When culturing microbes, it is critical that the media used meets the growth requirements of the target organism. For this reason, it is necessary to measure the pH of culture media during preparation to ensure that the media will perform as intended.
Most microbes grow best at a neutral or slightly alkaline pH. For this reason, it's no coincidence that the common general growth medium Tryptic Soy Agar is prepared with a pH specification of 7.1 to 7.5.
