Thanks! On bacteria - would be nice to know effect on gut bacteria. Also, metrics on various inflammatory markers, insulin, etc. If the major effect is on mTORC1, we know that fasting likely drops mTOR expression due to dramatic drop in amino acids and glucose stimulation, so wonder if a weekly fasting regime, say or 1 or 2 days at very low calories like Dr. Longo's FMD would have a similar effect. Maybe, 2 days a week for 12 weeks as a guess. In any case, having a road map of changes such as common blood markers and gene expression changes would allow for comparing different interventions. I will likely try rapa at some point for a few months, and if I do will track blood markers.

Is anyone using Rapamycin for periodontal health (toothpaste, rinse etc)? I am experimenting with 5 mg / week internally and would like to expand the experiment to dental health.
Rapamycin is a potent and selective inhibitor of the mechanistic target of rapamycin (mTOR) protein kinase, which acts as a central integrator of nutrient signaling pathways. mTor is a key regulator in maintaining skeletal muscle mass. Rapamycin is an inhibitor of mTOR and mTOR is critical to signaling muscle protein synthesis. Why would rapamycin be used as an anti-aging agent that inhibits muscle protein synthesis? Would resistance muscle training to maintain muscle mass be impaired and cause sarcopenis muscle atrophy?
I had frozen shoulder. Got a steroid injection after trying physical therapy (which didn't work) and it went away after the steroids. I think I'll try rapamycin because my shoulder feels like it might be starting again.
That’s an interesting question. I don’t think anyone has a trust this question in a research setting. Since fasting, like rapamycin, inhibit mTOR, there could be a synergy effect in combining the two. The question is whether this effect is desired, and for which people. As Dr. Kaeberlein pointed out in his interview, it’s me be best to combine rapamycin with exercise to counteract potential negative effects in muscle mass. Combining rapamycin with prolonged fasting, might do the opposite end, hence, be not indicated for some people.
I take akg because of a rotator cuff tear. I have had no incidents from akg. The first dose of vitamin k2 + d3 was an emergency room visit but with no after effects. I became very ill going to the restroom at 1:30 am after using our stairs at home. The emergency doc said it was my vegas nerve. After a few hours everything became normal again and I continued with the k2 vitamin. No further effects were observed. I will take berberine when I can afford to get it. I am trying to be keytonic with some success.❤
Rapamycin is an immunosuppressive medication.. of course it will work for inflammatory condition like frozen shoulder. There are many strong anti-inflammatory medications that can be offered and I reckon you would experienced similar benefit. The question is whether the anti-aging aspect of the medication is what helped the shoulder which I doubt happened over a few weeks course.
Many including me take nmn, Berberine, spermedine, fisetin to induce autophagy.. But it can take several weeks to see the effect.. Meanwhile one should train with muscle building and maintenance otherwise you start looking very spiky.

Keep in mind. An increase in mTor activity is not always bad. For example, this is what mediates muscle growth after exercise. However, several undesirable factors can contribute to the increase in mTOR activity with aging. In this context, an increase in mTOR activity can be viewed as a biomarkers of these undesirable factors, rather than a driving cause of aging. One of these factors is the accumulation of cellular damage over time. This damage can activate mTOR signaling pathways, leading to increased protein synthesis and cell growth. Another factor is changes in nutrient sensing and metabolism that occur with aging. As we age, our cells become less efficient at metabolizing nutrients, and this can lead to changes in mTOR activity. For example, insulin resistance, which is common in aging, can activate mTOR signaling and increase protein synthesis. Additionally, chronic inflammation is another factor that can increase mTOR activity with aging. Inflammation can lead to the activation of mTOR signaling pathways, leading to increased protein synthesis and cell growth. I hope this helps!
I hope more studies are one on Rapamycin as quickly as possible so we can figure out how good or bad it is. It seems to be very promising.

I believe Dr. Kaeberlein mentioned that he is planning some studies on rapamycin for periodontal health. Stay tuned ;)
Month sore, yes I concur, happens once at the beginning. 5mg/week.
Yup, I got 2. One at corner of mouth and big one at back of throat. 2 weeks in, 6mg week.

Interesting... I'm 🤔 curious, too
Thank you. I’m very curious about Rapamycin..And exactly what it can help with in terms of regeneration.
Isn't there supplements that mimic Rapamycin? I think I read that somewhere. Can anyone tell me what supplements those are??? Thank you.
Probably cost an arm and a leg😢

Try free hangs from chin up bar. Never had a shoulder issue since starting that after workouts.
I tried to get a prescription. My doctor refused to sign for it.
Raised my BP from 130/70 to 154/90 in 4 months. Anyone else have this happen?
Nope, but I am whole plant vegan for 22 years at 53 with perfect blood work and high normal testosterone.

I’d like to use it for the periodontal disease my dentist want me to have surgery on my whole mouth and my insurance won’t cover most of it

It’s expensive …1 5mg dose is about $100
Not if you buy generic brands from India. It's less than a dollar per 1 mg. I'm UK so it's my only option!
It costs $1.

not understanding the rationale for 8mg/week dosing. Mice were given 8mg/kg/day, which would translate into GRAMS for humans. Not getting it
We don't care if mice die. That is why experiments are done on mice.
It’s not a direct 1:1 ratio for rodent dosing vs human dosing. Don’t remember it off the top of my head, but there is a formula to calculate it
Indeed, from what I'm seeing there's a factor of 12 involved.
