No!!! Km measures enzyme affinity for a substrate, remember Km is half Vmax. Meaning if an enzyme has high Km, it means an enzyme requires high concretration of a substrate to reach Vmax. The increase in substrate has no effect on Vmax, as the Vmax remains the same
Why do enzymes or protein get saturated? Saturation: What Does That Mean? Does that imply fatigue? Is it comparable to a man who works extremely hard all day being exhausted (saturated) and losing all energy? please shed some light on this comment
Saturation means that all the available receptors are busy, and therefore can’t bind more substrate (think of it as a call center where all the customer service representative are busy with other clients, i.e. saturated).

The actual meaning of Km depends on specific aspects of the reaction mechanism, such as the number and relative rates of the individual steps. When k2 is rate-limiting, k2 ≪ k−1, and Km reduces to k−1/k1, which is defined as the dissociation constant, Kd, of the ES complex. Where these conditions hold, Km does represent a measure of the affinity of the enzyme for its substrate in the ES complex. However, this scenario does not apply for most enzymes" As I understand it, Km represents affinity only when k2 is wery low, and Km=Kd. I just can't understand why is it so. Also, I googled it and mostly everyone says that Km is an indicator of affinity. I hope you'll have a look at this, maybe you will be able to put it in simple words, as you do.
