance exercise for instance: the inflammation and post recovery and overcompensation is based on stress (caused by mechanical and metabolic work): protein synthesis occurs in the hours post exercise so I do assume resveratrol should not be taken anyway post exercise in these cases. Following your good work and perhaps we'll have a podcast together in the future;-)

I think cycling is not needed with NMN. It works in a "build up" mode. It more like taking magnesium supplement.
NAD+ mechanism isn't susceptible no benefited by cycling as far as current data.
For example, Sinclair found it takes 9 days for NMN to reach it peak active mode, and you don't want to go down from there.
Save your money. Niacin 60mg is equivalent 1000mg NMN in raising NAD levels.

Hi Claire, can't give personalized advice. I can tell you those fat-soluble antioxidants - unless taken for a specific time for a targeted goal (e.g. 1-3 weeks) can turn off the natural defense system and reduce longevity - WHEN we're young.
I"m very careful with them.
Astaxanthin should work better after age 70 when we begin to oxidize our brains, eyes, and ears...all fatty tissues.
And from what I've seen, it can cross the blood-brain barrier.
Today, applied topically against sunshine should be rather safe - I think. Also maybe taking orally before a very sunny vacation and that's it.

Stanfield for all his emphasis on methodology and rigorous/accurate interpretation of data, all too often selectively cherrypicks research just for the sake of being contrarian.
His assertion that it is not even clear that activating sirtuins is an anti-aging mechanism based on 1 sirt-2 study is downright ridiculous.
To make such a statement while omitting literal piles of research estabilishing the crucial role of sirtuins (and specifically sirt-1) in aging of various tissues from bones to ovaries, is either ignorant (again, Stanfield is MD, not a real researcher) or malicious.
He has proven himself to be a somewhat sleazy opportunist and considering his considerable lack of expertise or even academic merit, his public persona is all the more inapproperiate.

Hi, I hope this comments finds you well. I’m a scuba diver- few months ago I started to take resveratrol , nmn, metformin and some other molecules mentioned by Mr Sinclair. For the first time during my dives (I’m a nitrox diver) I started to experience scary heart palpitations when I was below 20m - it usually would happen at 30-25m depth. At first I thought it was narcosis but then I decided to stop taking resveratrol (intuition) and my dives were going perfectly again. The same problem occurred when I used a hyperbaric chamber (I have it at my gym) - I experienced the same reaction as during diving and they had to take me out of the chamber. When I used it without resveratrol I was again- absolutely fine.
Does it make sense to you? I hope you can shine some light on this as it’s been really on my mind. This days I never take resveratrol when I’m on a diving trip or when I know I may use hypoberic. Tks for your time .

It sounds like something that causes your body to become more irritable under physical stress (Water, oxygen levels).
I don't have an explainable mechanism.
Possible explanations:
Allergy to resveraratrol
Allergy to compound in the capsule itself, including remains from the plant from which resveratrol was extracted.
Over activation of the stress system by resveratrol via an unknown mechanism.
Too much high dose.
Have you asked your doctor maybe?

Hi Rimon, I’m very well thank you. Yes , it wasn’t the best experience to get a major heart palpitations at 27m and trying to ascend as slowly as possible. I’m glad I worked out what it was tbh as diving is my passion. I do think it might have been a combination of high doze (1g) and high oxygen… I do breathe 30-33% O2 being on nitrox. I was even more certain it was resveratrol after it happened in hyperbaric. Anyway I do agree with your comment of taking it every other day- I’ve also decreased a dose to 0.5g. I noticed that 1g makes me anxious somehow but I feel fine with 0.5g.
Regarding my doctor- I’ve only recently had my first visit with functional medicine doctor but we didn’t have a chance to talk Abt this particular problem. I’m sure I will get a chance during my second app. Thank you for your time. Keep well.

I know my mom had CT scan and no dementia or Alzheimer’s but has some blood vessel disease. I know blood flow is important. I want to try NMN with mom and myself. She’s 40 pounds overweight but her spine is damaged,so she’s not able to exercise. She can go for small walks but her spine and neck are really painful. Just a wild guess Edyta, resveratrol increased total cholesterol significantly, from the *No beneficial effects of resveratrol on methabolic syndrom* study Rimon forgot to mention at 1:25 (from Dr. Brad's video around the 8th min. mark). In higher pressurized environments, serum-cholesterol may clot arteries more easily which could cause palpitations... I bet you'd not be the only one showing those symptoms under those conditions.
There is possibly a link between cycling creatine and Resveratrol. One creates cellular on a small scale promoting recovery demands and the other providing the energy needed to meet those demands. Research would be needed but this is possibly a compass.
why would you not put that supplemental data out front. this does not pass the smell test.

Do you have a reference for the paper David Sinclair said showed regular diet fed mice had benefit when resveratrol was given on alternating days? I've looked at some of his papers but could not find that data in the supplemental material.

Hi David! I remember seeing that in 2019 when I delved into this for my own supplementation.
Now, I went intuitively to the original study, and you're right - No mention there in the supplemental data.
But it says in the paper: "Analyses of the other groups will be presented at a later date."
The standard diet + resveratrol on and off had to be "other groups".
I think that I've seen it in a subsequent paper, that I've seen that. Not easily dug, but you're right about the original paper here:

I've noticed that supplements seem to reduce my kidney function. So I've cut back to just good diet, daily exercise (alternating RT, Zone 2, HIIT and 10K steps) and occasional fasting. Maybe that's enough. I used to take resveratrol, but can't imagine taking it now.

there is no controversy, just a GP (Stanfield) that knows about as much molecular cell biology as my english school teacher. There is *some* discussion about the exact involvement of sirtuins, but the overwhelming evidence (and not just from the Sinclair lab) shows that they are absolutely involved in CR mediated lifespan increase. Obviously they are not the only pathway, but there is never just one pathway regulating "big" functions like cell senescene, so its not an either/or with respect to sirtuins and rapamycin. If anything, supplementing nutritionally, with resveratrol and NMN is far preferable than taking a drug like rapamycin. Also, if you read the paper showing the low level replicative stress carefully, they do discuss their results in the context of the plethora of evidence on resveratrol and lifespan extension, and they are not actually mutually exclusive because low level relicative stress is actually one of the putative mechanisms unerpining lifespan extension even accroding to Sinclair's layman explanation. OVerall the literature has an astounding amount of data linking caloric restriction, sirtuins and sirtuin activator compounds and the rest is various randos on the internet trying to catch a bit of the spotlight and make a name for themselves (like that stanfield guy) on youtube by throwing around provovative statements like "convtoversy about resveratrol", which is laughable. Thankfully real science doesn't care about them

I did study chemistry, biochemistry, metabolism (certified in Metabolic Typing), and statistics).

@Wellness Messiah with Rimon thank you for your response and that sounds like an impressive background. Your video is very informative and well presented. I am finding it so hard to parse information on ageing when the top ageing experts like Professor David Sinclair, Charles Brenner, Matt Kaeberlein and Brian Kennedy all disagree with each other and argue so vehemently about it on Twitter. It's honestly so bad for science communication and for lay people like me wanting to know if they should take a supplement and which molecule is better. I wish the ageing experts would come together for some sort of friendly joint debate or discussion.
My impression is that while there are some studies studies, there maybe aren't large-scale, strong human trials on supplements like NR, NMN, metformin, rapamycin, so forth? Or they are still underway? It's so confusing, honestly lol.
@Jessica _S I agree with you, it's very confusing. Remember that each lab researcher has seen different results. I think it's -- part of their identity.
Also, the answer many times is intuition.
Because each study gives a tiny, tiny cube of evidence (that may be incorrect), as well. Thus,
lab researchers, many have different intuitions about where we should go.
And it's very difficult to put intuition into a debate. You can only prove the cubes. Only your mind can see the big picture - hard to argue around that.
The results I have put were never done in a single study but followed by the intuition of how the human body works.
Metformin - many, many human studies. Been in use in humans for 60-70 years now.
NR& NMN - Most of the human studies are from NR because it has a patent. So there is money in it. It doesn't take anything from NMN
There is a lot of benefit in mice studies, our evolutionary cousins.

Did you not do a video where you suggested too much resveratrol was more beneficial to those who were sick (I.e. cancer patients) and less is more beneficial for healthy people? Why are you taking 1g, isn’t that higher dose? I thought you suggested between 250-500mg for healthy people?

The issue is that it could not be replicated. Ask the company that payed out millions to David.
That study about exercise was short term, people forget your body adapts to things over time. Why don't they run blood test on people who have taken resverstrol for years. Cause a lot of those people seem in great shape. It appears resverstrol didn't harm their exercise.

@ How aer you? If I got your point right (not sure), also plants have many other molecules and their levels of antioxidants are not as high as in supplements. They have additional benefits beyond the scope of youtube comments. So you have a point.

My point being is that these studies which indicate that antioxidants are bad have studied supplements.
However, when you study plant foods rich in antioxidants (Blueberries, Cacao, etc) you do not find the same results.
So, you cannot say antioxidants are bad, only that supplemental antioxidants are bad. Otherwise, this would a very reductionistic statement.
Plants contain thousands of phytochemicals, when ingested as a whole, they provide antioxidant effects but also other effects that have not been shown to be bad on exercise or on youth.
I’m not saying supplements are bad, I take more than 20 supplements. But I have noticed people claiming antioxidants are bad and they’re basing these claims on these studies which only studied supplements.
That’s like studying an anthocyanin supplement, getting a result, and claiming that antioxidants are bad. But antioxidants from blueberries aren’t bad. In fact, they’re good.
Plants contain hundreds of thousands of phytochemicals in a food matrix and this significantly changes how it acts on the body. You see it time and time again. Isolating one compound and studying it and seeing it doesn’t work. However, when you take plant itself, it works better.
Resveratrol might be an exception because of its low bioavailability and that it exists only in very small milligrams in plants.
