Seed Viability Test or TZ test

  • Seed viability test is very important for crops producers, seed producers, seed sellers, seed store keepers prior to proceed for final seed germination. Tetrazolium (TZ) test the most common and vastly used method for seed viability analysis. In this video I have shown step-wise; from preparation of TZ solution, seed dissection and placing the TZ solution soaked seeds into the water-bath and final color absorption by seeds.
    Minor mistake: In my speech mistakenly I used "ovule" but it might be "embryo", sorry for that mistake.

    How much time the seeds put in water bath and which type of water bath you are using

      It depends on seed types; can be 2 hours to 24 hours. We used Memmert brand Water Bath.

    How would you increase TZ before testing keeping seed cold ?

      You can prepare TZ solution of different concentrations (0.5%, 1% or 5%) as your own choice and can preserve in freezer for long time covering the vials with aluminium foil (or others); as the TZ is sensitive to direct light.

      Very simple, higher concentration. If you can afford better to use higher concentration for quick results

    Does the tetrazolium only react in the dark?

      Dark is not mandatory (but better) while keeping into 35 to 40 degree C temp. warm water in mandatory for several hours. As we normally use water bath and when we close the cover normally there's create a dark condition.
