    Hi Arpan! want to say a big THANK YOU for making these videos! help me to understand a lot!! I am self-studying bio, your animation makes the concepts very easy to understand! Am giving every video I watched a big thumb.
    Question for video "plasmid copy number regulation": what would lead to a higher number of plasmid? If I am understanding right, both with or without ROP1, RNA I and RNA II tend to bind and lessen the number of plasmids, right? So what would lead to a higher number of plasmid? In a situation when RNA I not replicating?

      High copy number plasmids have mutation in their Ori which leads to release the brake and they can increase in number. some plasmids have a mutation in RNAi sequence or lack of ROP protein thus preventing the RNAi-RNAii complex formation. hence these plasmids will be in high copy number. First of its kind is developed in University of California (UC) and the plasmids are named pUC. Some plasmids, such as the pUC series and derivatives, have mutations in Ori itself which allow them to reach very high copy numbers within the bacterial cell. Plasmids based on pBR322 and many cosmids are generally maintained at lower copy numbers. Very large plasmids are often maintained at very low copy numbers per cell while small plasmids tend to have higher copy number.after all the insert in the plasmid sometimes influence the copy number but it’s not clear how.

    Why in F-plasmid having 1-2 copies/cell but Col plasmids are having 15-20 copies/cell .

    sir the plasmid in category of high copy number also have both region of ori ,rna1 and rop1 but how are they getting high copies why not rna1 and rop1 is binding rna2 in them?

      You do mutation in genes for rop and sequences for RNA I and II to get more copies of plasmid

    Is replication regulated in host cells also? Like if u get a dna vaccine will the plasmid self-replicate in your human cells?

      Usually dna vaccines wont work in high eukaryote cells, if they even manage to enter tru electroporation or particle bombardement they will likely be cut into pieces and destroyed before expressing the protein. It also needs the nucleus proteins for transcription (no way it can get in there). If it is a bacteria it will regulate itself I think (using its own encoded proteins or both its own and host proteins that work together)

      With nucleus protein I mean that all transcription proteins are inside the nucleus and nowhere else
