Removing Spontaneous Differentiation from iPSC Cultures

  • In this video, we are going to revise the protocol to manually remove regions of spontaneous differentiation that can be found in induced pluripotent stem cell (iPSC) cultures. iPSCs are prone to spontaneously differentiating, therefore, daily checks and removal of any spontaneously differentiated regions are required to maintain a healthy iPSC culture.
    List of materials:
    - Phase-contrast microscope
    - A permanent marker
    - 200 µL sterile pipette tips or a cell scraper
    - iPSC media (mTeSR, mTeSR plus or E8) at room temperature
    - 5 mL serological sterile pipette
