Hello sir, is it necessary to do with EC MUG broth in complete phase?
For the presumptive phase we used lactose broth, for comfirmed phase we used EC broth and for complete phase we were told to used EC Mug Broth .

Sir please guide us why we use double stranded or single stranded media In presumptive phase?

This is because this broth media contain lactose as carbon source and only coliform bacteria are able to metabolize lactose sugar
