Defects in DNA repair& Cancer
DNA damage is ubiquitous
DNA damage is not the same as mutation though it can lead to mutation
DNA damage is a chemical alteration ( can be corrected)
Mutation is a change in the sequence of base pairs ( cannot be corrected )
If any kind of DNA damage is likely to lead to a mutation, we call it genotoxic
There are Mechanisms to repair the types of DNA damage
The Effective DNA repair is the backbone of cancer free survival
Genome instability is an Important hallmark of cancer
INSTABILITY: could be point mutations, trinucleotide repeat expansions and contractions, gene duplications, deletions, and inversions, to large-scale chromosome changes including translocations and whole chromosome gains and losses.
DNA repair systems play critical roles in genome stabilization
Defects in DNA repair pathways contribute to specific instabilities
Erroneous DNA repair leads to mutations or chromosomal aberrations
The Genes for DNA repair
Individuals with inherited defects in DNA repair genes are at increased risk of developing cancer.
Base Excision Pathway (BER):MDB4,Colorectal Ovary Multiple myeloma
NER Pathway:XPC, Bladder Skin Cancer
Direct reversal of Damage:MGMT,Colon Gastric Glioblastoma
Mismatch excision repair (MMR):MSH 6Colorectal
Non Homologous End joining: XRCC5, lung
Homologous recombination: BRCA 1,Breast Ovary Gastric

Thank you so much . As an oncology resident i come n see your videos to understand the roles of genes in various cancers.
ur vdos r simple n explanatory...it made my understanding of hallmarks of cancer easier..thanx alot..i wish that more n more students get benifited by ur vdos..looking fwd to c more vdos from side sir..
very useful to me and helping very much to understand clearly the topic
detailed explanation ,thank you sir
Nice explanation.Thanks
Superb. Absolutely fantastic
Very helpful !!! 👍
Great teaching and illustrations thank you for teaching and sharing ❤❤❤❤❤
i have a very tiny small correction to make. at 5'48, in your graphic it is MBD4 (instead of MDB4) for the BER pathway.
for the rest i highly recommand the serie on neoplasia
thanks for the videos, it's a lot of work.
Sir you're a life savoir ❤

Can I have this lecture as PDF?
Extremely Helpful Video
Can I please know the Textbook referred by Sir?
very nice video sir. Clear and to the point concept.
is this Dr Vijay Shankar Sir delivering lecture??
Vishal Parekar ... Yes :-)
Kindly make video on carcinogenesis and carcinogens sir
Nice sir
Video comes in handy with this covid virus running rampant.
look like it is better let it age and die than repair
This accent kills me (
