    Hi your explanation is good. Just a concern is, in the 0.33 time, you tell that there is binding of proteins in gel filtration column which is wrong

      I think you got me wrong @ .33 sec I was talking about a general workflow of column chromatography and later pointed out that GFC is bit different than the general workflow ,it involves seiving mechanism in contrast to binding

    can the elution volume be greater than the total volume of the column?

    Suppose if exclusion range is 5 to 600 then the molecules whose size is below 5 they also trapped between the voids then these molecules also eluted with small size molecules then our reqired size protein will not be obtained is this happen or not sir?????

      If the peptide fragment is previously tagged by his tag / HA tag then one can do affinity chromatography, if slight estimation of size is known then one can do gel filtration, if amino acid composition is known and it has charged residues then one can go for ion exchange as well. So it really depends on what we know about the polypeptide.
