    A lot of misinformed comments here. Sirolimus’ primary pharmacological effect when taken orally is immunosuppression, which is why there’s an increased risk of cancer in people who may be predisposed to developing cancer or live unhealthy lifestyles. Transplant patients typically receive much higher doses. Applied topically at a low dose, systemic absorption should be low or negligible. At most, you may experience dry skin or a rash, but consult your doctor if you’re considering taking it and you’re not confident enough to make the choice on your own.

    I am a heart transplant recipient.
    I need to take Sirolimus daily to stay alive.
    It’s a medicine which has many terrible side effects and the idea that somebody could just willingly take this for vanity’s sake blows my mind.

      @@--RBuo84not just like I ozempic. You would never tell a depressed person they shouldn’t use antidepressants to improve their condition. Obesity is a serious health problem and has many many negative impacts on patient’s life quality. You have no idea what you’re talking about.

      @@jayg6138that’s not addressing or investigating for the root cause of the obesity, though. And when medication helps their symptoms so effectively, people tend to rely on that medication and just stay on it and not look for or find the root cause.
      And it IS a huge problem considering Ozempic was in such short supply after so many people started taking Ozempic to fix their weight and there were so many people with diabetes who literally need it to survive and they couldn’t even get it.
      It’s fine if there’s more than enough for the people who have diabetes AND people with obesity to take, but not at the cost of the lives of people with diabetes and their accessibility to it.
      And it would be fine if most people were just going to use it temporarily whilst looking for the underlying cause of their weight issues, but a lot of them aren’t - because it works, so they don’t see the need to.

      I imagine that the topical has less systemic side effects than oral even though as he said the evidence is on oral not topical. But the problem is that as with all immunosuppressants it may increase the cancer rate??

      @@jayg6138 Congratulations, you did not understand my comment. But I am happy that you are belittling my remark, based on your highly qualified interpretation skills. I meant that Ozempic is abused by people that do not need it - that's why people that depend on it, can not get it. If Sirolimus is used by people for beauty purpose, they also do not depend on it. I never said people that need ozempic should not use it or it is bad per se. But it has side effects. Ancient Romans and Greeks were aware of some of the health hazards related to lead exposure, as documented by historical texts describing lead poisoning symptoms among workers in lead mines and individuals who used lead-containing products. Despite this knowledge, the full extent of the dangers of lead exposure, especially in cosmetics, was not fully understood or widely recognized until more recent centuries with advancements in scientific research and toxicology. And I would be brave to guess, that humanity is not at the end of pharmaceutical knowledge.

      I was curious too, so it took me just over 10 seconds to find out this is actually rapamycin (first isolated in the sixties): "Sirolimus is the USAN-assigned generic name for the natural product rapamycin". It's also FDA-approved for certain organ transplant patients since 1999. There are a lot of ongoing studies for various other outcomes, it's quite fascinating. Once some of those start coming out, you should be able to start seeing this available as a supplement in some countries where it's not controlled by the FDA.

      Here in Canada ,it goes under different name.this info from patient information leaflet:
      Serious Warnings and Precautions
       You will be prescribed Rapamune by a healthcare professional experienced in using
      immunosuppressive drugs and in the management of organ transplant patients. Your treatment
      will be managed in a medical centre that had access to the appropriate staff, laboratory facilities
      and supportive medical resources. The healthcare professional in charge of your maintenance
      treatment will be in direct contact with your transplant centre.
       Rapamune is not indicated for use in liver or lung transplant patients.
       Immune system effects:
      o Rapamune may reduce your body's ability to fight infections.
      o Patients taking immunosuppressant drugs, like Rapamune, are at risk of developing cancer
      of the lymphoid tissues (called lymphoma) and skin.
       Severe allergic reactions: Cases of severe allergic reaction, including skin reactions.

    I took oral Sirolimus (rapamycin) for a few months. It made me feel so amped up and gave me pretty severe acne, although I am homoronally acne prone even at 36. Bryan Johnson, the millionaire biohacker, also takes this and looks noticeably youthful. I did notice some glowyness and skin rejuvenation while taking it but given my general sensitivity and acne, I decided to stop taking it. Looking forward to trying the topical version!

      I also took it for a year and a half, it gave me acne and left marks😢 (in my adolescence I had acne but it never left marks). Plus I suddenly developed a lot of allergies.

    I think Tazarotene is better than Tretinoin too. I got results much faster after making the switch.

      Yes. I’ve been using tazarotene for 3yrs. It made my skin feel smooth right away, helps control my hormonal acne & reduced old acne spots I had on my face. I didn’t get any irritation except some slight flakiness.

    I really appreciated you sharing this! I've found a bunch of 'anecdotal evidence'. Interesting. Promising. think a lot of people confuse the oral treatment with the topical cream, which for anti aging as far as i have read, much weaker, than the topical used for the treatment of medical skin issues.

      The description definitely did not mention the many side effects of oral sirolimus. This is only a topical version. So it's possible that the side effects of immuno suppression and cell damage that is caused by internally taken sirolimus is not the same. Just like in retinoids. Dry skin, irritation, acne, contact, dermatitis, bruising, photosensitivity, acneiform eruption happens with tropical sirolimus. The oral version causes brain damage, seizures, difficulty breathing, hypersensitivity, kidney failure etc.

      once a doctor and.an old lady with few spots of vitiligo told me to be very careful with the way I processed emotions.
      The old lady told me that her vitiligo stopped when she decided to be happy, and keep happy thoughts. Off course each person should find their own peace of mind as they can.
      And the doctor, the first question she asked me was if I was suffering too much emotionally or bearing too much stress.
      I could not tell you if it is truth, but, I only got one despigmentation on my face and it stopped. Also, that moment when my skin lost the color occurred while I was on an activity of forgiveness
      it's a long story anyway

    A quick search about this online says there are side effects to skin.. Melanoma, squamous cell carcinoma, basal cell carcinoma. What do you make of this, especially if the individual has exisiting solar keratosis for example?

    I am very disappointed with the use of your words, especially in the description and the title. Calling it a best well kept secret. It is not. It is a molecule that has very few studies behind it for anti-aging and for normal skin. A person is not a cutting edge dermatologist for using it. A dermatologist who uses for anti-aging is someone who likes to be new and different and follow upcoming trends. I've seen it in almost every social media dermatologist. Many claims about different chemicals, calling them holy Grail or secrets to anti-aging, and then when chemical comes out, suddenly the old one seems to be useless.

    As soon as I heard mTOR pathway immediately thought of rapamycin and lo and behold it is it seen in the show notes. Peter Attia is a big proponent of it as well.

    I am not sure that you thought this through. You are advocating topical rapamycin here. The side effects can be VERY serious. Including infertility, heart failure... it is not safe during pregnancy is available only via prescription.. Various side effects should be reported immediately. Yes.... this could be a very interesting product to watch when necessary research has been done. This video is bordering on dangerous.

      I didn't watch his video for the reasons he is not what he present himself on RUclips. He is another fake, money greedy, ripping his patients off badly and he is heartless also.
      It depends what cells he's talking; i.e skeletal cells regenerate about every 10 years, skin cells about every 27 days. Dead cell - senescence cells - build up is one of the causes of old age related problems. We lose millions of millions of cells everyday. And the ones copying is i.e. Telomeres shortens as we age. Hence, not a good copy of the one before.

      Some cells don't turn over at all, like cells in the lens or your eye or neurons. Some cells turn over very often like blood cells or skin cells and some turn over less often like the cells of our internal organs.

    Here in Germany you don’t get Sirilimus without having a strong illness what some people still wrote before. For a face cream it is the same, it is here on what we call the red list, means only from a doctor and only for certain illnesses…

      No, they are two different kinds of macrolides.

      nope different mechanism of action but both can be used to treat post transplants patients when taken orally. Tacrolimus inhibits calcineurin whilst sirolimus inhibitors mTOR. Tacrolimus also comes in a cream form and is used for eczema so its similar with sirolimus in terms they are both used to treat transplants when taking via orally, and they can also both be used topically in creams to treat various conditions. If you're using the cream for anti aging benefits, sirolimus would be your best bet as its one of the most promising anti aging medications tested with the most profound results (atleast in various animal models) human trials are more complicated as we live a very long time. (Im a pharmacist).

      I used to work at the company developing sirolimus for organ transplant rejection. I actually worked with the man who discovered it. This was over 25 years ago. He was such a kind, soft-spoken man as well as a brilliant scientist.

    I have found the best serum is 10% niacinimide (B3) after trying them all.
    This works best on dark circles under eyes & works faster than any under
    eye cream for this purpose and all dark spots.

      Absolutely not. This would be too harsh for your skin. Stick to Trentnoin if your skin tolerates it. If not, then use retiNAL (1 step down from Trenton)

      Retinol is way weaker than tretinoin, adapalene and tazarotene. Tazoratene will give u even faster results than tretinoin and adapalene, but in the long run they will give the same anti aging results for wrinkles, more even skin and melasma (dark spots)

    Interesting I think what I haven’t liked about tret & even medik8 is that it’s not only thins the skin but it give you red skin when it’s something I have never had before,I’m also wondering if it contributed to a few spider veins 😮I have never heard of this so will investigate more.Thanks de Lim 🇬🇧

    I've been interested in this for some time, the problem is I've been unable to find it. I can find the oral form, but not the topical.
