Km is an indicator of binding capacity. It can be defined as the concentration of substrate that is required to achieve half of maximum velocity. So higher the Km value lower the affinity for the substrate towards the enzyme.
Sir in the bigining it is told that rate of reaction depend on concentration of both substrate and product but in the equation we only considered the concentration of substrate ...is there any reason for this ...sir Also in esic 2016 paper there was a question that km value indicate affinity Can u please explain it
Rate depends on concertation of substrate and concertation of enzyme. In rate equation, concentration of substrate is included which is again related with enzyme concentration. Km value is an indicator for enzyme-substrate interaction which indicates relative affinity of the substrate towards enzyme. Thanks for your interest.
All these are inter-related. Normally rate depends on concentration of reactants if it is an irreversible reaction. But in case of enzymes rate is mainly influenced primarily by substrate and enzyme initially but later as the product accumulates , the rate may be reduced. Even product may produce a negative feed back inhibition on enzyme. So concentration of product, which depends on conc of substrate and enzyme initially, also acts as one factor that may influence the rate. Hope it is clear now.
