    How can i or what it depend get the exact equation to be used in hormonal analysis like testosterone ?

    This is a linear standard curve, y=mx+b. Many if not most ELISAs recommend analysis by 4PL using a log transformed sigmoidal curve. I would advise everyone to check their ELISA kit for recommendations on how to analyze their data before using a linear curve.

      You are absolutely right. There are softwares available to do this kind of analysis. You can choose the most appropriate type of equation for quantitation.

    thanks for the video,
    I need please to calculate the concentration of a medical substance given to horses and prohibited for human consumption.
    I used an Elisa test kit to determine the presence of this drug and measured the absorbance at 650 nm.
    can you help me on this point.

      You need a concentration standard curve to convert OD to concentration

    i have this question that what if you don't have a standard (in case of indirect ELSIA ) so then how to analyse the data without knwing the concentration of the standard?

      Do you mean customized ELISA? It needs a lot of characterization with known amount of protein. Otherwise you can only compare the relative OD reading.

      @@sunnyscience3384 ​ yes i have customised the Indirect ELISA by using antigens coatings on plate and performed it, and now im just wondering how can i analyse my data as the purpose of my ELISA was only to check if there's any antibodies generation in experimental animal, though im not intrested in the conc but its just a semi qualitative type ELISA just to get a "yes" or "no" answer. i just wonder if somone can help or if there is any such links to videos for analysing this type of data that i mentioned?
