    Correction and clarification: At time point 1:07:37, I said I take 1 gram of Spermidine, but the active ingredient in the capsules is 1-2 milligrams. I apologize for the confusion.

    The co host is great. He has a very effective way to be both a “layman” as he asks questions while at the same time, generally savvy on the topics. Also, I congratulate you both on not taking yourselves too seriously. We already get enough of that with certain scientists.

      My uncle was more educated than this man, and in the same field, and he died in his late 50s from cancer.
      I promise you none of this stuff has any real effect on your longevity.
      Live a healthy life, don't go taking a bunch of pills and potions and spending a load of money.

    I just finished your incredible book Lifespan-for the third time. First reading was an audio version and I stopped every hour to cry hard. I read a lot for a leadership program that I teach for the Air Force and thank you btw for mentioning USA air supremacy.
    LITERALLY THE MOST INCREDIBLE INFORMATION!!!! I tried to relay it all to my own adult professional child and her husband and sounded like a blithering idiot bc I get so emotional. I am thrilled to find this podcast so I do not have to try.
    Your optimism is a leadership trait; you’re an educator, not merely a scientist. Your book’s messages were mostly about longevity, but also part existential, part environmental, and part economical. As a teacher myself a huge obstacle is keeping my students and coachees happy about humanity. I am as wound up at the end of your book as I am at the beginning.
    Thank you for your genius and your optimism.

    I have been taking resveratrol for several years now. Unfortunately I am confused. Many evidences suggest on the contrary. Why doesn't Dr. Sinclare clearly answer the question raised by Dr. Brad Stanfield. I think there are many worried people around the world who have been taking resveratrol waiting for Dr. Sinclair's response. Please!

    Love this series of episodes. Very informative and hopeful for those of us in their 70s. Vanity would have driven my interest before, now it is quality of life. Especially cognitive function as I'm watching friends in their late 70s and 80s, mentally declining. Not all but so many.

    I am a physicist by education. I used to think the biology was an easy subject. Not any more. Listening to this very interesting podcast, I marvel at the complexity of biological systems. This podcast is very informative and easy to understand. I plan to use some of the supplements outlined in this podcast. Excellent

      Wow exactly my thoughts. Physics and mathematics is very simple compared to biology. I came to the conclusion biology plays with physics the way we play with math, which makes it very difficult to predict and understand

      @@creativesource3514 perhaps. But it is an elementary matter. I can explain to you the orbits of planets, their gravitational pull, their magnetic sphere. I can plot trajectories across the stars, how the stara form, hell i can make a star here on earth. I can tell you the age of the universe.
      In the same breath I cannot tell you in a fundamental level how a cell transcribes genes, I cannot tell you how the proteins are transported and constructed, I cannot tell you how a protein is folded, I cannot tell you how a cell structures it's self, I cannot tell you how all this produces consciousness, I cannot explain the quantum mechanical effects of biology and how cells utilize it.
      My point isn't that we don't know per se, as there are things we don't know about physics. But my point is more so to showcase that to understand how these biology systems fully function is nearly impossible due to the complexity. I can breakdown the solar system and its functions and interactions much more easily then I can explain a drop of blood.

    I had heard of NMN on another podcast about a year ago.
    I then dud a little research and started taking NMN. I found out it that it has changed my life for the better. My anxiety had pretty much gone away. My energy is by far more focused, and for the most part, I feel great.
    I have other heath issues that dragged me down prio to NMN. I've tried many supplements and haven't had any luck, but NMN is doing wonders for me.
    I have bought a year supply since places have quit selling the product.

    hello Dr. SInclair, are there any studies and recommendations especially for women regarding hormones, menopause and intake of bioidentical hormons or herbs, or supplements etc. and prevention of diseases related to decline of hormones. I would like to learn more especially about womens health (so rare in the school medicin). Thank you.

    Second comment: I want to add that I am following much of your protocol and I have so much energy it is incredible! I am relatively young (33), but have been dealing with fatigue for a long time. Particularly the day after intense exercise (BJJ). I have trouble focusing and getting through the day even with heavily optimized sleep. I am currently alternating between 16 and 22 hour fasts. When I do eat, it is mostly a low protein, high carb, plant based food (with some junk thrown in because I'm human and still working on things). I am taking NMN, resveratrol and Metformin. And I feel like a million bucks! I am recovering well, focused, creativity is up. I am honestly shocked and hope this is able to sustain itself for the next few months (I do not plan on doing all of this indefinitely).

    My biggest takeaway: the conversation about mice being predisposed to cancer (certain type(?)) and NMN stimulating cancer growth (as in the 2019 study that was discussed). It feels as though that wasn't given enough emphasis, should I be cancelling my order for NMN?

      It stimulates it because if the body inside is younger cancer would also act as if it is in younger body, but then again, if you don't take nmn you will get older faster and chances to get cancer and other increse...

      GlaxoSmithKline has to entirely drop all research and studies based off the company bought from Sinclair, as the results were not only bad, but in fact produced negative outcomes.

      Taking a supplement or injecting young plasma in a very old body would likely lead to rejuvenation in some tissues, but would also lead to rescue of some very damaged cells and since the all tissues and cells are not rejuvenated to the same extent can lead to undesirable outcomes such as cancer. For e.g. suppose a supplement rejuvenates the liver and does not rejuvenate the immune system to the same extent, some liver cells on the brink of senescence would be rescued, but with mutations or damaged DNA and the dysfunctional and aged immune system would not be able to eliminate them

    Can you elaborate more on metformin and berberine for (aging) athletes? What is happening with glucose? Is the total amount of glucose storage in the body reduced? Are we excreting it? Or do the molecules just help glucose more effectively get into tissue? So let's say I have a big ride the next day and I'm trying to tank up on carbohydrates. What's happening if I take one of these molecules with my high carb dinner? THANK YOU!!! LOVE YOUR WORK!!!

    Resveratrol, 1gram per day, in the morning with yogurt or olive oil,basil, and vinegar (needs fat to be absorbed)
    Nicotinamide mononucleotide(NMN), 1-2 grams, in the morning with same yogurt/olive oil
    Quercetin and fisetin, 500 mg each, once per day, in the morning
    Spermidine, 1-2 mg per day, in the morning
    Metformin (prescription drug), 800 mg, in the evening

    I am marco from Italy, I have already read your book and I have found it fantastic! Naturally all your suggestion are focused on US market, I hope one day you'll have also some suggestion about italian or european provider for blood rest, dna test etc. Supplement are quite easy to find though.

    Hi David - I understand Metformin is a chemical derivative of Goat's Rue (Galega officinalis), which you can buy as a herbal supplement. Would this be as effective as Metformin?

    NR consistently raised my alt and ast liver enzymes during a year's worth of regular blood tests. NMN lowered my enzymes even lower than they were before taking NR in the first place.

      Hi Tim :) given what Dr Sinclair said about Pancreatic Cancer and NMN, I’m wondering if you screened for that genetic propensity and if so what test you used? It’s my only concern regarding starting that supplement. Thanks!

      @@larnapike6992 under my 23 and me account, I don't see anything that would indicate a propensity for that or any other form of cancer. There are others who've had this issue with NR, since NR requires additional methyl groups in order to be converted into a usable form. To get these methyl groups, your body has to borrow them from other molecules in the body, taxing the liver more. Which by the way, I'm discussing the liver, not the pancreas. As far as my pancreas goes, all of those health numbers, such as A1C are optimal.

      @@AmeliaLi-lz6kc yes, I stocked up a year or so ago when the FDA announced they were going to take it off the shelf for over the counter and rewarded a pharmaceutical company with the license to sell it as a drug

    Thank you for all your hard work Dr. Sinclair. I listened to your channel and then went out and bought resveratrol and yogurt. I've been taking it for a few days now (along with NMN and metformin) and I'm feeling great. But then I get a video on my feed from Dr Brad Stanfield saying that resveratrol is harmful to humans. Now I'm confused. Still taking the resveratrol with my yogurt, but I'm a little worried. Will my daily metformin dosages overcome any issues with resveratrol?

    Hi David! I’ve been implementing what you suggest (I haven’t yet implemented supplements yet) and I feel a lot better! One of the things I’ve done that’s helped a lot is actually something you haven’t mentioned in the podcast yet. I took a food sensitivity test and cut out the foods that tested positive for sensitivities. Milk was apparently causing me muscle and joint inflammation as well as stomach problems.

    As a person without any medical background I really wonder how to check my body response to the supplements recommended in this podcast🤷🏻‍♀️. I would like to know what blood parameters/blood tests I should check before taking the supplements and when to check out the results by retesting.
    My kind wish is to have the professor’s opinion whether the recommended supplements are safe for people with celiac disease?
    Could you also recommend supplements for people with serious problems with knees (deficiency of collagen and how to reverse that ?)

    Now when you know such a good info about the supplements what is the rule of thumb to pick the quality ones? I mean there is NMN sold on Aliexpress for third of a price, there are comments of producers using GMO in production of those supplements etc etc How to choose?

    I've been taking metformin for 6 months. I recently bought a red light/Near Infrared light. As I understand this video, metformin reduces mitochondrial energy whereas the red/NIR light increases mitochondrial energy. Is there a conflict with using both, e.g. daily metformin and every other day light therapy?

    Great podcast and a lot to learn. I live in europe and looked up what your whole supplemention would cost. For this I have exchanged metformin with berberine because you cant buy it. And im not sure if every supplier is 100% thrustworthy. That would be the equivalent of 250 dollars per person per month. Without Inside Tracker / Levels or vitamin supplement.
    Is this really affordable for the everyday person in the US?

    Important to emphasize ‘the threshold level’ as David discussed in his interview with Rhonda Patrick PhD , the ‘natural clearing process ‘ of the body that will eliminate NMN supplements , Resveratrol from the body - and David’s idea of ‘pulsing ‘ these supplements every other day , then fasting and exercising

      In that interview with Dr. Patrick he mentions that young mice lived longer if the protocol was pulsed rather than if they received the supplements daily, but he doesn't pulse his own protocol. Is that because he's middle aged and every day is more efficacious, or did you come away thinking that the pulsing is an emerging idea which he is still vetting before switching his own protocol

    Hello, really liking the series so far. When talking carbohydrate there is a lot of talk about how many plants utilize certain chemicals to ward of predation, and that because of this they are harmful to humans and should be avoided, making the case for "primal"/keto. The question I have is: is it possible that these chemicals could actually be stimulating an "adversity" response and in fact be very beneficial to humans?

      He addressed this in the previous episode or the one before I think. In short, the more stressed the plant, the better they are for you. Somebody correct if I’m wrong, btw.

      @@paulgennaro2001 the question is not the fact that the plants were stressed, but whether the chemicals that were stressing the plants is actually harmful to humans or in the end beneficial. Many people in the low carb, keto camp feel that those chemicals are very harmful to humans, really getting into the weeds now.

    What age would you recommend a person begin? Also, are there negatives to microdosing, say 2-3 days on, take a 2 weeks off, back on for 2-4 days , 1 week off. I am 28.

    I love this podcast, and the format. However, I was wondering if there is room for special episodes where you have another researcher alongside you that might be able to add some criticism or another perspective to the research. I came across Dr Rhonda Patrick's video on Metformin for healthy people who exercise, and she seems less enthusiastic about the research. I finally found a review of the literature from last year that gave me a better understanding, but I was wondering if you could make this more efficient. My thought is, maybe you can have someone like Dr Rhonda Patrick on, and discuss a set topic that neither of you too fully agree on, like Metformin. Not to debate necessarily, but to get a more complete view and really flesh out the current state of the research from multiple angles. Matt can still be the moderator, he's great!

    Hi David, there seems to be some confusion with the info you are giving on Spermidine. Everywhere I’ve looked it’s measured in grams not milligrams, even the highest dose supplements offer 8mg a day. How come you are taking 1 gram?

    This is exceptionally interesting. I wonder if there are any studies addressing the additivity of the benefits of different supplements! If two different supplements increase the lifespan of mice by say 30% each, what is the combined effect of the two?
