The thing is she has a point. When you remember all the things the top people in any walk of life have given up to get where they are. Sports stars and top musicians, the hours they put into practice (especially when they're young); leaders in business, the hours they devoted to building their companies... if you want to be a success you have to be willing to sacrifice *a lot*. But then, when you achieve that success, you'll be able to do all those things that others can't because "Wednesday night is Halo night."
If you won't be happy while on the way to success, you won't be happy when you get it. It's always said it's the journey that counts, not the end. Yes, sport stars and musicians practice. But they also get downtime. Too much work WILL drive you mad and you won't be happy even if your work is your dream. Balance is key which Sheldon has handled pretty well. He works his ass off when needed and cancels those Halo nights and stuff. But he's working just fine even with downtime. You don't have to give up who you are to get where you want. Do that and you'll die with too much regret. Better focus on enjoying the now that bust your ass to a future you can't even guarantee.
Ramona actually cares about Sheldon. She's pushing him hard here, but it's for his benefit and according to his own principles. Sheldon is literally a man who has built his entire personality on being a genius, and Ramona encourages him to pursue his excellence. For normal people, this would be too much pressure, but Sheldon isn't a normal person.
As a former PhD student in a STEM field, the funniest and really unrealistic part was the way she bossed around Sheldon, a fulltime researcher in the department. No PhD student would be dumb enough to do that, bossing around a researcher/professor in the department would invite the wrath of your PhD advisor and the department chair.
He really should have given her the credit. She held him back without any fun but she did cooperate and he wouldn't have solved it without her. As he said himself.
