    Hello, I just stumbled upon your channel. You did very well at explaining this topic, I didn't understand a single thing until I saw your video! Oh yeah, I want to ask a question, what if we had 3 types or more of restriction site/enzyme? Such as EcoRI, BamHI, HindIII and so on, how would we construct the map? Thank you in advance!

    About the size of dna fragments ???
    My question is in circular DNA is there any differences in the ladder ? Like it will be in descending order??? Or as same as in linear dna ?

    i am a little bit confused on how you know they were not overlapping. is it because they both show up in the combined well 5?

      @@khal2652 I think it is because if you notice that both the 200 and 250 fragment stay in tact on the plasmid--you could not have overlap. Even if you placed the 200 within the 250 fragment that would actually result in a fragment of 200 and a fragment of 50 base pairs--the 250 base pair would not exist. I hope this helps/is accurate! I was trying to think about the answer to that myself.
